Project description:Purpose: In this study we investigated the role of JASMONATE RESISTANT 1 (JAR1) and JAR1 mediated JA-Ile formation in drought stress tolerance in Arabidopsis thaliana. Methods: Global transcriptional changes in a newly generated over-expression line (JAR1-OE; 35S::JAR1-1-YFP)), a T-DNA insertion line in the JAR1 locus (jar1-11;SALK_034543), and wild-type Col-0 were investigated by RNA-seq analyses of rosette leaves from 32 day-old plant that were either well-watered (control) or not watered after day 18 (drought). Plants were grown on soil under long-day conditions Results: Under control conditions, using a stringent cut-off (DESeq, adjusted to FDR < 0.01 and LogFC ≥ 1), we found only four differentially expressed genes (DEGs) between jar1-11 and Col-0, all of them downregulated. By contrast, we found 339 DEGs between JAR1-OE and Col-0, of which 134 were downregulated and 205 were upregulated. A comparison of the RNA-seq data from Col-0 between control and drought conditions revealed 3401 DEGs, of which 2023 were down- and 1378 upregulated. By comparison, jar1-11 plants, which were most heavily affected by drought stress, showed a much higher number (6139 in total; 2616 up- and 3523 down-regulated) of DEGs, while the more drought-tolerant JAR1-OE line displayed a lower number (2025 in total; 971 up- and 1054 down-regulated) of DEGs. 2411 DEGs were found between Col-0 and jar1-11 under drought among which 966 genes showed a higher and 1445 genes a lower expression level in jar1-11. On the other hand, out of 998 DEGs found between Col-0 and JAR1-OE under drought, 737 genes showed a higher and 261 genes a lower expression level in JAR1-OE. Moreover, we found 391 DEGs counter-regulated between jar1-11 and JAR1-OE. Conclusion:RNA-seq analysis and additional experiments of plants under control and drought stress conditions provided insight into the molecular reprogramming caused by the alteration in JA-Ile content.

Project description:Jasmonates regulate plant development and defence. In angiosperms, the canonical bioactive jasmonate is jasmonoyl-isoleucine (JA-Ile), which is formed in Arabidopsis thaliana by JAR1 and GH3.10. In contrast to other jasmonate biosynthesis or perception mutants, however, gh3.10 jar1 knockout lines are still fertile. Therefore we investigated whether further jasmonates and GH3 enzymes contribute to regulation of fertility. This study found JAR1 and GH3.10 are the only contributors to JA-Ile biosynthesis in Arabidopsis and identified a number of JA conjugates as potential bioactive jasmonates acting in the absence of JA-Ile. However, their contribution in regulating fertility is yet to be conclusively determined. Transcriptomic analysis did not identify further GH3 enzymes with increased transcription rate in flowers of gh3.10 jar1 plants.

Bioactive jasmonates, such as JA, JA-Ile, OPDA and dn-OPDA, are derived from C18- and C16- fatty acids (FAs), which are found ubiquitously in the plant kingdom. Bryophytes also contain long chain C20- and C22- FAs (LCFAs), which are absent in most vascular plants but are found in organisms of other king-doms, including mammals. The existence of bioactive jasmonates derived from LCFAs is cur-rently unknown. Here, we describe the identification of an OPDA-like molecule derived from a C20-FA in Marchantia polymorpha, which we term C20-OPDA. This molecule accumulates upon wounding, and when applied exogenously can activate known COI1-dependent and -independent jasmonate responses. Furthermore, we identify a dn-OPDA-like molecule (Δ4-dn-OPDA) deriving from C20-OPDA and demonstrate it to be a ligand of the jasmonate co-receptor (MpCOI1-MpJAZ) in Marchantia. By analysing mutants involved in the production of LCFAs, we elucidate the major biosynthetic pathway of C20-OPDA and Δ4-dn-OPDA. Moreover, using a double mutant compromised in the production of both Δ4-dn-OPDA and dn-OPDA, we demonstrate the additive nature of these molecules in the activation of jasmonate responses. Taken together, our data identify LCFAs as a source of bioactive jasmonates that are essential to plant plasticity and resilience.

Project description:Photoperiodic floral initiation in the leaf is controlled by the hub gene CONSTANS (CO) while jasmonates (JAs) control flower senescence. Although both processes are chronologically ordered, no association between them has been described to date. We show that CO protein remains in Arabidopsis flowers after the floral induction, although displaying a different tissue and diurnal pattern than in leaves. We found that changes in CO expression alter flower senescence and abscission by interfering with the JA response, supported by petal specific analysis as well as CO overexpression in JA synthesis and signaling mutants. CO has a ZIM-like domain that mediates interaction with the JA response repressor JAZ3 (jasmonate ZIM-domain 3), inhibiting its repressor activity and activating downstream transcription factors involved in flower senescence. The complex CO-JAZ3 also interacts with the E3 ubiquitin ligase Coronatine Insensitive 1 (COI1) leading to its degradation in the presence of jasmonates. Therefore, the coordinated recruitment of photoperiodic and jasmonate signaling pathways would be an efficient way for plants to chronologically order the floral process and ensure the success of offspring production.

Project description:Jasmonates are well known signaling components required for diverse processes ranging from wound and pathogen responses to regulation of developmental processes. A prominent feature of jasmonate biosynthesis or signaling mutants is the loss of fertility. In contrast to the male sterile phenotype of Arabidopsis mutants, the loss of the co-receptor protein COI1 in the tomato mutant jai1-1 results in female sterility with additional severe effects on stamen and pollen development. While a general model of jasmonate effects on male gametophyte development has been proposed to involve the regulation of water transport, the molecular details of this response are scarce. Here we show an extensive temporal profiling of the development, hormone content, transcriptome and metabolome of tomato stamen in six distinct stages of flower development in wild type and jai1-1. We found that the premature stamen desiccation of jai1-1 stamens and the preponed pollen maturation coincide with an accumulation of desiccation-related metabolites. The wild type shows a transient increase of jasmonates up to mid-flowering that is absent in jai1-1. This finding coincides with an early increase of the ethylene precursor ACC in jai1-1 that is similarly reflected in the increased expression of ethylene biosynthesis and response genes. Our data suggests an essential role of jasmonates in the temporal inhibition of ethylene production to prevent premature desiccation of stamens and to ensure proper timing in flower development.

Methyl jasmonate is an elicitor that activates a suite of defense responses including phenylpropanoid metabolism. In contrast, AOPP acts as an inhibitor of phenylalanine ammonia lyase (PAL). Samples were harvested 48 hrs after initiation of the experiment. Total RNA was extracted and gene expression measured using Affymetrix poplar genome microarrays.

Project description:The maintained vegetative phase (mvp) mutant has a non-flowering phenotype caused by deletions including, but not limited to, the genes CYS, PHYC and VRN1. However the impact of these deletions on flowering genes and global gene expression is still unknown. In this study, we showed that these deletions caused the up-regulation of several pathogenesis related (PR) and jasmonate responsive genes using microarray analysis. Our results raise the hypothesis that jasmonates may be involved in flowering. To confirm this hypothesis, the methyl-jasmonate (MeJA) and jasmonic acid (JA) content in mvp and wild type plants was measured. The content of JA was comparable in all plants while the content of MeJA was higher in mvp plants. Our transcriptomic and chemical analyses of the mvp mutants plants reveal that the deletion of the major vernalization gene TaVRN1 induces the up-regulation of the major biotic stress related genes and the accumulation of MeJA. In addition, our results demonstrate an important role of MeJA during vernalization and flowering in wheat.
