Project description:The proper control of mitosis depends on the ubiquitin-mediated degradation of the right mitotic regulator at the right time. This is under the control of the anaphase promoting complex/cyclosome (APC/C) ubiquitin ligase that is regulated by the Spindle Assembly Checkpoint (SAC). The Checkpoint prevents the APC/C from recognizing Cyclin B1, the essential anaphase and cytokinesis inhibitor, until all chromosomes are attached to the spindle. Once chromosomes are attached, Cyclin B1 is rapidly degraded to enable chromosome segregation and cytokinesis. We have a good understanding of how the SAC inhibits the APC/C, but relatively little is known about how the APC/C recognises Cyclin B1 as soon as the SAC is turned off. Here, by combining live cell imaging, in vitro reconstitution, biochemistry, and structural analysis by cryo-electron microscopy, we provide evidence that the rapid recognition of Cyclin B1 in metaphase requires spatial regulation of the APC/C. Using fluorescence cross correlation spectroscopy, we find that Cyclin B1 and the APC/C primarily interact at the mitotic apparatus. We further show that this is because Cyclin B1, like the APC/C, binds to nucleosomes, and identify an ‘arginine-anchor’ in the N-terminus as necessary and sufficient for binding to the nucleosome. Mutating the nucleosome binding motif on Cyclin B1 reduces its interaction with APC/C and delays its degradation, and cells with the mutant, non-nucleosome-binding Cyclin B1 become aneuploid, demonstrating the physiological relevance of our findings. Together, our data demonstrate that mitotic chromosomes constitute a platform to promote the efficient interaction between Cyclin B1 and APC/C and ensure the timely degradation of Cyclin B1 and genomic stability.

Project description:Phosphorylation of substrates by cyclin-dependent kinases (CDKs) is the driving force of cell cycle progression. Several CDK-activating cyclins are involved, yet how they contribute to substrate specificity is still poorly understood. Here, we discovered that a positively charged pocket in cyclin B1, which is exclusively conserved within B-type cyclins and binds phosphorylated serine- or threonine-residues as well as acidic side chains, is essential for correct execution of mitosis. HeLa cells expressing pocket mutant cyclin B1 are strongly delayed in anaphase onset due to multiple defects in mitotic spindle function and timely activation of the E3-ligase APC/C. Non-functional pocket results in decreased APC/C phosphorylation at non-consensus CDK1 sites, compromised in vitro ubiquitylation activity, and reduced binding of the E2-enzyme UBE2S. Our results support a model in which cyclin B1’s pocket serves as specificity factor for sequential substrate phosphorylations involving initial priming events facilitating subsequent pocket-dependent phosphorylations at non-consensus CDK1 motifs.

Project description:Mammalian cell cycle is driven by several CDK-cyclin complexes, with CDK1/cyclin B1 (CCNB1) being the one that is essential for mitotic entry and progression. CDK5 is not considered a typical CDK due to its high expression in post-mitotic neurons and activation by non-cyclin proteins p35 and p39. We previously showed that CDK5 is active in mitosis and here we demonstrate that CCNB1 is the mitotic activator of CDK5. Using independent chemical genetic approaches to specifically abrogate CDK5 at the onset of and throughout mitosis, we observe mitotic defects, nuclear atypia, and significant alterations in the mitotic phospho-proteome. Importantly, the disruption of the CDK5-CCNB1 complex phenocopies CDK5 abrogation. Together, our results demonstrate that CCNB1 partners with both CDK5 and CDK1 to drive mitosis, defines CDK5-CCNB1 as a canonical cell cycle CDK/cyclin complex and describes the role of CDK5-CCNB1 complex in ensuring mitotic fidelity.

Project description:Mitosis in early embryos often proceeds at a rapid pace, but how this pace is achieved is not understood. Here we show that cyclin B3 is the dominant driver of rapid embryonic mitoses in the C. elegans embryo. Cyclins B1 and B2 support slow mitosis (NEBD to Anaphase ~600s) but the presence of cyclin B3 dominantly drives the ~3-fold faster mitosis observed in wildtype. Multiple mitotic events are slowed down in Cyclin B1&B2-driven mitosis and cyclin B3-associated Cdk1 H1 kinase activity is ~25-fold more active than cyclin B1-associated Cdk1. Addition of cyclin B1 to fast cyclin B3-only mitosis introduces an ~60s delay between completion of chromosome alignment and anaphase onset; this delay, which is important for segregation fidelity, is dependent on inhibitory phosphorylation of the anaphase activator Cdc20. Thus, cyclin B3 dominance, coupled to a cyclin B1-dependent delay that acts via Cdc20 phosphorylation, sets the rapid pace and ensures mitotic fidelity in the early C. elegans embryo.

Project description:Metabolic dysfunction-associated steatotic liver disease (MASLD) is increasingly prevalent with life-threatening complications, underscoring the need for new therapeutic targets. In this study, we identified a novel long non-coding RNA (lncRNA), WEE1-AS, which is transcribed from the antisense strand of the Wee1 gene locus. Hepatic WEE1-AS was expressed in a spatiotemporal manner in response to a high-fat diet challenge. Functionally, WEE1-AS enhanced mitochondrial fatty acid oxidation by activating the Cdk1/Cyclin B1 complex through two mechanisms. First, it suppressed the transcription of the Wee1 gene by preventing access to the transcriptional machinery. Second, WEE1-AS bound and stabilized the Cyclin B1 protein by suppressing ubiquitin/proteasome-mediated degradation. Notably, treatment with the Wee1 inhibitor Adavosertib ameliorated MASLD symptoms by improving mitochondrial function in the liver. Furthermore, we identified a human homolog, lnc106435.1, which improved mitochondrial function, suggesting that targeting lnc106435.1 could be a valuable therapeutic strategy for managing MASLD.

Project description:A major obstacle in diabetes is the metabolic or hyperglycemic memory, which lacks specific therapies. Here we show that glucose-mediated changes in gene expression largely persist in diabetic kidney disease (DKD) despite reversing hyperglycemia. The senescence-associated cyclin-dependent kinase inhibitor p21 (Cdkn1a) was the top hit among genes persistently induced by hyperglycemia and was associated with induction of the p53-p21 pathway. Persistent p21 induction was confirmed in various animal models, human samples and in vitro models. Tubular and urinary p21-levels were associated with DKD severity and remained elevated despite improved blood glucose levels in humans. Mechanistically, glucose-induced and sustained tubular p21 expression is linked to demethylation of its promoter and reduced DNMT1 expression. Exploiting signaling of the disease resolving protease activated protein C (3K3A-aPC, parmodulin-2) reversed sustained tubular p21 expression, tubular senescence, and DKD. Thus, p21-dependent tubular senescence is a pathway contributing to the hyperglycemic memory, which can be therapeutically targeted.

Project description:To further explore differentially expressed genes in high glucose-induced renal tubular fibrosis cells, we used whole transcriptome microarray expression profiling as a discovery platform to identify genes with a potential role in the high glucose-induced renal fibrosis process. Low-glucose stimulation (5.5 mM glucose) and high-glucose stimulation (25 mM glucose) of human renal tubular HK2 cells identified a consensus profile of 644 highly expressed genes to distinguish high-glucose fibrotic renal tubules from control samples.
