Project description:Periodontitis can impair the osteogenic differentiation of human periodontal mesenchymal stem cells, but the underlying molecular mechanisms are still poorly understood. Long noncoding RNAs (lncRNAs) have been demonstrated to play significant roles under both physiologic and pathological conditions. We performed comprehensive lncRNAs profiling by lncRNA microarray to identify differentially expressed long noncoding RNA expression between Periodontal ligament stem cells from healthy Periodontal tissue and periodontal ligament stem cells from inflammatory periodontal tissue. Our analysis identified 233 lncRNAs and 423 mRNAs that were differently expressed (fold change >2.0, p-value < 0.05) between the two groups of cells. The GO analysis revealed that the significantly down-regulated biological processes included multicellular organismal process, developmental process and multicellular organismal development and the significantly up-regulated biological processes included cellular process, biological regulation and response to stimulus in periodontal ligament stem cells from inflammatory periodontal tissue. The Pathway analysis revealed that the differentially expressed mRNAs may involved in Focal adhesion, ECM-receptor interaction, Bacterial invasion of epithelial cells, Long-term depression, Circadian entrainment and HIF-1 signaling pathway. Two-condition experiment, periodontal ligament stem cells from healthy periodontal tissue (hPDLSCs) vs. periodontal ligament stem cells from inflammatory periodontal tissue (pPDLSCs), Biological replicates: 3 control replicates (hPDLSCs), 3 testing replicates (pPDLSCs).

Project description:This study investigated the role of mitochondrial function and the downstream long noncoding RNAs (lncRNAs) in regulating inflammation-induced changes to the cementoblastic differentiation of PDLSCs. We found that inflammatory cytokine-caused impairment to cementoblastic differentiation of PDLSCs was closely correlated to their mitochondrial function, and in terms of lncRNA microarray analysis and gain/loss-of-function studies, gastric cancer associated transcript 2 (GACAT2) was identified as a regulator across the cellular events of both inflammation-mediated mitochondrial function and cementoblastic differentiation. Next, comprehensive identification of RNA-binding proteins by mass spectrometry (ChIRP-MS) and parallel reaction monitoring (PRM) assay revealed that GACAT2 was directly binded to protein pyruvate kinase M1/2 (PKM1/2). Further functional studies demonstrated that GACAT2 overexpression increased cellular protein expressions of PKM1/2, PKM2 tetramer and phosphorylated PKM2, led to an enhanced pyruvate kinase (PK) activity along with increased translocation of PKM2 into mitochondria. Finally, we found that GACAT2 overexpression could reverse inflammation-compromised mitochondrial function and cementoblastic differentiation of PDLSCs, and this function could be abolished by PKM1/2 knockdown. Our data suggest that lncRNA GACAT2 plays a critical role in inflammation-compromised mitochondrial function and cementoblastic differentiation by binding protein PKM1/2.

The long noncoding RNA AC018926.2 plays a crucial role in palmitic acid exposure-compromised osteogenic potential of periodontal ligament stem cells

Project description:This study investigated the cellular response of human periodontal ligament stem cells (PDLSCs) to elevated levels of a specific fatty acid (FA), namely, palmitic acid (PA) and the role of long noncoding RNAs (lncRNAs) in regulating PA exposure-compromised osteogenic potential of PDLSCs. It was found that exposure of PDLSCs to abundant PA led to decreased osteogenic differentiation of cells. Through microarray analysis and gain/loss-of-function studies, AC018926.2 was identified and validated as the lncRNA that was most sensitive to PA and a regulator in the process of PDLSC osteogenic differentiation under a PA condition. Then, RNA sequencing and KEGG analysis demonstrated that AC018926.2 upregulated integrin α2 (ITGA2) expression at the transcriptional level and therefore activated ITGA2/FAK/AKT signaling. Further functional studies revealed that inactivation of ITGA2/FAK/AKT signaling by silencing ITGA2 counteracted the enhancement of the PDLSC osteogenic potential induced by AC018926.2 overexpression. Moreover, the results of bioinformatics analysis and the RNA immunoprecipitation (RIP) assay suggested that AC018926.2 might transcriptionally regulate ITGA2 expression by binding to poly (ADP-ribose) polymerase 1 (PARP1). Our data suggest that AC018926.2 plays a crucial role in PA exposure-compromised osteogenic potential of PDLSCs.
