Project description:In this study, depending on quantitative liposome and transcriptomic analyses, we unveiled safflower seed lipidic landscapes at 5 different developmental stages of seeds and revealed a parallel classification of 417 lipid compounds and their co-expressed genes into 7 distinct lipidic metabolites vs gene modules (MM vs GM). An essential transcriptional regulatory network for major lipid compounds changes that occurred throughout the seed growth cycle was constructed via integrative bioinformatics coupled with functional genomic assays. Sixteen transcription factors were identified that regulate the accumulation of triacylglycerols (TAGs) and fatty acids (FAs). Our findings thus shed light on the seeds of safflower metabolic regulatory network involved in the biosynthesis of major lipid compounds and provide a prospective resource for the designed improvement of oilseed quality.

Investigation of the microRNAs in safflower seed, leaf and petal by high-throughput sequencing

Project description:We report that there are at least 236 known miRNAs expressed in safflower, of which 100 miRNAs with relatively high expression level exhibited evolutionary conservation across multiple plants. Comparison of their expression patterns among different tissues shows that a total of 116, 133 and 128 miRNAs are significantly differentially expressed between safflower seed/leaf, seed/petal and leaf/petal. Meanwhile, interestingly, the majority of the most significantly differentially expressed miRNAs between tissues are tissue-specific miRNAs. In addition, 15 putative novel miRNAs have been identified in safflower. The small RNA transcriptomes obtained in this study provide a basis for further investigation of the physiological roles of identified miRNAs in safflower.

Transcription profiling by array of human A2780 ovarianc cancer cells and cisplatin resistant derivate cells treated with cisplatin or liposomal cisplatin

Project description:A2780 ovarian cancer cells and a cisplatin resistant derivate of A2780 cells, obtained from ECACC, UK, No. 93112519 [A2780] and No. 93112517 [A2780 cis] were seeded out in T-75 flasks at a density of 2x106 for A2780sens and 3x106 for A2780cis cells in 15 ml of medium and preincubated overnight. Medium was removed and 15 ml fresh medium (37M-0C) with different concentrations of cisplatin, liposomal cisplatin or empty liposomes were added and incubated for 72 h at 37M-0C in a 5% CO2 incubator. In case of A2780sens cells, 1.72 M-5M cisplatin (IC50 concentration) and in case of A2780cis cells 8.94 M-5M cisplatin (IC50 concentration) were added. Liposomal formulations contained equal cisplatin concentrations. Empty liposomes were added in the same concentration as the liposomal cisplatin, to analyze the impact of liposomal lipids (A2780sens: 0.80M-5mol lipid, A2780cis: 4.15 M-5mol lipid). After incubation, medium was removed and cells were washed thrice with 10 ml PBS. 1 ml RLT-buffer was added and cells lysates were stored at -80M-0C until RNA extraction.

Intraarticular injection of liposomal adenosine reduces cartilage damage in established murine and rat models of osteoarthritis

Project description:Osteoarthritis (OA) affects nearly 10% of the population of the United States and other industrialized countries and, at present, short of surgical joint replacement, there is no therapy available that can reverse the progression of the disease OA. Adenosine, acting at its A2A receptor (A2AR), is a critical autocrine factor for maintenance of cartilage homeostasis. Here we report that injection of liposomal suspensions of a selective A2AR agonist, CGS21680, significantly reduced OA cartilage damage in a rat model of established post-traumatic OA (PTOA). Differential expression analysis of mRNA from chondrocytes harvested from knees of rats with PTOA treated with liposomal A2AR agonist revealed downregulation of genes associated with matrix degradation and upregulation of genes associated with cell proliferation as compared to liposomes alone.

Global analysis of lipid metabolism regulation in Brassica napus developing seeds

Project description:Gene expression profiles during seed development and fatty acid (FA) metabolism, as well as the relevant regulation, of Brassica napus were studied through multiple high-throughput genomic approaches. Serial Analysis of Gene Expression (SAGE) using seed materials obtained a total of 68,718 tags, of which 23,897 were unique and 503 tags were functionally identified, and revealed the transcriptome of approximately 35,000 transcripts in B. napus developing seeds. Further, ~22,000 independent ESTs were obtained by large-scale sequencing using immature embryos at different stages, and 8343 uni-ESTs and 3355 full-length cDNAs were identified respectively, resulting in the systemic identification of B. napus FA biosynthesis-related genes. Gene expression profiles were further studied employing cDNA chip hybridization to reveal the global regulatory network of FA metabolism in developing seeds. Together with the analysis on FA amounts and composition, it was shown that 17-21 days after pollination (DAP) was a crucial stage for transition of seed to sink tissue. High expressions of FA biosynthesis-related genes and transition of FA components are mainly at stages 21 DAP or 21-25 DAP respectively. In addition, compared to Arabidopsis, more critical roles of starch metabolism are detected for B. napus seed FA metabolism and storage components accumulation. Crucial effects of starch metabolism, carbon flux, oxidative pentose phosphate pathway (OPPP), photosynthesis, and other regulators in FA metabolism were discussed.
