Project description:To investigate the effects of oxidative stress induced by hydrogen peroxide(H2O2) on miRNA expression in porcine granulosa cells,we examined the impact of H2O2-mediated oxidative stress in procine GC through miRNA-Seq. We identified 33 (19 upregulated, 14downregulated) and 22 (14 upregulated, 8 downregulated miRNAs) differentially expressed miRNAs (DEmiRNAs) at 300μM and 100μM H2O2, respectively, compared with the control group.

Gene expression profile in porcine ovarian granulosa cells during short-term in vitro culture

Project description:The porcine ovarian granulosa cells, when analysed during in vitro cultures, may provide new interesting data on the processes associated with in vitro culture of porcine oocytes, as well as assisted reproduction and in vitro fertilisation processes used in the livestock industry. Porcine ovarian granulosa cells were obtained from 43 crossbred Landrace gilts with a median age of 170 days and weight of 98 kg after slaugheter. The cells were harvested after 0h, 48h, 96h and 144h after begining of culture and submited to RNA isolation and microarray analisys. In this study, we demonstrated the gene expression profile of short term primary cultured porcine ovarian granulosa cells.

Transcriptomic signature of in vitro heat stress challenged porcine granulosa cells

Project description:Exploration of heat stress induced genes in granulosa cells is critical to characterization of candidate genes for thermal stress research in pigs. This study aimed to gain insight into the differentially expressed genes (DEGs) and signalling pathways regulating porcine granulosa cell adaptation to in vitro heat stress challenge at 42OC for 6 hours. Transcriptome profiling of heat stressed (treated) and non-heat stressed (control) granulosa cells was conducted using Illumina NextSeq2000 sequencing platform. The significant DEGs were selected using NOISeq R package with cut-offs, probability value ≥ 0.95 and absolute log2(fold change) ≥1. Bioinformatics analysis of DEGs were conducted to explore functional annotations enrichment, protein-protein interaction network and hub genes regulating the cellular homeostasis and survivability during heat stress challenge.

Time course analysis of dedifferentiation in porcine follicular granulosa cells

Project description:Cellular dedifferentiation signifies the withdrawal of cells from a specific differentiated state into a M-bM-^@M-^Xstem cellM-bM-^@M-^Y-like undifferentiated state. However, the mechanism of dedifferentiation remains obscure. We showed that follicular granulosa cells (GC), which have distinct functions in vivo, can dedifferentiate during culture in vitro and acquire multipotency. We investigated the dedifferentiation of GC using global gene expression analyses. Total RNA was isolated from GCs to DFOG cells at 5 time points during dedifferentiation (cultured in 0day, 1day, 2day, 4day and 7day). Each timepoint was performed in triplicate (ie, biological replicates). Using Affymetrix porcine genome array, we performed microarray time course experiments to analyze gene expression profiles during GC dedifferentiation.
