Project description:Endometriosis (EM) is a well-recognized disease that causes infertility in women of reproductive age. Tubal EM adversely affects fallopian tube function and female reproductive outcomes. Our study aimed to evaluate the effect of tubal EM on tubal epithelial ultrastructure and to explore the possible mechanisms of tubal infertility by analyzing the differential expression of exosomal microRNAs (miRNAs) in tubal fluid. We obtained human fallopian tube epithelium and tubal fluid samples from patients with and without tubal EM. We used scanning electron microscopy (SEM) and transmission electron microscopy (TEM) to assess ultrastructural changes. Exosomal miRNAs in tubal fluid were extracted for microarray. We visualized epithelial damage in the tubal EM group using electron microscopy. The number of organelles decreased, and organelle structure was destroyed. A total of 14 differentially expressed exosomal miRNAs were detected in tubal fluid (fold change >2 and P value <0.05). Four miRNAs (miR-1273f, miR-5699-5p, miR-6087, miR-6747-5p) were validated by quantitative real-time polymerase chain reaction (qRT-PCR). Bioinformatic analysis showed that most of the target genes participated in embryo transport, regulation of cell communication, anatomical structure morphogenesis, and immune system processes. We verified that tubal EM damaged the tubal epithelial ultrastructure in human specimens and identified differentially expressed exosomal miRNAs in tubal liquid. These findings may elucidate the pathogenesis of tubal EM-associated infertility.

Project description:Endometriosis is a gynaecological disease facing significant diagnostic challenges, contributing to an extensive diagnostic delay. Non-invasive diagnostic biomarkers of endometriosis could expedite diagnosis and minimise the reliance on surgery to confirm endometriosis. This pilot study aimed to identify candidate protein biomarkers of endometriosis from cervicovaginal fluid i) collected from consenting people having surgery for suspected endometriosis and ii) sampled via low vaginal swab. Samples from people with (n=20) and without (n=19) endometriosis across were analysed by data independent acquisition mass spectrometry using the Sequential Window Acquisition of All Theoretical Mass Spectra (SWATH)-MS approach. Samples were measured and analysed in two experimental runs (E1 and E2) based on time periods of sample collection. ELISA was performed to validate SWATH-MS findings.

Project description:<p><strong>BACKGROUND:</strong> Endometriosis is a common benign gynaecological disorder. A number of studies have confirmed that the occurrence of disease in endometriosis patients is associated with metabolic changes in biological fluids and tissues. We set out to investigate the metabolic characteristics of follicular fluid in patients with ovarian endometriosis undergoing in vitro fertilization (IVF).</p><p><strong>METHODS:</strong> The current research project is an exploratory cohort study on endometriosis. A total of 19 infertile patients with ovarian endometriosis diagnosed by laparoscopy were enrolled in this study and 23 age- and BMI-matched controls (women with infertility due to male or tubal factors). All the patients underwent IVF treatment with a gonadotropin-releasing hormone antagonist protocol and completed follicular fluid (FF) collection at oocyte retrieval. The metabolomics of FF samples was analyzed by ultra high performance liquid chromatography-orbitrap exploris-mass spectrometer (UHPLC-OE-MS). The best combination of biomarkers was then selected by performing stepwise logistic regression analysis with backward elimination. </p><p><strong>RESULTS:</strong> Fifteen metabolites were identified as biomarkers associated with endometriosis. A final model containing 8-hydroxy-2-deoxyguanosine, biotin, n-acetyl-L-methionine and n-methylnicotinamide was constructed. Receiver operating characteristic analysis confirmed the value of these parameters in diagnosing endometriosis, with a sensitivity of 94.7% and a specificity of 95.7%. Analysis of fortification via the Kyoto Encyclopedia of Genes and Genome showed that 15 metabolites were enriched in 8 metabolic pathways.</p><p><strong>CONCLUSION:</strong> Metabolomics based on UHPLC-OE-MS effectively characterized the metabolomics analysis of FF in patients with ovarian endometriosis. These findings may provide a new basis for a better understanding of how diseases progress and for the discovery of new biomarkers. </p>

Project description:Background Reduced endometrial receptivity is a major factor for impaired fertility in endometriosis (EMS). Endometrial deposition of collagen I proteins may account for poor endometrial receptivity in endometriosis. Methods We investigated the expression of collagen I expression in endometrium of endometriosis patients and in constructed EMS mice model. Effect of collagen I on ESCs was evaluated. Co-culture of exosomes with ESCs was conducted to evaluate the uptake of exosomes by different endometrial cell lines and the effect on decidualization of eutopic ESCs and embryo implantation. MiRNA expression profiles were compared between ectopic ESC derived exosomes and eutopic ESC derived exosomes. Luciferase reporter and its mutant plasmids were applied to confirm the direct target of miR-25-3p. Findings Here we found endometrial collagen I deposition with impaired decidualization in endometriosis patients and EMS mice model. Treatment of collagen I with ESCs contributed to impaired decidualization and inhibited BLS expansion in vitro. Endometriotic stromal cell-derived exosomes were detected in eutopic endometrium and the ectopic endometrial stromal cell derived exosomes were more taken-up by the same cell line in eutopic endometrium. Treatment of endometriotic stromal cell-derived exosomes increased the expression of endometrial collagen I in vitro and in vivo, while inhibited BLS expansion. Exosomal miR-25-3p was significantly increased in endometriotic stromal cell-derived exosomes compared with control group, and PTEN is a certain target of miR-25-3p. The promotion of endometrial miR-25-3p significantly increased collagen I expression in vitro through the PTEN/Akt pathway. Interpretation These results suggested that endometriotic stromal cell-derived exosomal miR-25-3p played key role in inducing endometrial collagen I deposition to impair embryo expansion in endometriosis via PTEN/Akt pathway. Project description:Endometriosis is an inflammatory chronic gynecological condition that affects 10-15% of reproductive-age women which causes pelvic pain and infertility. The disease is characterized by the growth of endometrial tissue outside the uterine cavity. Currently, early diagnosis of endometriosis is difficult due to non-specific symptoms and the absence of reliable biomarkers. Understanding these early molecular alterations could support the development of diagnostic tools for timely detection and treatment. The objective of this study was to identify changes in circulating microRNA (miRNA) profiles during the initial stages of endometriosis using a longitudinal non-human primate baboon model. Endometriosis was induced in female baboons by laparoscopic inoculation of menstrual endometrial tissue and plasma was collected at various timepoints. Plasma miRNAs at 3, 6, 9, and 15 months were analyzed using RNA sequencing. A total of 2,731 miRNAs were detected, 76 of which showed significant differences at least at one time point. There are four, three, and one miRNA exclusively differentially expressed at 3-, 6- and 9-month timepoints respectively while several (68) miRNAs are significantly expressed at 15 months. Four miRNAs (miR-210-3p, miR-448, miR-1260a/1260b, and miR-1298-5p) exhibited significant alterations across all time points. Of these, miR-210-3p increased, while the others decreased significantly. Hierarchical clustering revealed separation between the early (pre-inoculation and three months) and late (nine and 15 months) stages, indicating progressive divergence of miRNA profiles. These findings identify candidate miRNAs that may provide insight into early pathophysiological mechanisms and represent candidate circulating biomarkers associated with temporal molecular changes following disease induction, warranting further validation.
