Project description:Transcriptional profiling of human glioma cells comparing control GFP expressing cells with glioma cells transfected with a human PDGF-A gene. The isogenic cell lines were used to study the impact on glioma tumorigenesis and invasion. Goal was to determine the effects of PDGF-A gene transfection on global ES gene expression. Two set of glioma cell lines, LN444 vs LN444/PDGF-A and LN443 vs LN443/PDGF-A. Biological replicates: 2 control replicates, 2 transfected replicates.
Project description:Glioblastoma (GBM) is a deadly disease without effective treatment. Because glioblastoma stem cells (GSCs) contribute to tumor resistance and recurrence, improved treatment of GBM can be achieved by eliminating GSCs through inducing their differentiation. Prior efforts have been focused on studying GSC differentiation towards the astroglial lineage. We used microarrays to detail the global programme of gene expression underlying ZNF117 knockout and identified distinct classes of up-regulated genes during this process.

Project description:Differential mRNA expression patterns were seen in GSC272-vector compared to GSC272-POSTN shRNA tumors. We used microarrays to POSTN regulated gene expression in glioma stem cells. Gene expression comparisons were shown between: 1. mRNA control from GSC272 glioma stem cells implanted mouse brain tumor at 7 weeks after implantation, and 2. mRNA deplection of POSTN from knock down of POSTN of GSC272 glioma stem cells implanted mouse brain tumor at 7 weeks after implantation.
