Project description:Plant premature senescence is a major reason for agricultural losses caused by abiotic stress. Tools for suppressing stress-induced plant senescence are important but rare. Herein we report that diacetyl, a natural compound emitted by a plant-beneficial bacterium, suppresses ABA-mediated foliar senescence in Arabidopsis thaliana under various abiotic stress conditions. Our results established diacetyl as an effective protector of stress-induced plant senescence and disclosed a molecular mechanism for bacteria-enhanced plant stress-resistance.

Project description:Alveolar rhabdomyosarcoma (aRMS) is an aggressive sarcoma of skeletal muscle characterized by expression of the PAX3-FOXO1 fusion gene. Despite its discovery over almost 20 years ago, PAX3-FOXO1 remains an enigmatic tumor driver. Previously, we reported that PAX3-FOXO1 supports aRMS initiation by enabling bypass of cellular senescence. Here, we show that bypass occurs in part by PAX3-FOXO1-mediated upregulation of RASSF4, a Ras-association domain family (RASSF) member, which then suppresses the evolutionarily conserved mammalian Hippo/Mst1 pathway. RASSF4 loss-of-function activates Hippo/Mst1 and inhibits downstream YAP, causing aRMS cell cycle arrest and senescence. This is the first evidence for an oncogenic role for RASSF4, and a novel mechanism for Hippo signaling suppression in human cancer. Human skeletal muscle myoblasts (HSMMs) were retrovirally transduced with either an empty vector (Vp, pK1) or PAX3-FOXO1 (PFp, pK1-PAX3-FOXO1) and selected on puromycin. Presenescent (presen) cells were harvested before the senescence checkpoint. Project description:Arabidopsis microRNA expression regulation was studied in a wide array of abiotic stresses such as drought, heat, salinity, copper excess/deficiency, cadmium excess and sulphur deficiency. A home-built RT-qPCR mirEX platform for the amplification of 289 Arabidopsis microRNA transcripts was used to study their response to abiotic stresses. Small RNA sequencing and Northern hybridization were performed to study the expression of mature microRNAs. In the case of common climate change related stresses such as drought, heat and salinity we observed broad induction of the level of primary miRNAs that was not observed at the level of microRNAs.. In the case of local soil pollution stresses, that are represented by heavy metal contaminations or deprivation of a specific micro- or macroelement, the transcriptional response of pri-miRNAs was quite limited but also not predictive to the level of the mature microRNA. This points to an essential role of posttranscriptional regulation of microRNAs expression. We found that the level of several microRNAs can be differentially regulated in early and late response to stress. New Arabidopsis microRNAs responsive to abiotic stresses were discovered. Three microRNAs: miR319a/b, miR319b.2, and miR400 have been found to be responsive in several abiotic stresses and thus can be regarded as a general stress-responsive microRNA species. Additionally, a new target for miR319b.2 – TBL10 has been experimentally confirmed. However, its level under different abiotic stresses is unchanged in comparison to control conditions. In the promoter region of the TBL10 gene we found the presence of many stress-responsive elements. We suggest that transcriptional induction resulting in the increase of transcript levels is downregulated by the increase of the miR319b.2 ultimately resulting in a stable level of TBL10 mRNA. Our experiments show the existence of a complex regulatory network involved in the microRNA level control that is necessary to fine-tune plant response to environmental cues.

Here we show that a subset of short-chain leaf volatiles with an α,β-unsaturated carbonyl bond in their structure (reactive short-chain leaf volatiles, RSLVs) like (E)-2-hexenal and (E)-2-butenal can act as signal chemicals that strongly induce the gene expression of abiotic-related transcription factors, such as heat stress-related transcription factors (HSFA2, MBF1c) and other abiotic stress-related transcription factors (DREB2A, ZATs). RSLV-induced expression of HSFA2 and MBF1c was eliminated in HSFA1s-, known as heat stress response master regulators, knockout mutant, whereas those of DREB2A and ZATs were not, suggesting that the RSLV signaling pathway is composed of HSFA1-dependent and -independent pathways. RSLV treatment induced production of chaperon proteins, and the RSLV-treated Arabidopsis thus demonstrated enhanced abiotic stress tolerance. Because oxidative stress treatment enhanced RSLV production, we concluded that commonly found RSLVs produced by environmental stresses are powerful inducer of abiotic stress-related gene expression as oxidative stress signals.

Project description:We report the identification of RCI5, an Arabidopsis cold inducible gene encoding a FMO. RCI5 seems to participate in the biosynthesis of TMAO, a new plant methabolite. We also demonstrate that TMAO positevely controls Arabidopsis tolerance to low temperature, high salt in the soil and drough stress, by promoting a wide transcriptomic reprograming of stress-relate genes. Finally, we found that diferent crops also contain TMAO in their tissues, and that exogenous applications of TMAO also increases tomato tolerance to abiotic stress.

Project description:Plants physiological mechanisms are affected by Abscisic acid (ABA) via changing gene expression and empowers plants to adapt in numerous environments. Plants have evolved their protection mechanisms for seed germination and abiotic environments to deal with critical harsh conditions. Here, we have explored those changes in Arabidopsis thaliana plants subjected to multiple abiotic stresses. AtBro1 transcripts showed up-regulation in the presence of salt, ABA and mannitol stress conditions. AtBRO1 over-expression lines exhibited robust tolerance to drought and salt stress. Further, ABA elicits resistance responses in loss-of-function bro1-1 mutant plants and AtBro1 positively regulates drought resistance in Arabidopsis. Promoter of AtBro1 fused with GUS showed GUS expression mainly in the rosette leaves and floral clusters, especially in anthers. AtBro1 protein was found to be localized in the plasma membrane of Arabidopsis at the subcellular level by using AtBro1::GFP fusion. Using a broad RNA-sequencing analysis, we observed that early transcriptional responses prompted by ABA induction exhibit specific quantitative differences at different time points, suggesting that ABA stimulates resistance responses in bro1-1 mutant plants. Additionally, transcripts levels of MOP9.5, MRD1, HEI10, and MIOX4 were altered in loss-of-function mutant plants which are involved in different stress conditions. Collectively, our results have shown that AtBro1 is involved in a significant role by regulating plant transcriptional response to ABA and induction of resistance response against abiotic stress.

Project description:The WRKY gene family has a very ancient origin but has faced extensive duplication only in the plant kingdom so much that Arabidopsis (Arabidopsis thaliana) has 74 copies of WRKY genes encoding transcription factors while 109 can be found in Rice (Oryza sativa L.). Several studies in the last decade has pointed their involvement in an heterogeneous number of biological processes, from development to hormone signalling, dormancy and senescence, but a wide number of WRKY genes are transcriptionally regulated during biotic or abiotic stresses. To investigate involvement of WRKY genes upon host and non-host infection (different strain of Magnaporthe grisea) and osmotic stress in Rice, we performed a gene family transcription analysis using custom microarray. Results indicate that a relevant part of WRKY genes are involved during at least one of these stresses, that there is little difference in transcriptional regulation between host and non-host infection or between different tissues upon the same osmotic stress. Moreover, are evident groups of genes that, often with opposite behaviour, are co-regulated in all or most of the studied conditions. We thus formulated the hypothesis that WRKY genes might be part of co-regulatory networks with other WRKY genes. Keywords: stress response
