Valorization of seaweed hydrolysate for production of the high-value reverse antibiotic nybomycin using metabolically engineered Streptomyces explomaris

Project description:This study develops a pipeline for high-level production of the reverse antibiotic nybomycin from three seaweed species: Himanthalia elongata, Palmaria palmata, and Ulva lactuca. Screening Streptomyces strains identified S. explomaris, a marine species, as the best host to express the nybomycin gene cluster. The accumulated low yields in artificial seawater with brown seaweed hydrolysate. Gene expression analysis revealed downregulation of precursor supply pathways and upregulation of repressors, limiting biosynthesis. Metabolic engineering addressed these bottlenecks, leading to a superior S. explomaris mutant achieving 57 mg/L, a five-fold increase as compared to reported yields. The strain effectively valorized commercial seaweed hydrolysates, highlighting marine feedstocks' potential for antibiotic production.

Project description:The algal holobiont, which consists of algae and the bacteria living on it, has developed strategies to handle environmental stresses, such as changes in temperature. The green alga Ulva (Chlorophyta) is home to bacteria that help the algae grow and develop, from germlings to mature plants. These bacteria release chemicals like thallusin, N-acyl homoserine lactone, and ectoine, which support the health and growth of the algae. However, there is limited research on the metabolome of cold-adapted Ulva holobionts, especially those from hard-to-reach regions like Antarctica, and how they respond to environmental changes. In this study, we examine the metabolites found in Ulva and its associated bacteria from Antarctica (Potter Cove) using high-resolution mass spectrometry. We identified small polar and zwitterionic metabolites, such as cysteinolic acid, ectoine, glutamine, glycerol, and proline, in the surface tissues of the algae with MALDI-MS. By profiling these metabolites using UHPLC-HRMS and ZIC-HILIC chromatography, we found that their levels change significantly in response to heat-stressed Antarctic Ulva holobionts. Our results suggest that environmental change can affect the interaction between seaweeds and their microbes by altering the metabolites involved in their chemical exchanges. The Data contains the Ulva metabolome profiles (.mzXML) analyzed using ZIC-HILIC chromatography coupled with HRMS (QExactive). Acquisition was conducted with the polarity switch mode and ddMS acquisition was done on the QC pool samples. Integrated transcriptomics and proteomics analysis reveals muscle metabolism effects of dietary Ulva lactuca and ulvan lyase supplementation in weaned piglets

Project description:Seaweeds, including the green Ulva lactuca, can potentially reduce competition between feed,food, and fuel. They can also contribute to the improved development of weaned piglets. However,their indigestible polysaccharides of the cell wall pose a challenge. This can be addressed throughcarbohydrase supplementation, such as the recombinant ulvan lyase. The objective of our study wasto assess the muscle metabolism of weaned piglets fed with 7% U. lactuca and 0.01% ulvan lyasesupplementation, using an integrated transcriptomics (RNA-seq) and proteomics (LC–MS) approach.Feeding piglets with seaweed and enzyme supplementation resulted in reduced macronutrientavailability, leading to protein degradation through the proteasome (PSMD2), with resulting aminoacids being utilized as an energy source (GOT2, IDH3B). Moreover, mineral element accumulationmay have contributed to increased oxidative stress, evident from elevated levels of antioxidantproteins like catalase, as a response to maintaining tissue homeostasis. The upregulation of the geneAQP7, associated with the osmotic stress response, further supports these findings. Consequently, anincrease in chaperone activity, including HSP90, was required to repair damaged proteins. Our resultssuggest that enzymatic supplementation may exacerbate the effects observed from feeding U. lactucaalone, potentially due to side effects of cell wall degradation during digestion.
