Single-nucleus transcriptomic profiling of adult male Japanese medaka brain following parental benzo[a]pyrene exposure

Project description:Benzo[a]pyrene (BaP) is a polycyclic aromatic hydrocarbon associated with developmental and neurobehavioral toxicity. This study examined whether parental BaP exposure produces transcriptional changes in the brains of unexposed adult first filial generation (F1) Japanese medaka. Adult parental fish were exposed to solvent control or 1 µg/L BaP for 21 days; eggs were collected after exposure and F1 offspring were reared in clean water. Whole brains from three control-lineage and three BaP-lineage adult males were analyzed by single-nucleus RNA sequencing. Fourteen transcriptionally distinct brain cell populations were resolved, and treatment-associated differential expression was evaluated within each cluster.

New insights into benzo[⍺]pyrene osteotoxicity in zebrafish

Project description:Persistent and ubiquitous organic pollutants, such as the polycyclic aromatic hydrocarbon benzo[⍺]pyrene (BaP), represent a major threat to aquatic organisms and human health. Beside some well-documented adverse effects on the development and reproduction of aquatic organisms, BaP was recently shown to affect fish bone formation and skeletal development through mechanisms that remain poorly understood. In this work, several bone-related in vivo assays were used in zebrafish to evaluate the osteotoxic effects of BaP during bone development and regeneration. Exposure to BaP for 3 days induced a dose-dependent reduction of the size of the opercular bone in 6 days post-fertilization (dpf) larvae, and affected osteoblast maturation as observed by the expression of the mature marker, osteocalcin. Exposure to BaP for 27 days affected the development of the axial skeleton and increased the incidence and severity of skeletal deformities. During bone regeneration, BaP affected the mineralization of newly formed fin rays and scales, while it impaired fin ray patterning and scale shape, through mechanisms that may involve an imbalance of bone remodeling. Transcriptomic and gene expression analyses indicate that BaP induced the activation of xenobiotic and metabolic pathways, while negatively impacting extracellular matrix formation and organization. Interestingly, BaP exposure positively regulated inflammation markers in 6 dpf larvae and increased neutrophil recruitment. A direct interaction between neutrophils and bone extracellular matrix or bone forming cells was observed in vivo, suggesting a role for neutrophils in the mechanisms underlying BaP osteotoxicity. Our work provides novel data on the cellular and molecular players involved in BaP osteotoxicity and brings new insights into a possible role for neutrophils in inflammatory bone reduction.

Effect of benzo(a)pyrene on the replication of vesicular stomatitis virus and transcriptomic analysis.

Project description:Benzo(a)pyrene (Bap), a key component of cigarette smoke (CS), is a pollutant and widely present in our daily life. The roles of Bap on viral replication have been amply discussed and controversial. In our present study, we investigated the effect and mechanism of Bap on the replication of vesicular stomatitis virus (VSV) and respiratory syncytial virus (RSV) in vitro. HeLa cells were pre-exposed to Bap at 10 μM for 48 h and then infected with VSV (MOI=1) for 24 h. RNA sequencing analysis was conducted to identify Bap-dysregulated genes and qPCR analysis was used to identify the result. Chronic exposed to Bap significantly inhibited the replication of VSV and RSV in vitro and in vivo.

Sex differences in embryonic gonad transcriptomes and benzo[a]pyrene metabolite levels after transplacental exposure

Project description:Polycyclic aromatic hydrocarbons like benzo[a]pyrene (BaP) are generated during incomplete combustion of organic materials. Prior research has demonstrated that BaP is a prenatal ovarian toxicant and carcinogen. However, the metabolic pathways active in the embryo and its developing gonads and the mechanisms by which prenatal exposure to BaP predisposes to ovarian tumors later in life remain to be fully elucidated. To address these data gaps, we orally dosed pregnant female mice with BaP from E6.5-11.5 (0, 0.2 or 2 mg/kg-day) for metabolite measurement or E9.5-11.5 (0 or 3.33 mg/kg-day) for embryonic gonad RNA-sequencing. Embryos were harvested at E13.5 for both experiments. The sum of BaP metabolite concentrations increased significantly with dose in the embryos and placentas, and concentrations were significantly higher in female than male embryos and in embryos than placentas. RNA sequencing revealed that enzymes involved in metabolic activation of BaP are expressed at moderate to high levels in embryonic gonads and that greater transcriptomic changes occurred in the ovaries in response to BaP than in the testes. We identified 490 differentially expressed genes (DEGs) with FDR p-values <0.05 when comparing BaP-exposed to control ovaries, but no statistically significant DEGs between BaP-exposed and control testes. Genes related to monocyte/macrophage recruitment and activity, prolactin family genes, and several keratin genes were among the most upregulated genes in the BaP-exposed ovaries. Results show that developing ovaries are more sensitive than testes to prenatal BaP exposure, which may be related to higher concentrations of BaP metabolites in female embryos.

Project description:Benzo[a]pyrene (BaP) is a widespread polycyclic aromatic hydrocarbon and environmental contaminant with potential adverse effects on early development. To investigate the transcriptional response of human pluripotent stem cells to BaP exposure, H9 human embryonic stem cells were exposed to 1 μM BaP for 7 days. Transcriptomic changes following BaP exposure were subsequently assessed by bulk RNA sequencing. This dataset provides a resource for investigating the molecular responses and potential mechanisms associate with BaP exposure during the pluripotent stage of human development.
