Surface mechanics and compressive stress impact mammalian follicle development

Project description:The maturation of functional eggs in ovaries is essential for successful reproduction in mammals. Despite its biological and clinical importance, the underlying mechanisms regulating folliculogenesis remain enigmatic. Here, using murine ovaries, we report that the theca cells surrounding secondary follicles play a critical role in regulating follicle development through mechanical signalling. Using biophysical approaches, we found that the contractile theca cells exert significant compressive stress to the follicular interior through active assembly of fibronectin. Manipulation of compressive stress by targeting theca cell contractility, basement membrane integrity or intrafollicular pressure leads to changes in follicle size and mechanics, granulosa cell YAP signalling and oocyte-granulosa cell communications. Transcriptomics and quantitative immunofluorescence reveal that compressive stress impacts functional follicle growth through regulating the balance between granulosa cell proliferation and death that drives tissue pressure homeostasis. Altogether, our study identifies new mechanical functions of theca cells and provides quantitative evidence of the role of compressive stress in regulating mammalian folliculogenesis.

Intracellular zinc chaperones in plants

Project description:Metal availability is tightly regulated within cells through metal homeostasis to prevent cytotoxicity from excess and protein inactivity from deficiency. Limited bioavailability of essential metals in soil can negatively impact crop health and yield. A plant zinc (Zn)-homeostatic mechanism involving Zn transferases has been discovered, which plays a crucial role in delivering zinc to vital zinc-dependent proteins, particularly under zinc limitation. This study focused on investigating the effects of loss of AtZNG1 (AT1G26520), a key component of this mechanism, on growth, transcriptome, and proteome in Arabidopsis thaliana during zinc deficiency. The findings from multi-omics analyses highlight the broad consequences of this mechanism's disruption. The goal of this experiment wass to identify differentially abundant proteins in A.thaliana root tissue upon Zn withdrawal, in S999 zng1 mutant cells and wild-type control.

Differential gene expression of static and intermittent compressive force treated human periodontal ligament cells

Project description:Mechanical force has been shown to regulate periodontal ligament cells (PDLs) behaviors. However, different force types lead to distinct PDLs’ responses. Here, the differential gene expression profiling of PDLs subjected to static and intermittent compressive forces was examined using RNA sequencing technique. Results demonstrated that the static and intermittent compressive force treated PDLs exhibited the differential regulation genes. KEGG pathway enrichment analysis revealed that focal adhesion and transforming growth factor beta signaling pathway were commonly upregulated while calcium signaling pathway was downregulated in both static and intermittent compressive force-treated PDLs. Interestingly, Wnt signaling pathway was upregulated only in the PDLs that subjected to the intermittent compressive force.

Compressive Force Induces Differential Gene Expression in Uterine Fibroids

Project description:We investigated how compressive force influences fibroid and myometrial cells. Patient-matched cells were isolated from women undergoing hysterectomy or myomectomy for the treatment of uterine fibroids. Samples from five women with no hormonal modulation within three months of surgery were used for this study. Spheroids fabricated from fibroid or myometrial cells were embedded in agarose hydrogels and exposed to compressive force for seven days. RNA sequencing was performed to identify genes and signaling pathways altered in fibroids in response to compression.

Effects of dynamic compressive loading on MSC chondrogenesis

Project description:Long-term dynamic compression enhanced the mechanical properties of MSC-seeded constructs only when loading was initiated after 21 days of chondrogenic differentiation. This study examined the molecular differences of chondrogenic MSCs compared to undifferentiated MSCs (TGF-beta vs no TGF-beta) and the effects of dynamic loading on MSC chondrogenesis (loading vs free-swelling). Free-swelling MSC-seeded constructs were cultured for 21 days in chemically defined media. Chondrogenesis was induced with TGF-beta3. Undifferentiated controls were maintained in parallel. After 21 days of chondrogenic differentiation, a subset of constructs were subjected to 21 days of dynamic compressive loading. On days 21 and 42, construct mechanical properties and biochemical content were assessed. Microarray analysis was carried out on day 3, day 21 and day 42 constructs. 6 arrays.
