Project description:Early-life stress (ELS) leads to increased vulnerability to psychiatric illness including cognitive impairment later in life. Neuroinflammatory processes have been implicated in ELS-induced negative health outcomes, but, how ELS impacts microglia, the macrophages of the central nervous system, is unknown. Here, we determined the effects of ELS induced by limited bedding and nesting material during the first week of life (P2 – P9) on the morphology and gene expression of microglia from young (postnatal day (P)9) and adult (P200) mice. ELS led to a change in the proportion of morphological microglia subtypes in adulthood, associated with immune reactivity. At the transcriptional level, whereas at P9 no ELS mediated changes were detected, at P200 ELS induced transcriptional changes in microglia genes involved in the immune response and protein ubiquitination. Additionally, ELS altered microglia gene expression changes during development from P9 to P200 and in response to LPS at P200, in both cases marked by GO-terms associated with the immune response. Concluding, these data show that ELS has persistent effects on the morphology and gene expression program of microglia and results in an altered transcriptional response to a systemic LPS challenge.

Project description:Early-life stress (ELS) leads to increased vulnerability to psychiatric disorders including depression later in life. Neuroinflammatory processes have been implicated in ELS-induced negative health outcomes, but how ELS impacts microglia, the main tissue-resident macrophages of the central nervous system, is unknown. Here, we determined the effects of ELS-induced by limited bedding and nesting material during the first week of life (postnatal days [P]2-9) on microglial (i) morphology; (ii) hippocampal gene expression; and (iii) synaptosome phagocytic capacity in male pups (P9) and adult (P200) mice. The hippocampus of ELS-exposed adult mice displayed altered proportions of morphological subtypes of microglia, as well as microglial transcriptomic changes related to the tumor necrosis factor response and protein ubiquitination. ELS exposure leads to distinct gene expression profiles during microglial development from P9 to P200 and in response to an LPS challenge at P200. Functionally, synaptosomes from ELS-exposed mice were phagocytosed less by age-matched microglia. At P200, but not P9, ELS microglia showed reduced synaptosome phagocytic capacity when compared to control microglia. Lastly, we confirmed the ELS-induced increased expression of the phagocytosis-related gene GAS6 that we observed in mice, in the dentate gyrus of individuals with a history of child abuse using in situ hybridization. These findings reveal persistent effects of ELS on microglial function and suggest that altered microglial phagocytic capacity is a key contributor to ELS-induced phenotypes.

Project description:The purpose of this project was to investigate whole genome expression in primary microglia cultured from homozygous Trem2 R47H knock-in mice versus wild type littermates under basal conditions and in response to IL-4 treatment. The primary phenotype of microglia from Trem2 R47H knock-in mice was decreased expression levels of Trem2. IL-4 induced the expression of an anti-inflammatory programme of genes in microglia, which was attenuated when Trem2 expression was decreased.

Project description:The OPTN gene is linked to neurodegenerative diseases like amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD), which are characterized by chronic microglial activation. Optineurin regulates inflammatory signaling, autophagy, and trafficking, but its role in microglia is not well understood. We used bulk RNA sequencing to profile CRISPR-Cas9-mediated optineurin knockout (KO) and wild-type (WT) BV2 microglia under basal conditions and upon LPS stimulation. At baseline, optineurin KO altered approximately 7% of the transcriptome, with a notable downregulation of type I interferon and antiviral pathways. LPS stimulation in wild-type cells triggered a broad transcriptional shift (~35% of genes). This LPS-induced response was blunted in optineurin-deficient microglia, with ~16% of genes changed relative to the KO baseline. Furthermore, LPS-treated optineurin KO microglia notably diverged from LPS-treated wild-type cells, with ~26% differentially expressed genes. Our findings establish optineurin as a key regulator of the microglial transcriptome. Its loss weakens basal interferon-mediated immune surveillance and decouples the canonical inflammatory response from cell cycle arrest upon activation.

Project description:Microglia, the resident immune cells of the central nervous system (CNS), are important regulators of normal brain activities. In CNS demyelinating diseases like multiple sclerosis (MS), the functions of these cells are of particular interest. Based on the observation of diminished Dicer positive microglia/macrophages number in demyelinating white matter lesions (WMLs) of human MS brains, here we probed the impact of impaired microRNA (miRNA)-mediated post-transcriptional gene regulation in a mouse model lacking microglia/macrophage-specific Dicer1 expression during demyelination and remyelination. Conditional Dicer1 ablation and loss of miRNAs expression in adult microglia led to extensive demyelination and impaired myelin processing. Interestingly, demyelination was accompanied by increased apoptosis of mature oligodendrocytes (OLs) and arresting oligodendrocyte progenitor cells (OPCs) in the precursor stage. At the transcriptional level, Dicer1-deficient microglia led to downregulation of microglial homeostatic genes, increased cell proliferation, and a shift towards a disease-associated phenotype. Loss of remyelination efficiency in these mice was accompanied by stalling of OPCs in the precursor stage. Collectively, finding from this study highlight a new role of microglial miRNAs in promoting a pro-regenerative phenotype in addition to promoting OPC maturation and differentiation during demyelination and remyelination.

Transcriptomic analysis unveils bona fide molecular signatures of microglia under conditions of homeostasis and viral encephalitis

Project description:Microglia provide a front-line defense against neuroinvasive viral infection, however, determination of their bona fide transcriptional profiles under conditions of health and disease is challenging. Here, we used a combination of experimental tools to delineate the overall transcriptional landscape of microglia during viral infection. By exploiting the ribosomal tagging approach, we developed the concept of enrichment of relevant marker genes by comparing immunoprecipitated RNA with total RNA. Enriched transcripts corresponding to genes expressed in target cells were instrumental in defining bona fide signatures of microglia. With this approach, we generated a comprehensive and accurate transcriptome of microglia at the in situ environment under conditions of health and virus infection. These unified microglial signatures may serve as a benchmark to retrospectively assess ex vivo artefacts from available atlases. Leveraging the microglial translatome, we found enrichment of genes implicated in T-cell activation and cytokine production during the course of VSV infection. These data linked microglia with T-cell re-stimulation and further underscored that microglia shape anti-viral T-cell responses in the brain. Collectively, this study faithfully defines the transcriptional landscape of microglia in steady state and during viral encephalitis and highlights cellular interactions between microglia and T cells that contribute to the control of virus dissemination.
