Project description:Osteosarcoma (OS) is a life-threatening disease affecting mainly children and adolescents, it is urgently needed to find novel anticancer agents against it. In the present study, we demonstrated that Ailanthone (AIL), a major component of the Chinese medicine Ailanthus altissima, significantly suppressed cell growth, inhibited cell migration and invasion, induced cell cycle arrest instead of apoptosis in OS cells. Mechanistically, combined transcriptomics, proteomics and metabolomics analyses revealed that AIL induced widespread changes in metabolic programs in OS cells, while the serine biosynthetic pathway was the most significantly affected pathway. The results were verified by reverse test. Metabolic reprogramming is an essential hallmark of malignancy including OS. Our findings suggest that AIL holds a great potential to be an OS treatment strategy through mediating metabolic reprogramming.

Ribosomal protein S3 regulates GLI2-mediated osteosarcoma invasion

Project description:It has been reported that GLI2 promotes proliferation, migration, and invasion of mesenchymal stem cell and osteosarcoma cells. To examine the molecular mechanisms of GLI2-mediated osteosarcoma metastasis, we performed a microarray analysis. The gene encoding ribosomal protein S3 (RPS3) was identified as a target of GLI2. Real-time PCR revealed that RPS3 was upregulated in osteosarcoma cell lines compared with normal osteoblast cells. Knockdown of GLI2 decreased RPS3 expression, whereas forced expression of a constitutively active form of GLI2 upregulated the expression of RPS3. RPS3 knockdown by siRNA decreased the migration and invasion of osteosarcoma cells. Although forced expression of constitutively active GLI2 increased the migration of human mesenchymal stem cells, knockdown of RPS3 reduced the up-regulated migration. In contrast, forced expression of RPS3 increased migration and invasion of osteosarcoma cells. Moreover, reduction of migration by GLI2 knockdown was rescued by forced expression of RPS3. Immunohistochemical analysis showed that RPS3 expression was increased in primary osteosarcoma lesions with lung metastases compared with those without. These findings indicate that GLI2–RPS3 signaling may be a marker of invasive osteosarcoma and a therapeutic target for patients with osteosarcoma.

Adaptor protein CIN85 potentiates migration of osteosarcoma cells

Project description:The adaptor protein CIN85, encoded by the SH3KBP1 gene, is a member of the CIN85/CMS family of adaptor molecules involved in many signaling pathways. CIN85 functionally contributes to different biological processes in normal and cancer cells, such as vesicle-mediated transport, cell adhesion, migration, and invasiveness. In this study, we found that the expression of CIN85 was elevated in osteosarcoma compared to normal bone, and in metastases compared to primary bone tumors. Overexpression of CIN85 in osteosarcoma cell lines enhanced their migration and collagen invasion while silencing CIN85 suppressed cell migration. We used human osteosarcoma HOS cells with the overexpression of CIN85 or its knockdown to study the transcriptomic changes induced by this protein. Differentially expressed genes were enriched in cell proliferation, apoptosis, cell differentiation, extracellular matrix, adhesion, focal adhesion, tight junctions, and cell migration. Several abundant signaling pathways involved in tumorigenesis were identified, including NF-κB, TNF, MAPK, Wnt, PI3K/Akt/mTOR, TGF, and VEGF.

Small molecule activation of PKM in cancer cells induces serine auxotrophy

Project description:Proliferating tumor cells use aerobic glycolysis to support their high metabolic demands. Paradoxically, increased glycolysis is often accompanied by expression of the lower activity PKM isoform, effectively constraining lower glycolysis. Here, we report the discovery of novel PKM activators with a unique allosteric binding mode. Characterization of how these compounds impact cancer cells revealed an unanticipated link between glucose and amino acid metabolism. PKM activation resulted in a metabolic rewiring of cancer cells manifested by a profound dependency on the non-essential amino acid serine for continued cell proliferation. Induction of serine auxotrophy by PKM activation was accompanied by reduced carbon flow into the serine biosynthetic pathway and increased expression of high affinity serine transporters. These data support the hypothesis that PKM expression confers metabolic flexibility to cancer cells that allows adaptation to nutrient stress. A549 cancer cells were treated with compound-16 for up to 24 hours in the presence and absence of serine in the media.

ASCT2 is a major contributor to serine uptake in cancer cells

Project description:The non-essential amino acid serine is a critical nutrient for cancer cells due to its diverse biosynthetic functions. While some tumors can synthesize serine de novo, others are auxotrophic and therefore reliant on serine uptake. Importantly, despite several transporters being known to be capable of transporting serine, the transporter(s) that mediate serine uptake in cancer cells are not known. Here, we characterize the amino acid transporter ASCT2 (SLC1A5) as a major contributor to serine uptake in cancer cells. ASCT2 is well-known as a glutamine transporter in cancer, and our work demonstrates that serine and glutamine compete for uptake through ASCT2. We further show that ASCT2-mediated serine uptake is essential for purine nucleotide biosynthesis and that ERα promotes serine uptake by directly activating SLC1A5 transcription. Together, our work defines an additional important role for ASCT2 as a serine transporter in cancer and evaluates ASCT2 as a potential therapeutic target.
