Project description:Potato plants are sensitive to multiple abiotic stresses such as drought, low temperature and high light. We analyzed the transcriptome of WT potato plants as well as that of transgenic potato plants expressing the Arabidopsis stress related transcription factor CBF1 that confers tolerance to multiple stresses. Wild type and AtCBF1OX transgenic potato plants were exposed to low temperature, high light, drought or kept under control conditions as described below in detail, and transcriptional changes induced by the different stresses were analyzed.

Proteomic analysis of sap reveals root-shoot communication dynamics in potato and barley following flg22 treatment

Project description:Potato (Solanum tuberosum, variety Princ) and barley (Hordeum vulgare, variety Sebastian) plants were cultivated in a controlled environment. Plants were grown in Potgrond H soil (Klasmann-Deilmann GmbH, Germany) under a 12-hour photoperiod with a constant temperature of 21 °C and a photon flux density of 100 μmol m⁻² s⁻¹. Potato tubers were used as starting material, while barley plants were grown from surface-sterilized seeds. After six weeks of cultivation for potato and three weeks for barley, the sap proteome was collected by cutting the stems 10–20 mm above the soil surface. Root exudates were sampled into three fractions over a two-hour period: F1 (15 min), F2 (75 min), and F3 (135 min). Each experiment included at least three independent biological replicates. A subset of plants was pre-incubated with flg22, a conserved peptide sequence derived from bacterial flagellin (QRLSTGSRINSAKDDAAGLQIA, >95% purity; ProteoGenix, France). Flg22 solution (1 μM flg22, 0.025% v/v Silwet L-77) was applied by spraying the leaves and pouring the solution under the pot. Potato plants received 30 ml and 200 ml of the solution for spraying and watering, respectively. Due to their smaller size, barley plants were treated with half the solution volume. Mock-treated plants received a solution containing only 0.025% Silwet L-77. After 24 hours, plants were cut, and the third fraction of the sap (75-135 min) was collected. Root and shoot tissues were collected in liquid nitrogen, the sap was collected as described above. Each experiment included at least three independent biological replicates.

Potato cyst nematode repsonses in potato

Project description:Sets of seven 2-week old potato plants carrying the nematode resistance gene H1, grown from tuber ‘chits’ in sandy loam at a constant temperature of 20 ºC and a light cycle of 16 hour light/8 hour dark, were each inoculated in the roots evenly with 2000 juveniles of the virulent potato cyst nematode Globodera pallida, or with the avirulent G. rostochiensis pathotype Ro1, or with water. Plants were manually watered throughout the duration of the experiment. 5, 17 and 33 days after inoculation, the roots were carefully washed and root tissue samples were individually flash-frozen in liquid nitrogen and stored at –80 ºC. Total RNA isolations were performed using the Qiagen RNeasy kit. All samples were treated with DNase. The experiment was replicated twice. Keywords: Direct comparison

Transcriptome sequencing identifies tuberous root development related genes in sweet potato

Project description:To screen genes related to the development of sweet potato tuberous roots, the high throughput sequencing of different stages of sweet potato tuberous roots was performed. The fibrous roots (FR; roots at 20 dap), developing tuberous roots (DR; roots at 60 dap) and mature tuberous roots (MR; roots at 120 dap) of Ipomoea batatas (L.) Taizhong 6 and MBP3 overexpressed lines were used for transcriptome analysis. Totally, we identified 5488 differentially expressed genes between different stage tuberous roots of Taizhong6 and 14312 differentially expressed genes between the tuberous roots of Taizhong6 and MBP3 overexpressed lines, by calculating the gene FPKM in each sample and conducting differential gene analysis. This study provides a foundation for the mechanism analysis of sweet potato tuberous root development.

Metabolic barriers controlling root microbiota assembly

Project description:Plant roots are an essential plant organ, which is functionally related to nutrient and water uptake. These functions are conditioned by their morphology since water and nutrients infiltrate the root from the soil through different tissue layers to reach the central vasculature. We have revealed that microbial composition affects root morphology complexity. Importantly, we also found that the presence of the microbiota changes morphological traits in the root under low nutrient conditions, suggesting an unexplored coordination between root morphology complexity and microbial communities with consequences for plant health. Using Synthetic Communities (Syncom), we identified three bacteria that are capable of modifying plant root morphogenesis. So, in this experiment, we exposed duckweed plants to a bacterial Syncom as well as three alternative Syncoms, each of which excludedimportant bacteria. Additionally, we also exposed the plants to single bacteria.
