Project description:In summary, we have for the first time generated a Dox inducible and switchable tdTomato pig strain with dual knock-in at Hipp11 and Rosa26 locus. This pig model provide a versatile tool model to produce stable transgenic pigs with inducible overexpressing any gene of interest by using the technology of phiC31 integrase-mediated cassette exchange. Based on this approach, an inducible hKRASG12D-expressing pig model line was established and tumor formation was induced after Dox administration. We expect that the inducible tool pig model would greatly facilitate the production of controllable transgenic pigs, and broaden the applications of transgenic pigs in biomedicine and agriculture fields.

TET-dependent regulation of retrotransposable elements in mouse embryonic stem cells [RNA-seq]

Project description:TET enzymes oxidise DNA methylation as part of an active demethylation pathway. Despite extensive research into the role of TETs in genome regulation, little is known about their effect on transposable elements (TEs), which make up nearly half of the mouse and human genomes. Epigenetic mechanisms controlling TEs have the potential to affect their mobility and to drive the co-adoption of TEs for the benefit of the host. We have performed a detailed investigation of the role of TET enzymes in the regulation of TEs in mouse embryonic stem cells (ESCs). We found that TET1 and TET2 bind multiple TE classes that harbour a variety of epigenetic signatures indicative of different functional roles. Namely, TETs co-bind with pluripotency factors to enhancer-like TEs that interact with highly expressed genes in ESCs, whose expression is partly maintained by the demethylating action of TET2. TETs and 5-hydroxymethylcytosine are also strongly enriched at the 5’ UTR of full-length, evolutionarily young LINE1 elements, a pattern that is conserved in human embryonic stem cells. TET depletion leads to a marked increase in DNA methylation levels at LINE1s, but surprisingly their expression is stably maintained through additional TET-dependent activities. Specifically, we find that TET1 recruits the SIN3A co-repressive complex to LINE1s, ensuring their repression upon TET-driven hypomethylation. This dual nature of TET action may reflect the evolutionary battle between TEs and the host. Our data implicate TET enzymes in the evolutionary dynamics of TEs, both in the context of exaptation processes and of retrotransposition control.

Project description:This paper gives the available data for the maximum equilibrium solubility limits of each of the first 83 elements (H to Bi) of the periodic table in each of the others. This is expressed in the form of the maximum value of the equilibrium solid solubility for terminal solid solutions, expressed in atomic percentage (at%), occurring at any temperature where the room temperature phase of the pure element acting as the solvent is stable. The values thus represent the compatibility between different elements in the formation of alloys and similar combinations, and will be of use for research into fundamental solid state physics relating to elemental interactions, for the interpretation of phase structures in materials research and for the design of alloys and materials involving elemental mixing of the elements.

OGT prevents DNA demethylation and suppresses the expression of transposable elements in heterochromatin by restraining TET activity genome-wide

Project description:The O-GlcNAc transferase OGT interacts robustly with all three mammalian TET methylcytosine dioxygenases. We show here that deletion of the Ogt gene in mouse embryonic stem cells (mESC) results in a widespread increase in the TET product 5-hydroxymethylcytosine (5hmC) in both euchromatic and heterochromatic compartments, with concomitant reduction of the TET substrate 5-methylcytosine (5mC) at the same genomic regions. mESC treated with an OGT inhibitor displayed increased 5hmC, and attenuating the TET1-OGT interaction in mESC resulted in a genome-wide decrease of 5mC, indicating that OGT restrains TET activity and limits untoward DNA demethylation in a manner that requires the TET-OGT interaction and the catalytic activity of OGT. DNA hypomethylation in OGT-deficient cells was accompanied by de-repression of transposable elements (TEs) predominantly located in heterochromatin. We suggest that OGT protects the genome against TET-mediated DNA demethylation and loss of heterochromatin integrity, preventing the aberrant increase in TE expression noted in cancer, autoimmune-inflammatory diseases, cellular senescence and ageing.

Water Distribution Systems in Pig Farm Buildings: Critical Elements of Design and Management.

Project description:Drinking water distribution systems (WDSs) within buildings on pig farms have critical elements of their design and management that impact water provision to pigs, water quality, the efficacy of in-water antimicrobial dosing, and, thus, pig health and performance. We used a mixed-methods approach to survey managers of 25 medium to large single-site and multi-site pig farming enterprises across eastern and southern Australia. We found wide variation in the configuration (looped or branched) and total length of WDSs within buildings across farms and in pipe materials and diameters. Within many conventional buildings and some eco-shelters, WDSs were 'over-sized', comprising large-diameter main pipelines with high holding volumes, resulting in slow velocity water flows through sections of a WDS's main pipeline. In over half of the weaner buildings and one-third of grower/finisher buildings, the number of pigs per drinker exceeded the recommended maximum. Few farms measured flow rates from drinkers quantitatively. WDS sanitization was not practiced on many farms, and few managers were aware of the risks to water quality and pig health. We identified important aspects of water provision to pigs for which valuable recommendations could be added to industry guidelines available to pig farm managers.
