Project description:Urea can serve as nitrogen source for coral holobionts and plays a cruscial role in coral calcification, although the degradation of urea by coral symbionts is not fully understood. In this study, we investigated the urea utilized pathway and the responses of the Symbiodiniaceae family to urea under high temperature conditions. Genome screening revealed that all Symbiodiniaceae species contain the urease (URE) and DUR2 subunit of urea amidolyase (UAD) system. However, only three speciesCladocopium goreaui, Cladopium c92, and Symbiodinium pilosum possess a complete UAD system, including both DUR1 and DUR2. Phylogentic analyses revealed that the UAD system in Symbiodiniaceae clusters more closely with symbiotic bacteria, indicating that horizontal gene transfer of UAD system has occured in coral symbionts. Physiology analysis showed that the symbiodiniacean species Cladocopium goreaui, which containing both URE and UAD, grew better under urea than ammonium conditions, as indicated by higher maximum specific growth rates. Furthermore, most genes of Symbiodiniaceae involved in urea utilization appeared to be stable under various conditions such as heat stress (HS), low light density, and nitrogen deficiency, wheras in ammonium and nitrate transporters were significantly regulated. These relatively stable molecular regulatory properties support sustained urea absorption by Symbiodiniaceae, as evidenced by an increase in δ15N2-urea absorption and the decreases in δ5N-NO3-, and δ15N-NH4+ from cultural environment to Symbiodiniaceae under HS conditions. Token together, this study reveals two distinct urea utilization systems in coral ecosystem and highlights the importance of the urea cycle in coral symbionts when facing fluctuating nitrogen environment in future warming ocean.

Project description:The metabolic bases of the interaction between the coral Acropora millepora and its dinoflagellate symbiont were investigated by comparing gene expression levels under light and dark conditions at the whole transcriptome level. Among the differentially expressed genes identified, a suite of genes involved in cholesterol transport was found to be up-regulated under light conditions, confirming the significance of this compound in the coral symbiosis. Although ion transporters likely to have roles in calcification were not differentially expressed in this study, expression levels of many genes associated with skeletal organic matrix composition and organization were higher in light conditions. This implies that the rate of organic matrix synthesis is one factor limiting calcification at night. Thus, LEC during the day is likely to be a consequence of increases in both matrix synthesis and the supply of precursor molecules as a result of photosynthetic activity.

Project description:The association with photosymbiotic algae is crucial for the proliferation of many coral reef organisms, but increases their sensitivity to environmental changes. Large benthic foraminifera (LBF) are a diverse group of carbonate producers harboring algal photosymbionts. They act as key ecological engineers and are widely used as bioindicators. As in corals, elevated temperatures and light intensities are known to induce bleaching in LBF, but the combined effects of ocean acidification and warming remain unclear. To shed light into the adaptive physiology of LBF, we linked the assessment of the holobiont and photosymbiont physiological condition (mortality, growth, coloration, and chlorophyll a) to a bottom-up proteomics approach that allows the examination of cellular responses of host and symbionts simultaneously. In a two-months experiment, we exposed Amphistegina lobifera to the combined effects of ocean acidification (400, 1000 and 2800 ppm pCO2) and warming (28-control and 31°C). More than 1,000 proteins were identified by label-free mass spectrometry-based whole proteome analysis and assigned to the host or photosymbionts. Photopigment concentrations declined in response to elevated pCO2, visible by discoloration. These indicate the reduction of photosymbiont densities under ocean acidification, despite the fertilizing effects suggested for high inorganic carbon availability, and imply metabolic adjustments. Increases of proteolytic proteins suggest active host regulation of photosymbiont density in order to maintain homeostasis with its algal photosymbionts. Growth rates, however, were unaffected by elevated pCO2 levels at control temperatures, but high pCO2 levels (2800 ppm, pH 7.52) combined with thermal stress (31°C) impaired growth, though mortality and shell dissolution was negligible. While growth was unaffected by intermediate pCO2 levels (1000 ppm, pH 7.98) combined with ocean warming, this treatment induced the most distinct proteome responses. These include the regulation of ion transporters and host cytoplasmic proteins that likely abet calcification under ocean acidification. This study reveals a highly complex cellular response in both the host and the photosymbiont, which appears to facilitate a high resilience potential of A. lobifera to end of the century ocean conditions. Nevertheless, our results imply that when pCO2 levels rise above 1000 ppm during persistent ocean warming or extreme heating events these adaptive mechanisms become disrupted.

Project description:Abstract The coral–dinoflagellate symbiosis is increasingly disrupted by global and local anthropogenic stressors. Coral bleaching is primarily a result of high sea surface temperatures, while eutrophication is associated with reef ecosystem degradation. Excess inorganic nitrogen relative to phosphate has been proposed to directly sensitise corals to thermal bleaching and accelerate reef decline. We assessed the proteomic response of the dinoflagellate coral symbiont Symbiodinium microadriaticum to elevated temperatures under multiple nutrient conditions by mass spectrometry. Elevated temperatures resulted in reductions of many chloroplast proteins, particularly light-harvesting complexes, with simultaneous increases in chaperone proteins. N:P imbalance had a larger effect on the proteome than temperature, but the biological processes and proteins responding to each stressor largely overlapped. The proteomes were highly similar at low N:P ratios but were strongly affected by phosphate starvation. High N:P ratios inhibited cell division, reflected by changes in proteins involved in protein translation. Imbalanced N:P did not increase sensitivity to high temperatures as measured by physiological means; however, imbalanced N:P strongly upregulated cell redox homeostasis proteins at high temperatures. As redox balance is critical during thermal bleaching, these data provide insight into the mechanisms of cellular responses to thermal and multiple stresses in the coral–dinoflagellate symbiosis.
