Project description:Cardiomyocyte-like cells can be reprogrammed from somatic fibroblasts by combinations of genes, providing a new avenue for cardiac regenerative therapy. Here we show that functional cardiomyocytes can be rapidly and efficiently generated from human fibroblasts by specific combination small molecules. Microarray analysis has been used to compare the expression profile of cardiomyocyte-like cells derived from human foreskin and lung fibroblasts, and human ES cell-derived cardiomyocytes.

Gene expression analysis of embryonic photoreceptor precursor cells using BAC-Crx-EGFP transgenic mouse.

Project description:We generated a transgenic mouse line which express EGFP in the retina driven by the Crx promoter using BAC transgenesis. We sorted EGFP-positive photoreceptor precursors at E17.5 using FACS, and subsequently performed microarray analysis of the FACS-sorted cells. In order to clarify a molecular role of Crx in developing cone photoreceptor precursors, we investigated the expression profile of the BAC-Crx-EGFP-positive cells compared with that of the BAC-Crx-EGFP-negative cells at E17.5.

Direct Reprogramming of Human Fibroblasts into Retinal Progenitor Cells by Small Molecules to Treat Photoreceptor Degeneration

Project description:Retinal progenitor cells (RPCs) hold promise for restoring vision in advanced stages of photoreceptor degeneration. We have developed a technique to directly reprogram human Tenon's fibroblasts (HTFs) into induced RPC-like cells (iRPCs) using small molecules. To explore the transcriptomic profile of iRPCs, we performed bulk RNA-seq analysis of HTFs and FACS-sorted iRPCs. The Gene Ontology (GO) enrichment analysis demonstrated significantly highly enriched GO terms include Proteinaceous Extracellular Matrix, Axon and Dendrite, Synaptic Membrane and Transport Vesicle, Presynapse and Postsynapse, Basement Membrane and Collagen Trimer, Platelet Alpha Granule and ER Lumen. The significantly down-regulated GOs were Spindle and Chromosomal Structures, Sarcomere and Muscle Fiber Components, Cell Adhesion Complexes.
