Project description:The target of rapamycin (TOR) kinase is a central signaling hub that plays a crucial role in precisely orchestrating plant growth and development. It is increasingly evident that TOR also has a significant impact on plant stress responses. This suggests that TOR is intricately involved in maintaining the balance between plant growth and stress responses. Nevertheless, despite the observed effects, the specific mechanisms through which TOR operates in these processes remain obscure. In this study, we investigated how the tomato (Solanum lycopersicum) TOR (SlTOR) affects plant growth and cold responses. We demonstrated that the inhibition of SlTOR transcriptionally primes cold stress responses, consequently enhancing tomato cold tolerance. Furthermore, a widely targeted metabolomics analysis revealed the disruption of amino acid metabolism homeostasis under cold stress upon SlTOR inhibition. This disruption led to the accumulation of two important defense metabolites: salicylic acid (SA) and putrescine (Put). Subsequent gain- and loss-of-function analyses validated the crucial roles of salicylic acid and putrescine in enhancing tomato cold tolerance. Our study provides a mechanistic framework that elucidates how SlTOR modulates amino acid-related metabolism to enhance tomato cold tolerance, which sheds light on the complex interplay between growth and stress responses orchestrated by TOR.

Project description:Free and glycosylated sterols play a central role in maintaining the structural integrity and proper functioning of plasma membrane, which serves as the primary sensor of cold and initiates signaling cascades to mitigate chilling-induced damage. Here we characterize the cold response of tomato (Micro-Tom) mutants with opposite glycosylated to free sterol ratio resulting from the overexpression and silencing of the sterol glycosyltransferases SlSGT2 and SlSGT1. A high ratio in the SlSGT2 overexpressing mutants increases cold tolerance, membrane stability, and oxidative stress responses, while a low ratio in the SlSGT1-silenced mutants causes the opposite phenotypes. The different cold sensitivity in these mutants is due to the altered membrane sterol profiles, rather than to a different capacity to adjust sterol levels during cold exposure. Changes in the glycosylated to free sterol ratio also trigger distinct transcriptional programs that establish a preconditioned stress-responsive state under basal conditions and a more efficient response under cold exposure in the SlSGT2 overexpressing mutants and compromise the capacity to withstand the effects of cold stress in the SlSGT1-silenced mutants. The SlSGT2 overexpressing mutants accumulate higher basal levels of jasmonates and display enhanced biosynthetic capacity under cold stress compared to SlSGT1-silenced and control plants. The elevated glycosylated to free sterol ratio in SlSGT2 overexpressing mutants facilitates JA biosynthesis and signaling, leading to the activation of cold-responsive genes, including those of the CBF–COR pathway, antioxidant defenses and polyamine biosynthesis. Our findings provide key insights into the mechanisms by which glycosylated sterols improve cold tolerance in tomato.

Project description:Plants orchestrate stress responses through the integration of light and temperature signals, yet the molecular mechanisms involved in this crosstalk are incompletely elucidated. Here, we identify the transcription factor SlWRKY2 as a critical hub linking phytochrome-mediated light perception with cold stress adaptation in tomato (Solanum lycopersicum). Under cold stress, low red/far-red light ratios significantly induce SlWRKY2 expression and protein accumulation, with phytochrome B (SlphyB) repressing and SlphyA promoting this process. Genetic analyses confirm that SlWRKY2 functions downstream of SlphyB and SlphyA, physically interacts with both SlphyB1 and SlphyA, and acts in a regulatory module where SlphyB negatively regulates cold tolerance while SlphyA exerts a positive effect. Moreover, both cold stress and light signals promote the formation of SlWRKY2 condensates consistent with liquid-liquid phase separation (LLPS). Functionally, SlWRKY2 directly interacts with and stabilizes the light-signaling regulator PHYTOCHROME-INTERACTING TRANSCRIPTION 4 (SlPIF4). Furthermore, SlWRKY2 directly activates SlPIF4 transcription, and the two factors function synergistically to enhance the expression of SlCBFs, as evidenced by genetic data indicating that SlPIF4 is required for SlWRKY2-dependent cold tolerance. Mechanistically, SlWRKY2 competes with SlphyB1 for interaction with SlPIF4. Our findings reveal the SlphyB1/A-SlWRKY2-SlPIF4 signaling module as a pivotal mechanism underlying light quality-dependent cold adaptation, advancing our understanding of how plants integrate environmental cues while providing mechanistic insights into how plants integrate environmental cues to fine-tune cold adaptation.

Comparison of fiber cell transcription profiles is between the two germplasm lines and over a developmental time-course from 8 to 24 days post anthesis in four day intervals. Cotton plants grown in 3-4 row plots of approximately 300-400 individual plants. Bulked fiber samples from multiple plants per each plot represented a biological replication. There were 3-4 spatially distinct plots per cotton germplasm line. Loop microarray hybridization experimental design. Biological replicates: 2 for each germplasm line at each time-point. Project description:Plants have evolved mechanisms to perceive and face mechanical stress (MS) caused by physical forces, including compacted soils, winds, rain, pathogens, and interactions with animals and plants. Previous research indicated that applying mechanical treatment (MT) to Arabidopsis increases both xylem area and seed yield. To explore sustainable tomato production, we applied MT - combining stem bending, weighting, and particular touching - to 10-day-old seedlings, using a specific weight on the upper stem for 48 h. Two days after the treatment, we observed stem enlargement and increased the number of xylem vessels and area in MT plants. Additionally, we noticed earlier flowering, leading to increased tomato production. The transcriptome of MT-treated plants revealed significant changes in the expression of several essential genes involved in central metabolism, growth responses, and crucial phytohormone signalling. By studying different tomato mutants in the ethylene and auxin signalling pathways, we demonstrated that both hormones play essential roles in the plant responses to combined MT. Our findings suggest that combined MT generates a beneficial MS in tomato plants that induces plant morphoanatomical changes that promote early flowering and increased yield, providing a promising strategy for sustainable agriculture.
