Project description:The present work identified gene expression changes in response to total body irradiation (TBI) in heart, lungs, and liver tissue in Gottingen minipigs. Animals were exposed to 1.7 Gy, 1.9 Gy, 2.1 Gy and 2.3 Gy of TBI and were followed for 45 days. Organ samples were collected at the end of 45 days or sooner if the animal displayed morbidity necessitating euthanasia. RNA was extracted and gene expression profiling studied using Agilent arrays.

Project description:The population's increasing risk of acute large-scale radiological/nuclear exposures underlines the need for functional radiation biomarkers. As ionizing radiation triggers a complex response on the genome, proteome, and metabolome level, all were already reported as suitable indicators of radiation-induced damage in vitro or in animal models. We aim to analyze the blood of total-body irradiated (TBI) leukaemia patients using a mass spectrometry approach and to identify and quantify plasma proteins before and after irradiation. Combining two independent mass spectrometry approaches we identified 15 plasma proteins differing in the blood of TBI patients before and after irradiation. The majority of proteins were associated with the inflammatory responce of the immune system and with metabolism of lipids. These candidate radiation biomarkers might have implications for practical biological dosimetry.

Project description:This study utilized RNA-seq data to comprehensively analyze the transcriptome across five tissues (heart, liver, kidney, back fat, and loin) of minipigs. As the demand for organ transplantation continues to rise, xenotransplantation has gained significant attention as a promising solution to address the critical shortage of organ donors. Minipigs are considered a highly promising animal model for xenotransplantation research due to their remarkable physiological and anatomical similarities to humans. To evaluate the potential of minipigs, this study identified tissue-specific genes and differentially expressed genes (DEGs) related to body size, offering an in-depth exploration of their transcriptomic characteristics. The findings revealed that tissue-specific genes such as MYH7B in the heart and ITIH4 in the liver play pivotal roles in physiological and pathological processes. Additionally, DEGs associated with body size, such as DES in the heart and NRAP in the loin, provided valuable insights into the relationship between organ size and functionality. Notably, the expression patterns of immune-related genes such as IFI30, NCK2, NFKB1, and SOS2 demonstrated that minipigs exhibit a greater similarity to humans than to conventional pigs, highlighting their potential as an indispensable model for xenotransplantation and biomedical research.

Project description:Gottingen minipigs mirror the physiological radiation response observed in humans and hence make an ideal candidate model for studying radiation biodosimetry for both limited-sized and mass casualty incidents. We examined the whole blood gene expression profiles at day 1, 3 and 7 after total-body irradiation with 1.7, 1.9, 2.1 and 2.3 Gy doses of gamma-rays. Blood taken one day prior to radiation served as control. Six animals per dose group were used The minipigs were monitored for up to 45 days or time to euthanasia necessitated by radiation effects. Survival correlative signatures were identified from the data.

Project description:In the current study, we investigated the gene expression response of blood cells of non-human primates that were total body irradiated with a 4 Gy dose. We isolated total RNA from peripheral blood at 3 days before irradiation, and then from the same animals on days 3, 5 and 7 days after irradiation. Using Agilent Rhesus microarrays, we identified genes that were significantly differentially expressed across the 7-day time course of this study. We identified common biological functions affected that persisted across the 7-day study.

Project description:Background: The brain undergoes ionizing radiation exposure in many clinical situations, particularly during radiotherapy for brain tumors. The critical role of hippocampus in the pathogenesis of radiation-induced neurocognitive dysfunction is well recognized. Aim: The goal of this study is to test the potential contribution of non-targeted effects in the detrimental response of hippocampus to irradiation and to elucidate the mechanisms involved. Material and Methods: C57Bl/6 mice were whole body (WBI) or partial body (PBI) irradiated with 0.1 or 2.0 Gy of X-rays or sham irradiated. PBI consisted in the exposure of the lower third of the mouse body, whilst the upper two thirds were shielded. Hippocampi were collected 15 days or 6 months post-irradiation and a multi-omics approach was adopted to assess the molecular changes in non-coding RNAs, proteins and metabolic levels, as well as histological changes in the rate of hippocampal neurogenesis. Results: Notably, at 2.0 Gy the pattern of early molecular and histopathological changes induced in the hippocampus at 15 days following PBI was similar in quality and quantity to the effects induced by WBI, thus providing a proof of principle of the existence of out-of-target radiation response in the hippocampus of conventional mice. We detected major alterations in DAG/IP3 and TGF- signaling pathways as well as in the expression of proteins involved in the regulation of long-term neuronal synaptic plasticity and synapse organization, coupled with defects in NSCs self-renewal in the hippocampal dentate gyrus. However, compared to the persistence of the WBI effects, most of the PBI effects were only transient and tended to decrease at 6 months post-irradiation, indicating important mechanistic difference. On the contrary, at low dose we identified a progressive accumulation of molecular defects that tended to manifest at later post-irradiation times. These data, indicating that both targeted and non-targeted radiation effects might contribute to the pathogenesis of hippocampal radiation-damage, have general implications for human health
