Project description:Phospholipase C-gamma 2 (PLCγ2) is highly expressed in hematopoietic and immune cells, where it is a key signalling node enabling diverse cellular functions. Within the periphery, gain-of-function (GOF) PLCγ2 variants, such as the strongly hypermorphic S707Y, cause severe immune dysregulation. The milder hypermorphic mutation PLCγ2 P522R increases longevity and confers protection in central nervous system (CNS) neurodegenerative disorders, implicating PLCγ2 as a novel therapeutic target for treating these CNS indications. Currently, nothing is known about what consequences strong PLCγ2 GOF has on CNS functionality, and more precisely on the specific biological functions of microglia. Using the PLCγ2 S707Y variant as a model of chronic activation we investigated the functional consequences of strong PLCγ2 GOF on human microglia. PLCγ2 S707Y expressing human inducible pluripotent stem cells (hiPSC)-derived microglia exhibited hypermorphic enzymatic activity under both basal and stimulated conditions, compared to PLCγ2 wild type. Despite the increase in PLCγ2 enzymatic activity, the PLCγ2 S707Y hiPSC-derived microglia display diminished functionality for key microglial processes including phagocytosis and cytokine secretion upon inflammatory challenge. RNA sequencing revealed a downregulation of genes related to innate immunity and response, providing molecular support for the phenotype observed. Our data suggests that chronic activation of PLCγ2 elicits a detrimental phenotype that is contributing to unfavourable CNS functions, and informs on the therapeutic window for targeting PLCγ2 in the CNS. Drug candidates targeting PLCγ2 will need to precisely mimic the effects of the PLCγ2 P522R variant on microglial function, but not those of the PLCγ2 S707Y variant.

Project description:Microglia are the resident immune cells of the CNS and integral to maintaining homeostasis. They rapidly respond to injury or infection within the CNS by altering their morphology, proliferating, and releasing cytokines and other signaling molecules. Microglia also play critical roles in development, regulating synaptic pruning and debris clearance. In disease, they are powerful mediators of neuroinflammation. Understanding the molecular pathways involved in microglial function is pivotal for advancing neurobiological research and developing effective strategies for CNS disorders. In this context, P2RY12 is a G protein-coupled receptor (GPCR) that is enriched in microglia in the brain parenchyma and is part of the transcriptional signature of homeostatic, ramified microglia. However, P2RY12 is downregulated in activated microglia and in neurological conditions. The downstream effects of P2RY12 downregulation in disease-associated microglia and how they influence microglial activation remain inadequately understood. In this study, we apply transcriptional and histological methods to explore the changes to microglia upon a genetic P2RY12 loss. Our findings reveal that P2RY12-deficient microglia experience alterations in distinct metabolic pathways while preserving overall homeostatic microglial transcriptional identity. P2RY12 deficiency dysregulates signature genes involved in homeostatic iron metabolism. Importantly, the Glutathione Peroxidase 4 (GPX4)-Glutathione (GSH) antioxidant pathway related to ferroptosis susceptibility is impaired upon microglial activation with LPS treatment. These results highlight the critical role of P2RY12 in regulating microglial immune and metabolic responses under both homeostatic and inflammatory conditions, providing insights into its involvement in CNS pathophysiology.

Within the CNS, we identify ARID5A as a microglial-specific RBP and integrate multi-omics analyses of ARID5A's RNA, DNA, and protein interactions to uncover its RNA-mediated regulation of microglial functions. We show ARID5A regulates the splicing and translation of its RNA targets, many of which are integral to microglial functions. We find ARID5A modulates cytokine secretion, lysosome activity, and iron accumulation, as well as ferroptosis sensitivity in microglia and co-cultured neurons. We illustrate the neuroprotective potential of ARID5A modulation, in which knockdown restored dysregulated functions in TREM2 mutant microglia. Our results highlight the potential of leveraging ARID5A-RNA interactions to restore dysregulated pathways in neurodegeneration.

Project description:Microglia maintain brain homeostasis through coordinated pathways, including cell migration, phagocytosis, and secretion of immune-related signaling factors. Microglial reactivity is associated with a variety of functional outcomes that are highly context-dependent with epigenetic regulation through DNA methylation and histone modificationsimplicated in this complex control of microglial plasticity. Specifically, in relation to histone methylation, lysine-specific demethylase 5B (KDM5B) has been associated with several pathophysiological states, including neurodevelopmental and inflammatory disorders, cancer, and alcohol use disorder; however, the cell-type-specific role of KDM5B in microglial reactivity has not been investigated. In this study, transcriptomic and proteomic analyses were used to characterize the effects of KDM5B depletion on microglial pathways in clustered regularly interspaced short palindromic repeats (CRISPR)-edited adult-derived murine microglial cells. Additionally, various immunomodulatory stimuli, including lipopolysaccharide, interleukin-4, and alcohol, were used to study the effects of KDM5B depletion on immune reactivity in microglia using deep proteomics and bioinformatic analysis. Through this comprehensive characterization, KDM5B-depleted microglia exhibited broad remodeling of immune reactivity, including reduced intracellular and secreted inflammatory responses to lipopolysaccharide as well as distinct modulation of alcohol- and interleukin-4-induced pathways, that was functionally demonstrated by altered cytokine secretion profiles. Results from this study provide critical insight into KDM5B-mediated immunological effects on microglial function.

Project description:Cannabinoids are known to exert immunosuppressive activities. However, the mechanisms which contribute to these effects are unknown. Using lipopolysaccharide (LPS) to activate BV-2 microglial cells, we examined how Δ9-tetrahydrocannabinol (THC), the major psychoactive component of marijuana, and cannabidiol (CBD) the non-psychoactive component, modulate the inflammatory response. Microarray analysis of genome-wide mRNA levels was performed using Illumina platform and the resulting expression patterns analyzed using the Ingenuity Pathway Analysis to identify functional subsets of genes, and the Ingenuity System Database to denote the gene networks regulated by CBD and THC.

Project description:Microglial cells are resident macrophages in the central nervous system (CNS) and play a pivotal role in the innate and adaptive immune responses against microbial infections. The immune functions of microglia are regulated by a milieu of cytokines including interferon (IFN)-gamma. We here performed a series of experiments to acertain the transcriptional profile of human fetal microglial cells at 1, 6, and 24 h after IFN-gamma treatment. Primary human microglial cells were either untreated or treated with 200u/ml IFN-gamma. Affymetrix U133A chips were utilized. Four different tissue samples (B18, O, W, and Y20) were analyzed at the three time points.
