Project description:Osteoporosis is characterized by bone loss, reduced bone strength, and increased fracture risk. Studies have shown that epigenetic mechanisms play a role during bone formation and bone-related disorders, including osteoporosis. However, the contribution of bivalent domains, which are chromatin regions that are co-occupied by the gene activating Histone 3 lysine 4 trimethylation (H3K4me3) and the gene suppressive Histone 3 lysine 27 trimethylation (H3K27me3) marks, has not been elucidated during osteogenesis. Previous work demonstrated that the enhancer of zeste homolog 2 (Ezh2), a histone 3 lysine 27 (H3K27) methyltransferase, suppressed differentiation of osteogenic progenitor cells and that a small molecule inhibitor of Ezh2 (GSK126) enhanced osteogenic commitment of osteoblast progenitors in vitro and bone formation in vivo. Tazemetostat (or EPZ6438) is an Ezh2 inhibitor (iEzh2) that is FDA approved for the treatment of epithelioid sarcoma and might be repurposed to mitigate bone loss. The present study aimed to elucidate mechanisms by which Tazemetostat-mediated H3K27me3 reduction regulates bivalent domain associated enhancers critical for bone formation. Tazemetostat enhanced the expression of bone-related genes (e.g., Runx2-p57, Bglap, Ibsp) as measured by qPCR analysis and stimulated extracellular matrix deposition as revealed by alizarin red staining of cell cultures. Western blotting analysis revealed that Tazemetostat reduced H3K27me3 levels and concomitantly enhanced H3K27Ac, as well as increased protein levels of the osteoblastic master regulator Runx2. Integrative analysis of RNA-seq and ChIP-seq datasets revealed that upregulation of group of genes that are associated with bivalent domains. Importantly, gene ontology analysis showed that genes associated with Wnt, Pth and Hh signaling pathways are enriched within these up-regulated genes. Follow-up mechanistic-based studies revealed that the pro-osteogenic effects induced by Ezh2 inhibition are attenuated by inhibition of Hh signaling and enhanced by disruption of Wnt signaling. Our data demonstrate that the genes controlled by bivalent domains are associated with key osteogenic signaling pathways and are induced upon Ezh2 inhibition and subsequent H3K27me3 reduction. Our data also demonstrate that iEzh2-induced osteogenic differentiation is modulated by Wnt and Hh signaling pathways.

Project description:Bivalent epigenomic regulatory domains containing both activating histone 3 lysine 4 (H3K4me3) and repressive lysine 27 (H3K27me3) trimethylation are associated with key developmental genes. These bivalent domains repress transcription in the absence of differentiation signals but maintain regulatory genes in a poised state to allow for timely activation. Previous studies demonstrated that enhancer of zeste homolog 2 (Ezh2), a histone 3 lysine 27 (H3K27) methyltransferase, suppresses osteogenic differentiation and that inhibition of Ezh2 enhances commitment of osteoblast progenitors in vitro and bone formation in vivo. Here, we examined the mechanistic effects of Tazemetostat (EPZ6438), an Food and Drug Administration approved Ezh2 inhibitor for epithelioid sarcoma treatment, because this drug could potentially be repurposed to stimulate osteogenesis for clinical indications. We find that Tazemetostat reduces H3K27me3 marks in bivalent domains in enhancers required for bone formation and stimulates maturation of MC3T3 preosteoblasts. Furthermore, Tazemetostat activates bivalent genes associated with the Wingless/integrated (WNT), adenylyl cyclase (cAMP), and Hedgehog (Hh) signaling pathways based on transcriptomic (RNA-seq) and epigenomic (chromatin immunoprecipitation [ChIP]-seq) data. Functional analyses using selective pathway inhibitors and silencing RNAs demonstrate that the WNT and Hh pathways modulate osteogenic differentiation after Ezh2 inhibition. Strikingly, we show that loss of the Hh-responsive transcriptional regulator Gli1, but not Gli2, synergizes with Tazemetostat to accelerate osteoblast differentiation. These studies establish epigenetic cooperativity of Ezh2, Hh-Gli1 signaling, and bivalent regulatory genes in suppressing osteogenesis. Our findings may have important translational ramifications for anabolic applications requiring bone mass accrual and/or reversal of bone loss.

Project description:Bivalent domains marked with repressive H3K27me3 and activating H3K4me2/3 are a molecular signature of totipotency in stem cells and development. While bivalent domains are retained throughout the germline to recover totipotency in the next generation, the mechanisms establishing bivalent domains remains unknown. Here we demonstrate that a germline-specific Polycomb protein, SCML2, binds to chromatin containing hypomethylated DNA to induce H3K27me3, thereby initiating the establishment of germline-specific bivalent domains in mice. SCML2 regulates two distinct classes of autosomal bivalent domains, the first are maintained constitutively through spermatogenesis (Class I), and the second are specifically established during meiosis (Class II). In postmeiotic spermatids, the loss of H3K27me3 leads to an increase of H3K4me2/3 on bivalent domains and disorganization of pericentromic heterochromatin. We propose that SCML2 regulates dynamic bivalent domains in the germline as a molecular imprint to recover totipotency after fertilization.

However, how their epigenetic status might affect their potential to differentiate toward specific lineages remains a puzzling question. Using ChIP-on-chip, the status of bivalent domains on gene promoters (i.e. H3K4 and H3K27 trimethylation) was monitored for both undifferentiated and BMP2 induced cardiac committed cells. A marked difference in the epigenetic profile of HUES cell lines was observed and this was correlated to the pattern of gene expression induced by BMP2 as well as to their potential to generate cardiac progenitors and differentiated myocytes. Thus, the epigenetic H3trimeK4 and H3trimeK27 prints generating bivalent domains on promoters, could be used to predict a preference in their differentiation toward a specific lineage. Project description:The male germline transcriptome changes dramatically during the mitosis-to-meiosis transition to activate late spermatogenesis genes and to transiently suppress genes commonly expressed in somatic lineages and spermatogenesis progenitor cells, termed somatic/progenitor genes. These changes reflect epigenetic regulation. Induction of late spermatogenesis genes during spermatogenesis is facilitated by poised chromatin established in the stem cell phases of spermatogonia, whereas silencing of somatic/progenitor genes during meiosis and postmeiosis is associated with formation of bivalent domains which also allows the recovery of the somatic/progenitor program after fertilization. Importantly, during spermatogenesis mechanisms of epigenetic regulation on sex chromosomes are different from autosomes: X-linked somatic/progenitor genes are suppressed by meiotic sex chromosome inactivation without deposition of H3K27me3.
