Project description:We profile the transcriptomes of over 20,000 mouse single cells, yielding molecular definitions for mesenchymal and epithelial cell types present in the mouse uterus during post-menstrual tissue repair. We identify a novel population of fibroblasts that express genetic markers associated with both mesenchymal and epithelial phenotypes in the repairing mouse uterus. Further trajectory analysis indictates a differentiation trajectory between mesenchymal and epithelial cells indicative of a putative mesenchymal-to-epithelial transition (MET) event occuring during post-menstrual tissue repair. Our work provides a platform to further understand the roles of these cell populations in uterine physiology and disease and for comparisons between wound healing events in the endometrium and other adult tissue which are prone to fibrosis.

Single-cell RNA sequencing of human endometrium across the luteal phase of the menstrual cycle

Project description:The fate of the human endometrium is determined during the mid-luteal window of implantation, a crucial period when endometrial stromal cells (EnSCs) differentiate into specialized decidual cells. Upon embryo implantation, these differentiating EnSCs transform the endometrium into the decidua of pregnancy. Conversely, in the absence of pregnancy, decreasing progesterone levels trigger tissue breakdown, leading to menstruation. Despite our understanding of these processes, the precise mechanisms governing this tissue transformation remain elusive. To bridge this knowledge gap, we conducted single-cell RNA sequencing, mapping the transcriptomic profiles of timed endometrial biopsies throughout the luteal phase of the menstrual cycle.

Transcription profiling of human endometrial endothelial cells of eutopic endometrium of patients with endometriosis compared with control

Project description:The pathogenesis of endometriosis may result from aberrant angiogenesis that occurs in eutopic endometrium with retrograde menstruation. The difference in gene expression profile between human endometrial endothelial cells (HEECs) from eutopic endometria of patients with and without endometriosis would be determinant that affects the occurrence of endometriosis. To explore this kind of difference, we performed in vitro culture and identified their endothelial origin, as well as observed growth features of HEECs from the two different origins. Finally we identified the difference in gene expression profile when combined suppression subtractive hybridization(SSH) with genechip and confirmed the results by quantitative reverse transcription polymerase chain reaction (qRT-PCR). The HEECs derived from endometriosis exhibited potent survival ability in vitro compared to that from non-endometriosis. We found that gremlin and fibronectin genes were up-regulated in HEECs derived from eutopic endometrium of patients with endometriosis when compared with that from patients without endometriosis. Our study implies that enhanced angiogenic capacity of eutopic HEECs may be an independent determinant in endometriotic aberrant angiogenesis in addition to the interaction of exfoliated endometrium and peritoneal environment elements such as activated macrophages and soluble cytokines. Project description:We examined the molecular profile of human endometrium on days 2, 3 and 4 of menstruation to identify genes and pathways that play a role in the menstrual process. Endometrial samples were collected by Pipelle biopsy on days 2 (n=9), 3 (n=9) or 4 (n=6) of menstruation and arrayed. We identified key gene pathways involved in menstruation and identified a number of genes not previously associated with the process (Paiva et al. 2016. Mol Hum Reprod. In press).

Murine model of decidulization and menstruation

Project description:A murine model that mimic the decidualization and regression observed in human was used to investigate the molecular mechanisms underlying the dynamic processes in endometrium. Ovariectomized mice were treated sequentially with steroid hormones and then, to induce decidualization, oil was injected into the uterine lumen. A process similar to menstruation was induced by hormone-withdrawal. The uterine tissues were collected at 4 time-points after the induction of decidualization. Transcriptome analysis of MET-constitutively active versus MET-silenced colorectal cancer cells

We then silenced MET in LoVo cells using CRISPR/Cas9 knock-out (KO) protocol, and performed gene expression profiling analysis using data obtained from RNA-seq of cells with hyperactivated-MET versus MET-KO.
