The effect of ionomycin-induced oocyte activation on multiple morphological abnormalities of the sperm flagella

Project description:Artificial oocyte activation (AOA) is considered an effective method to improve clinical outcomes in patients with some forms of male factor infertility and does not increase the risk of birth defects. However, the effects of AOA on patients with multiple morphological abnormalities of the sperm flagella (MMAF) caused by a DNAH1 mutation are still unknown. To explore the effects, our study analyzed a case with MMAF due to DNAH1 homozygous mutation who underwent testicular sperm extraction (TESE) combined with intracytoplasmic sperm injection (ICSI). The case had 28 MII oocytes. The 28 oocytes were divided randomly and equally into AOA and non-AOA groups. Ionomycin was used for AOA. We compared the clinical outcomes of two groups and selected three blastulation failure embryos from each group for transcriptome analysis. Differentially expressed genes (DEGs) were determined with an adjusted P-value < 0.05 and a |log2-fold change| ≥ 1. The comparison of clinical outcomes showed that the two pronuclei (2PN) rate and grade 1–2 embryo rate at day 3 were not significantly different between the two groups. Transcriptome analyses of blastulation failure embryos showed that the use of AOA had potential risks of chromosome structure defects, transcriptional regulation defects, and epigenetic defects. In conclusion, when the case with MMAF due to DNAH1 mutation underwent TESE-ICSI, ionomycin-induced oocyte activation could not improve the clinical outcomes and introduced the risks of chromosome structure defect, transcriptional regulation defect, and epigenetic defect.

Profiling of circulating microRNAs by RT-PCR arrays in atypical hemolytic uremic syndrome and controls samples

Project description:In this experiment, we explore if circulating microRNAs are altered in patients with atypical hemolytic uremic syndrome (aHUS). To do so, we analyze 4 different pooled samples of aHUS (Pool 1-4) in the arrays and 2 pools of age and gender-matched control samples (Pool F and Pool M). aHUS pools consist of patients samples with different characteristics regarding the presence of complement gene mutations: Pool 1: patients without identified mutations (n=5) Pool 2: patients with mutations in CFH (n=5) Pool 3: patients with mutations in MCP (n=5) Pool 4: patients with mutations in C3 or CFB (n=5) In the case of the control samples, pool F consists of 10 samples of healthy women and pool M consists of 10 samples of healthy men. In conclusion, circulating miRNAs levels are altered in aHUS compared with controls. This alteration is variable and could be mediated by the presence of the complement genetic defect.

Comparison of transcriptome landscape between adenine-induced and nephrectomy-induced uremic cardiomyopathy

Project description:Adenine and nephrectomy can be used to establish models of uremic cardiomyopathy in rodents. Although these two approaches cause similar phenotypes of heart, adenine may directly act on cardiomyocytes and lead to different molecular changes from nephrectomy-induced uremic cardiomyopathy. Transcriptome analysis on left ventricles showed 789 upregulated and 345 downregulated genes in mice with nephrectomy-induced uremic cardiomyopathy compared to control mice, while 432 upregulated and 155 downregulated genes in mice with adenine diet-induced uremic cardiomyopathy compared to control mice. Functional analysis revealed that these DEGs were involved in pathways related to immune inflammation, metabolic responses, and synaptic transmission. Further analysis demonstrated that nephrectomy-induced uremic cardiomyopathy has enhanced cell cycle process/extracellular matrix remodeling and suppressed fatty acid metabolism, whereas adenine diet-induced uremic cardiomyopathy only exhibits increased inflammatory response/extracellular matrix remodeling. Compared with nephrectomy-induced uremic cardiomyopathy, adenine diet-induced uremic cardiomyopathy showed improved oxidative phosphorylation, aerobic electron transport chain, and tricarboxylic acid cycle, and suppressed cell cycle-related process, indicating that adenine and its derivates may affect the process of chronic kidney induced cardiac changes.

Project description:To investigate the differential dynamic plasma miRNA profiles during the peri-implantation period in patients with different reproductive outcomes following embryo transfer. 10 negative pregnancy (NP) patients, 10 biochemical pregnancy loss (BPL) patients, and 10 clinical pregnancy (CP) patients were enrolled for miRNA-seq in this study. Peripheral blood samples were collected at three time-points (ET0, ET11, and ET14) from BPL patients and CP patients, and samples were collected at two time-points (ET0 and ET11) from NP patients. Small RNA libraries were constructed using peripheral plasma RNA. miRNA-seq was used to investigate the differential dynamic peripheral miRNA changes between the three groups during the peri-implantation period.

Project description:Temozolomide resistance is a major obstacle in the therapy of glioblastoma. This is also due to the insufficient understanding of glioma heterogeneity in relation to temozolomide resistance. More in depth biological knowledge would benefit the development of different approaches to tackle the problem from its root. We explored the different outcomes derived from the long-term exposure to temozolomide and identified two different behavioral phenotypes associated to specific categories of pathways. In addition, we investigated the intra-tumoral heterogeneity of these phenotypes revealing the complexity of the temozolomide resistance phenomenon.
