Project description:Serratia marcescens, a member of the order Enterobacterales, is adept at colonizing healthcare environments and an important cause of invasive infections. Antibiotic resistance is a daunting problem in S. marcescens because in addition to plasmid-mediated mechanisms, most isolates have considerable intrinsic resistance to multiple antibiotic classes. To discover endogenous modifiers of antibiotic susceptibility in S. marcescens, a high-density transposon insertion library was subjected to sub-minimal inhibitory concentrations of two cephalosporins, cefoxitin and cefepime, as well as the fluoroquinolone ciprofloxacin. Comparisons of transposon insertion abundance before and after antibiotic exposure identified hundreds of potential modifiers of susceptibility to these agents. Using single gene deletions, we validated several candidate modifiers of cefoxitin susceptibility and chose ydgH, a gene of unknown function, for further characterization. In addition to cefoxitin, deletion of ydgH in S. marcescens resulted in decreased susceptibility to multiple 3rd generation cephalosporins, and in contrast, to increased susceptibility to both cationic and anionic detergents. YdgH is highly conserved throughout the Enterobacterales, and we observed similar phenotypes in Escherichia coli O157:H7 and Enterobacter cloacae mutants. YdgH is predicted to localize to the periplasm and we speculate that it may be involved there in cell envelope homeostasis. Collectively, our findings provide insight into chromosomal mediators of antibiotic resistance in S. marcescens and will serve as a resource for further investigations of this important pathogen.

Project description:Cephaloridine (CER) is a classical beta-lactam antibiotic that has long served as a model drug for the study of cephalosporin antibiotic-induced acute tubular necrosis. In the present study, we analyzed gene expression profiles in the kidney of rats given subtoxic and toxic doses of CER in order to identify gene expression alterations closely associated with CER-induced nephrotoxicity. Male Fisher 344 rats were intravenously injected with three different doses (150, 300, and 600 mg/kg) of CER, and sacrificed after 24 h. Only the high dose (600 mg/kg) caused mild proximal tubular necrosis and a slight renal dysfunction. Microarray analysis identified hundreds of genes differentially expressed in the renal cortex following the exposure to CER, which could be classified into two main groups that were deregulated in dose-dependent and high dose-specific manners. The genes upregulated dose-dependently mainly included those involved in detoxification and antioxidant defense, which was considered to be associated with CER-induced oxidative stress. In contrast, the genes showing high dose-specific (lesion-specific) induction included a number of genes related to cell proliferation, which appeared to reflect a compensatory response to CER injury. We also found a subset of G2/M phase genes that exhibited hormesis-like (U-Shape) biphasic dose response; namely, downregulation only at the low and/or middle (subtoxic) doses. Furthermore, we could predict potential transcription regulators responsible for the observed gene expression alterations, such as Nrf2 and E2F family. Among the candidate gene biomarkers, kidney injury molecule 1 was markedly upregulated at the mildly toxic dose, suggesting that this gene can be used as an early and sensitive indicator for cephalosporin nephrotoxicity. Project description:Acremonium chrysogenum is the industrial producer of cephalosporin C. We isolated a mutant (AC554) from a T-DNA inserted library of A. chrysogenum. AC554 exhibited reduced conidiation and lack of cephalosporin C production. In consistent, the transcription of cephalosporin biosynthetic genes pcbC and cefEF was obviously decreased in AC554. TAIL-PCR and sequence analysis indicated that a T-DNA was inserted in the upstream of an open reading frame (ORF) which was designated AcmybA. Sequence analysis indicated that AcmybA encodes a novel Myb domain containing transcriptional factor. Observation of red fluorescence protein (RFP) tagged AcMybA showed that AcMybA is naturally located in the nuclear of A. chrysogenum. Transcription analysis demonstrated that AcmybA was overexpressed in AC554. In contrast with AC554, the AcmybA deleted mutant (DAcmybA) overproduced conidia in LPE medium and increased cephalosporin production during fermentation. To determine the genes under the influence of AcmybA, we sequenced and compared the transcriptome of DAcmybA, AC554 and the wild-type strain at different developmental stages. Results confirmed the repression of AcMybA on the key conidiation regulatory gene AcbrlA and the cephalosporin biosynthetic genes. Among the targets of AcMybA, 10 putative regulatory genes were selected and overexpressed in A. chrysogenum. Taken together, our results indicate that AcMybA negatively regulates conidiation and cephalosporin production in A. chrysogenum.

Project description:Serratia marcescens (S. marcescens), an opportunistic human pathogen, has been identified as a major cause of nosocomial infection and outbreaks. The purpose of this analysis is to examine the S. marcescens (ATCC 13880) protein profile using a high resolution mass spectrometry (MS). S. marcescens ATCC 13880 strain was grown in Luria-Bertani broth and the protein extracted underwent trypsin digestion followed by simple reverse phase liquid chromatography fractionation. Peptide fractions were then analyzed using Orbitrap Fusion Mass Spectrometry and raw MS data was processed using Proteome Discoverer software. The proteomic study identified 2,541 unique protein groups, corresponding to approximately 54% of the measured protein-coding genes. Bioinformatics analysis of these identified proteins demonstrated their involvement in biological processes such as cell wall organization, caperone-mediated protein folding and ATP binding. To our knowledge, this is the first high-performance S.marcescens proteomics analysis (ATCC 13880). These novel observations provide a key baseline molecular profile of the S. marcescens proteome which will prove to be helpful for the future research in understanding the host-pathogen interactions during infection, elucidating the mechanism of multidrug resistance and for developing novel diagnostic markers or vaccine for the disease.

First goal was to identify genes that are involved in immune response of germline-ablated P. pacificus when exposed to S. marcescens. Second, this was compared to longevity-regulating genes in germline-deficient animals (see series GSE37331). One-condition experiments. Germline ablated P. pacificus worms exposed to S. marcescens for 8 hours versus germline-ablated P. pacificus exposed to the control E. coli OP50. 4 biological replicates for each condition, including 2 dye-swaps.

Project description:The acid tolerance of industrial strains is a significant challenge in the fermentation process. The bacterium Serratia marcescens is part of the Enterobacteriaceae family of eubacteria, which is a potential industrial microorganism. However, the molecular mechanism behind S. marcescens acid resistance is not properly understood. In this study, we screened for novel regulators that respond to acidic conditions by a Tn5G transposon insertion mutagenesis of S. marcescens and found mutations in a gene encoding for the HTH_XRE super-family regulatory protein member, here named xrpA. Using transcriptomics and further experiments, we showed that the xrpA disruption conferred pleiotropic phenotype changes, including highly decreased biomass, altered cell shape, H+-ATPase activity, and deficiency of cell membrane permeability and integrity, compared with those of the parent (JNB5-1) strain at low pH. These data revealed that the molecular mechanism by which xrpA affects acid resistance of S. marcescens is through positive regulation of cell membrane permeability, integrity, and H+-ATPase activity to maintain intracellular homeostasis at low pH. Finally, we constructed an acidic resistant strain JNB5-1/pSX1314 by overexpression of xrpA in S. marcescens and found that its ability to produce prodigiosin by fermentation increased by 21.74% compared with that of parent strain at pH4.0. These results indicated that xrpA regulates tolerance to low pH by transcriptional regulation of acid stress response genes to maintain cell membrane function in S. marcescens.
