Project description:Maternal folate deficiency during pregnancy results in adverse outcomes. We have found miR-30e-3p and 34b-5p to be directly downregulated by folate deficiency in placental trophoblast cells and wanted to determine cell functions/pathways affected by these miRNAs. Using microarrays we identified a number of functional pathways for further analysis that could contribute to placental dysfunction in folate deficient women.

Project description:Studies have shown that miR-361-3p is associated the progression of HCC cells and has potential to be used as a biomarker for cancer therapy. However, the mechanism of how miR-361-3p regulates the progression of HCC cells is not clear. To explore this mechanism, we transfected miR-361-3p mimics into Huh 7 cells , and verified the transfection efficiency with qRT-PCR. Then, we performed transcriptomic analysis using data obtained from RNA-seq of 3 pairs of these transfected cells.

MiR-30e in Aortic Smooth Muscle Cells and Bone Marrow Mesenchymal Stem Cells [Experiment 3]

Project description:MiR-30e represses the osteogenic program in mesenchymal stem cells (MSCs) and smooth muscle cells (SMCs) by targeting IGF2, and drives their differentiation into adipogenic or smooth muscle lineage, respectively. In Experiment 1, SMCs were cultured from mouse aortas and transduced with miR-30e or control lentivirus. In Experiment 2, MSCs were extracted from mouse bone marrow and transduced with miR-30e or control lentivirus. In Experiment 3, the MSCs were transfected with a scrambled oligo or anti-miR-30e oligo. In all 3 experiments, N=3 per group. RNA was extracted from each experiment and run on Affymetrix arrays.

To study the each function of let-7d-3p, let-7e-5p,miR-146a-5p,miR-130a-3p, miR-151a-3p which are significantly upregulated in the plasma of atopic patients, we performed mimic-transfected THP-1 cells, a mononuclear cell line, and performed comprehensive genetic analysis.
