Project description:Clinically important broadly reactive B cells evolve during multiple infections. The phenotype and function of B cells re-activated after secondary infection is different compared to B cells activated after a primary infection. Here we studied CD27highCD38high plasmablasts from patients with a primary or secondary dengue virus infection. Three transcriptionally and functionally distinct plasmablast clusters were identified. The largest cluster 0/1 was plasma cell-related and contained cells coding for virus serotype cross-reactive antibodies of the IgG1 isotype, consistent with memory B cell activation during an extrafollicular response. In contrast, cells in clusters 2 and 3 expressed low levels of antibody genes and high levels of genes associated with oxidative phosphorylation, EIF2 pathway and mitochondrial dysfunction. Clusters 2 and 3, which we name T cell-dependent plasmablasts, showed a transcriptional footprint of T cell help, in line with activation from either naÃƒÂ¯ve B cells or memory B cells. Our results contribute to the understanding of the parallel B cell activation events that occur in humans after natural primary and secondary infection.

SMART-seq2 of activated human plasmacytoid dendritic cells

Project description:Plasmacytoid and conventional dendritic cells (pDCs and cDCs, respectively) are closely related immune lineages with distinct functions. Whether pDCs can switch fate into the cDC lineage has remained controversial. Here, we sequenced purified pDCs and DCs to create a reference map of the transcriptome and chromatin landscape of all major human blood DC populations (GSE267174 and GSE267100). We compared this reference map to the transcriptomic and epigenetic profile of activated pDCs (GSE267099 and GSE266889). We show that activation of purified, bona fide human pDCs induces transcriptional and epigenetic heterogeneity at the single-cell level. A fraction of activated pDCs retains classical pDC features, while others acquire a cDC2-like identity through an intermediate state resembling transitional dendritic cells (tDCs). These datasets—generated using single-cell multiomic profiling (scRNA-seq and scATAC-seq) and SMART-seq—provide a resource to investigate the molecular basis of pDC-to-cDC2 fate transition.

PD-1highCXCR5–CD4+ Peripheral Helper T (Tph) cells Promote CXCR3+ Plasmablasts in the early phase of COVID-19

Project description:T cell–B cell interaction is the key immune response to protect host from severe viral infection. We used COVID-19 as a model of acute viral infections and analyzed CD4+ T cell subsets associated with plasmablasts expansion and clinical outcome. Peripheral helper T cells (Tph, denoted as PD-1highCXCR5–CD4+ T cells) were significantly increased, as were plasmablasts. Tph cells exhibited “B-cell help” signatures and induced plasmablasts differentiation in vitro. Expanded plasmablasts showed increased CXCR3 expression, which is positively correlated with higher frequency of activated Tph cells and better clinical outcome. These results elucidate a critical role for Tph cells in regulating protective B cell response during acute viral infection.
