Project description:Polyethylene terephthalate (PET) is one of the most commonly used plastics, utilized in synthetic fibers, water containers, and food packaging. From the 1990s onwards, the demand for PET, and therefore its production, increased exponentially. This increased usage of PET has resulted in a staggering accumulation of undegraded plastic waste. Nearly 80% of the 6300 million tons of plastic waste that had been generated as of 2015 were accumulated in landfills or the natural environment. Moreover, the production of PET relies heavily on non‐renewable fossil fuels, exacerbating environmental concerns over its widespread use. In alignment with principles of environmental sustainability, the biotechnological upcycling of PET has recently emerged as a compelling solution. Since the discovery of PETase, a hydrolase capable of depolymerizing this polyester, enzymatic plastic breakdown is increasingly considered as a promising solution for managing PET waste. This enzyme and its improved derivatives, such as Fast‐PETase, enable the breakdown of PET into bis(2‐41 hydroxyethyl) terephthalate (BHET) and mono(2‐hydroxyethyl) terephthalate (MHET). Subsequently, the enzyme MHETase is responsible for further degradation of MHET into ethylene glycol and terephthalic acid. The metabolic capability of microorganisms to utilize these monomers of PET for growth has been explored in various biotechnological applications, especially in the context of bioremediation and bioconversion processes aimed at transforming plastic waste into useful products using genetically engineered bacteria.

Project description:Plastic is widely used in daily life, but it also brings serious environmental pollution. Plastic products can degrade into microplastic particles after physical, chemical, and biological processes, and microplastic particles can further degrade into nanoplastic particles. Microplastics and nanoplastic particles exist in water, atmosphere, and food, and enter the human body through the digestive and respiratory tracts, causing harm to animal and human health. In this study, Caenorhabditis elegans was exposed to a culture medium containing polystyrene plastic particles at a concentration of 1000 μ g/L to detect the healthy lifespan of the nematodes, further revealing the impact of polystyrene nanoparticles on human health and providing a theoretical basis for further evaluating the biological toxicity effects and potential health risks of polystyrene nanoparticles.

Project description:Somatic cells can be chemically reprogrammed into a pluripotent stem cell (CiPSC) state, mediated by an extraembryonic endoderm- (XEN-) like state. We found that the chemical cocktail applied in CiPSC generation initially activated a plastic state in mouse fibroblasts before transitioning into XEN-like cells. The plastic state was characterized by broadly activated expression of development-associated transcription factors (TFs), such as Sox17, Ascl1, Tbx3, and Nkx6-1, with a more accessible chromatin state indicating an enhanced capability of cell fate conversion. Intriguingly, introducing such a plastic state remarkably improved the efficiency of chemical reprogramming from fibroblasts to functional neuron-like cells with electrophysiological activity or beating skeletal muscles. Furthermore, the generation of chemically induced neuron-like cells or skeletal muscles from mouse fibroblasts was independent of the intermediate XEN-like state or the pluripotency state. In summary, our findings revealed a plastic chemically activated multi-lineage priming (CaMP) state at the onset of chemical reprogramming. This state enhanced the cells’ potential to adapt to other cell fates. It provides a general approach to empowering chemical reprogramming methods to obtain functional cell types bypassing inducing pluripotent stem cells.

Project description:Humans are exposed to chemicals leaching from plastics, yet many remain data-poor and lack toxicological evaluation. High-throughput transcriptomics (HTTr) offers a scalable approach to screen chemicals of concern and derive mechanistic insights for human health risk assessment. We applied HTTr in MCF-7 breast cancer cells to evaluate plastic additives and dyes effects. Transcriptomic points of departure (tPODs) were derived from global gene perturbations, pathway-specific, and estrogen receptor α (ERα)–specific measures of gene expression. Transcriptomic biomarkers were also used to assess ERα activity and stress responses. Most plastic additives showed similar toxicological potencies, with the majority producing tPODs within one order of magnitude. Bisphenol K (BPK) was the most potent chemical tested, activating ERα at the lowest concentrations, followed by plastic additive 08 (PA08), and bisphenol A (BPA). Despite similar potencies, transcriptomic biomarkers revealed mechanistic differences: plastic additives that shared similar chemical features as BPA activated the ERα, whereas several others with different functional groups inhibited the biomarker. Pathway and upstream regulator analyses further showed that BPA-like plastic additives perturbed ERα–related pathways, while other plastic additives enriched fewer and often of opposing directionality gene sets. At the highest concentrations tested, many plastic additives also activated stress biomarkers and inhibited proliferation. These findings suggest that while many plastic additives display broadly similar in vitro potencies, they diverge in ERα regulation and downstream pathways. This study demonstrates the reproducibility and utility of HTTr for chemical screening, supports grouping of ERα-active plastic additives for read-across, and highlights the need for further screening of plastic additives.
