Project description:To study how exercise induces bone remodeling in diet-induced obesity in a comprehensive way.8-week male C57/B6 mice were fed with HFD to establish an obese mice model. Then mice were randomly divided into 2 groups: the exercise and the control groups. The exercise group of mice was put on a moderate-intensity treadmill running for 12 weeks.We employed unbiased RNA-Seq to comprehensively investigate the exercise effect on the bone marrow of diet-induced obesity mice in vivo.

Exercise Promotes Bone Marrow Microenvironment by Inhibiting Adipsin in Diet-Induced Male Obese Mice.

Project description:Obesity is a growing global epidemic linked to many diseases, including diabetes, cardiovascular diseases, and musculoskeletal disorders. Exercise can improve bone density and decrease excess bone marrow adipose tissue (BMAT) in obese individuals. However, the mechanism of exercise regulating bone marrow microenvironment remains unclear. This study examines how exercise induces bone marrow remodeling in diet-induced obesity. We employed unbiased RNA-Seq to investigate the effect of exercise on the bone marrow of diet-induced obese male mice. Bone mesenchymal stem cells (BMSCs) were isolated to explore the regulatory effects of exercise in vitro. Our data demonstrated that exercise could slow down the progression of obesity and improve trabecular bone density. RNA-seq data revealed that exercise inhibited secreted phosphoprotein 1 (Spp1), which was shown to mediate bone resorption through mechanosensing mechanisms. Interactome analysis of Spp1 using the HINT database showed that Spp1 interacted with the adipokine adipsin. Moreover, exercise decreased BMAT, which induced osteoclast differentiation and promoted bone loss. Our study reveals that exercise improves the bone marrow microenvironment by at least partially inhibiting the adipsin-Spp1 signaling pathway so as to inhibit the alternative complement system from activating osteoclasts in diet-induced obese mice.

Transcription profiling of soleus muscle from lean and obese Zucker rats following exercise training

Project description:Endurance exercise training has been shown to decrease whole-body and skeletal muscle insulin resistance and increase glucose tolerance in conditions of both pre-diabetes and overt type 2 diabetes. However, the adaptive responses in skeletal muscle at the molecular and genetic level for these beneficial effects of exercise training have not been clearly established in an animal model of pre-diabetes. The present study identifies alterations in skeletal muscle gene expression that occur with exercise training in pre-diabetic, insulin-resistant obese Zucker (fa/fa) rats and insulin-sensitive lean Zucker (Fa/-) rats. Treadmill running for up to 4 weeks caused significant enhancements of glucose tolerance as assessed by the integrated area under the curve for glucose (AUCg) during an oral glucose tolerance test in both lean and obese animals. Using microarray analysis, a set of only 12 genes was identified as both significantly altered (>1.5-fold change relative to sedentary controls; p<0.05) and significantly correlated (p<0.05) with the AUCg. Two of these genes, peroxisome proliferator-activated receptor-g coactivator 1a (PGC-1a) and the z-isoform of protein kinase C (PKC-z), have known involvement in the regulation of skeletal muscle glucose transport. We confirmed that protein expression levels of PGC-1a and PKC-z were positively correlated with the mRNA expression levels for these two genes. Overall, this study has identified a limited number of genes in soleus muscle of lean and obese Zucker rats that are associated with decreased insulin resistance and increase glucose tolerance following endurance exercise training. These findings could guide the development of pharmaceutical M-^Sexercise mimeticsM-^T in the treatment of insulin-resistant, pre-diabetic or overtly type 2 diabetic individuals.

Exercise rescues aberrantly expressed genes in diet-induced obese rats: a potential role of miR-211 modulation in the hypothalamic arcuate nucleus

Project description:Obesity is a major public health concern that is associated with negative health outcomes. Exercise and dietary restriction are commonly recommended to prevent, or combat obesity. The purpose of this study was to examine the mechanism by which aberrantly expressed genes in the hypothalamic arcuate nucleus (ARC) of diet-induced obese (DIO) rats is attenuated through voluntary exercise. Methods: A transcriptomic approach assessed novel genes in the ARC that affect DIO by voluntary wheel running and a combined approach of physiology, pharmacology, transcriptomic and bioinformatics analysis were used to evaluate the role of miR in the reversal of obesity. Results: Exercise attenuated body weight gain, decreased fat mass, and restored gene expression in the ARC. As high-fat diet (HFD) consumption can cause dysregulation of satiety/hunger regulation mechanisms in the ARC, we outlined transcriptional clusters which depicted alterations in patterns of gene expression through running. Among them were genes involved with inflammation and cellular structure. To uncover regulatory mechanisms governing gene expression in DIO attenuation, we explored the role of miR-211, as it has been implicated in systemic inflammation. The DIO overexpression of miR-211 was ameliorated by exercise, demonstrating its pivotal role in the regulation of inflammation in the ARC. Further, in vivo central administration of miR-211-mimic affected the expression of immunity and cell cycle related genes. By comparing genes attenuated by running and genes downregulated by miR-211, we produced a list of candidate genes that might be involved in the modulation by miR-211 in attenuating obesity by exercise. Conclusion: This research suggests that exercise may have a rescue effect on obesity through changes in gene expression that are mediated partially though miR-211.

Gene expression changes in adipose tissue with diet- and/or exercise-induced weight loss

Subcutaneous adipose-tissue biopsies were performed at baseline and after 6 months and changes in adipose-tissue gene expression were determined by microarray with an emphasis on pre-specified candidate pathways, as well as by unsupervised clustering. Analyses were conducted first by randomization group, and then by degree of weight change at 6-months in all women combined.
