Project description:Hypertension affects one-third of the world's population, leading to cardiac dysfunction that is modulated by resident and recruited immune cells. Cardiomyocyte growth is essential to withstand hypertensive stress, however whether immune cells are involved in this cardioprotective process is unclear. In normotensive animals, single-cell transcriptomics of fate-mapped self-renewing cardiac resident macrophages (RMs) revealed transcriptionally diverse cell states with a core repertoire of reparative gene programs, including high expression of Insulin-like Growth Factor-1 (Igf1). Hypertension drove selective in situ proliferation and transcriptional activation of some cardiac RM subsets, directly correlating with increased cardiomyocyte size and cardiac mass. Inducible ablation of RMs, or selective deletion of RM-derived Igf1 caused complete absence of adaptive cardiomyocyte growth and development of cardiac dysfunction without adaptive organ growth. Single cell transcriptomics identified a conserved IGF1-expressing macrophage subpopulation in human cardiomyopathy. Here, we defined the absolute requirement of RM-produced IGF-1 in cardiac adaptation to hypertension.

White adipose tissue resident macrophages are required for adipogenesis and energy storage.

Project description:White adipose tissue (WAT) stores energy in the form of lipids. WAT macrophages cause adipose tissue inflammation and insulin resistance when intake exceeds expenditures, but their contribution to homeostasis remain poorly understood. Here, we show that two populations, ã and ä, of TNF-producing bone marrow-derived monocytic cells are transiently recruited to damaged adipocytes and mediate insulin resistance. In contrast, we identify α/β yolk-sac derived resident macrophages that produce adipogenic growth factors, and form a niche required for adipogenesis as white adipocytes do not differentiate in their absence. Production of adipogenic growth factors by α/β macrophages increases with lipid storage, but they do not contribute to inflammation. Thus the molecular identities and functions of resident macrophages and BM-derived cells are specified by their lineage rather than diet variation. These results identify an essential function of WAT resident macrophages in adipogenesis and energy homeostasis, separate from inflammation mediated by the recruitment of inflammatory leukocytes.

Nutrient regulation of the islet epigenome controls adaptive insulin secretion

Project description:Adaptation of the islet β-cell insulin secretory response to changing insulin demand is critical for blood glucose homeostasis, yet the mechanisms underlying this adaptation are unknown. Here, we show that nutrient cues adapt insulin secretion by modulating chromatin state and transcription of genes regulating β-cell nutrient sensing and metabolism. Feeding stimulates histone acetylation at sites occupied by the chromatin-modifying enzyme Lsd1 in islets. We demonstrate that β-cell-specific deletion of Lsd1 leads to insulin hypersecretion, aberrant expression of nutrient response genes, and histone hyperacetylation, features we also observed in the db/db model of chronically increased insulin demand. Moreover, genetic variants associated with fasting glucose levels and type 2 diabetes risk are enriched at LSD1-bound sites in human islets, suggesting interindividual variation in β-cell functional adaptation in humans. These findings reveal nutrient state-dependent modulation of the islet epigenome and identify Lsd1 as a regulator of feeding-stimulated chromatin modification and adaptive insulin secretion.
