Project description:In mammalian testes, a valve-like structure called Sertoli valve is located at the border region of the seminiferous tubule and the rete testis. Having identified Sox17 in the rete testis as a positive regulator of the valve formation, this study aim to elucidate the mechanism of Sox17-expressing rete testis to modulate the testicular homeostasis. Our scRNA-seq data demonstrate the altered gene expression levels of several growth factors in Sox17-depleted rate testis epithelia, and highlighted the altered proportion of Sertoli cells located around the proximal part of the testis. This study provides a comprehensive profile of the proximal part of the testis including the rete testis and Sertoli valve for the first time, and further illustrate the functional role of the Sox17+ rete tesits to orchestrate the testicular homeostasis.

Project description:The rete testis (RT) is a region of the mammalian testis which plays an important role in testicular physiology. The RT epithelium consists of cells sharing some well-known gene markers with supporting Sertoli cells (SCs). However, little is known about the differences in gene expression between these two cell populations. Here, we used fluorescence-activated cell sorting (FACS) to obtain pure cultures of neonatal RT cells and SCs and identified differentially expressed genes (DEGs) between these cell types using RNA sequencing (RNA-seq).

Stool-derived 16s rRNA V4 sequencing from intestinal-epithelium-LSD1-cKO mice against WT controls (both at 2-month-old and P14 timepoints)

Project description:Villin-Cre+ Lsd1fl/fl (cKO) mice display an immature intestinal epithelium characterized by an incomplete differentiation of enterocytes and secretory lineages, reduced number of goblet cells and a complete loss of Paneth cells. This experiment aims to elucidate the differences in stool microbial composition derived from WT (Villin-Cre- Lsd1fl/fl) and cKO mice both in adult (2-month-old) and neonatal (14 days postpartum P14) stages. Different timepoints are crucial to understand the role of intestinal maturation in microbiome composition since said maturation is dependent on time-dependent external cues happening at P14-21 (weaning and transition from milk to solid foods).

Rete ridges form via evolutionarily distinct mechanisms in mammalian skin

Project description:The loss of fur during human evolution has long mystified scientists and the public. Reduced hair density coincides with the acquisition of epidermal rete ridges, whose developmental timing and molecular mechanisms are poorly understood despite their prominence in humans. Examination of human and pig skin development revealed that rete ridges form through an independent mechanism from hair follicles and sweat glands by establishing interconnected epidermal invaginations. We documented rete ridge occurrence across Mammalia, including in grizzly bears and dolphins, and showed that neonatal pig wounds can regenerate them de novo. Multi-species spatiotemporal transcriptomics identified significant signaling interactions between epidermal and dermal cells during rete ridge morphogenesis, particularly through BMP. We also demonstrated that murine fingerpad skin forms rete ridges and functionally requires epidermal BMP signaling. We propose that evolution of rete ridges in mammalian skin involved replacement of the molecular program for formation of discrete microscopic appendages, including hair follicles and sweat glands, with a distinct program for the interconnected appendage network. Broad epidermal activation of BMP is required for the development of rete ridge networks organized around underlying dermal pockets. Understanding rete ridge mechanisms may enable the development of therapeutic approaches to regenerate epidermal appendages lost during wounding or disease in humans.

SOX17-positive rete testis epithelium is required for Sertoli valve formation and normal spermiogenesis in the male mouse.

Project description:Seminiferous tubules (STs) in the mammalian testes are connected to the rete testis (RT) via a Sertoli valve (SV). Spermatozoa produced in the STs are released into the tubular luminal fluid and passively transported through the SV into the RT. However, the physiological functions of the RT and SV remain unclear. Here, we identified the expression of Sox17 in RT epithelia. The SV valve was disrupted before puberty in RT-specific Sox17 conditional knockout (Sox17-cKO) male mice. This induced a backflow of RT fluid into the STs, which caused aberrant detachment of immature spermatids. RT of Sox17-cKO mice had reduced expression levels of various growth factor genes, which presumably support SV formation. When transplanted next to the Sox17+ RT, Sertoli cells of Sox17-cKO mice reconstructed the SV and supported proper spermiogenesis in the STs. This study highlights the novel and unexpected modulatory roles of the RT in SV valve formation and spermatogenesis in mouse testes, as a downstream action of Sox17.

Project description:We investigated transcriptional and translational changes in rete mirabile tissue of European yellow and silver eels. We therefore analyzed the transcriptome as well as the proteome of rete capillaries of specimens of both developmental stages. The results clearly show the presence of a large number of membrane transport proteins in the transcriptome as well as in the proteome. ATP required to energize ion and metabolite transport can be generated in the aerobic metabolism. A high ROS defense capacity is established in rete endothelial cells, probably connected to the hyperbaric oxygen partial pressures, generated in the rete by back-diffusion of oxygen.

Project description:This dataset consists of 33 raw MS files, an associated peak list and result file, acquired on an Q Exactive plus mass spectrometer operated in Data Dependent Acquisition mode. The files are associated with a manuscript submitted for publication. Publication title: " Expression of transport proteins in the rete mirabile of European silver and yellow eel " The swimbladder rete mirabile is known for its highly efficient countercurrent concentrating ability, allowing for the generation of hyperbaric oxygen partial pressures in the range of several 10th or even more than 100 atmospheres. The transcapillary exchange between venous and arterial capillaries is often explained by passive diffusion and hydraulic (osmotic) transport. The reported countercurrent concentration of lactate ions, however, raises the possibility that specific transport proteins contribute to the countercurrent concentrating ability of the rete. We therefore analyzed the transcriptome as well as the proteome of rete capillaries of the European eel. The results clearly show the presence of a large number of membrane transport proteins in the transcriptome as well as in the proteome. Transcriptome profile of MafB-cKO Sertoli cells.

Project description:MafB is a transcription factor acts as an important regulator of the development and differentiation of various organs and tissues. MafB is expressed in sertoli, leydig, and germ cells in adult mice testis. However, analysis of MafB cKO mice testis showed no dissruption of spermatogenesis or obvious abnormal structure. Thus, the exact function is unclear. We examined any change in the transcriptome profile of MafB-cKO Sertoli cells. Results of signaling pathway analysis of the downregulated genes was related to immune-function but not spermatogenesis.
