Lactobacillus acidophilus potentiates oncolytic virotherapy through modulating gut microbiota homeostasis in hepatocellular carcinoma

Project description:Oncolytic viruses (OVs) hold promise for cancer treatment. However, the antitumor efficacy is limited. Microbiota plays a pivotal role in cancer treatment and its impact on oncolytic virotherapy is unknown. Here, we show that VSVΔ51 has higher antitumor efficacy for hepatocellular carcinoma in the absence of microbiota in female mouse models. VSVΔ51 infection causes microbiota dysbiosis, increasing most of the gut bacteria abundance, while decreasing the commensal Lactobacillus. VSVΔ51 reduced intestinal expression of SLC20A1 that binds to Lactobacillus acidophilus (L. acidophilus) CdpA cell wall protein through IL6-JAK-STAT3 signaling, thereby attenuating attachment and colonization of L. acidophilus. L. acidophilus supplementation confers sensitivity to VSVΔ51 through restoring gut barrier integrity and microbiota homeostasis destroyed by VSVΔ51. In this work, we show that targeting microbiota homostasis holds substantial potential in improving therapeutic outcomes of oncolytic virotherapy

Project description:Oncolytic virotherapy is a unique and promising immunotherapy. In our prior study, a recombinant oncolytic vaccinia virus carrying IL-7 and IL-12 (hIL-7/mIL-12-VV) showed robust antitumor efficacy in both immunogenic CT26.WT and poorly immunogenic Lewis lung carcinoma (LLC) within the treated tumors. However, the efficacy was limited in virus non-treated (distant) LLC tumors. Here we identified oxaliplatin as a powerful combination partner for an oncolytic vaccinia virus to achieve strong efficacy in the virus-injected tumors and the distant tumors. To understand the molecular mechanism of enhanced antitumor efficacy, we evaluated RNA-sequencing signatures in LLC tumors treated with oxaliplatin alone, hIL-7/mIL-12-VV alone and their combination.

IDH1-R132H Enhances Oncolytic HSV-1 Therapy by Facilitating Viral Entry and Immune Activation in Glioma

Project description:Oncolytic virotherapy represents a promising yet under-explored approach for precision cancer treatment, particularly when tailored to tumor-specific molecular profiles. Patients with high-grade isocitrate dehydrogenase (IDH) mutant astrocytomas have limited treatment options and experience poor prognoses. Here, we investigate the therapeutic potential of rQNestin34.5v.2 (CAN-3110), an engineered oncolytic herpes simplex virus 1 (oHSV-1), in the context of IDH1-R132H-mutant high-grade diffuse gliomas. We demonstrate that the IDH1-R132H mutation increases glioma susceptibility to viral infection through upregulation of Nectin 1, the main cellular entry receptor for HSV-1 found in gliomas. Concurrently, IDH1-R132H-driven DNA hypermethylation suppresses interferon (IFN) signaling, an essential antiviral pathway, contributing to a permissive tumor microenvironment that facilitates viral replication and increases tumor cell susceptibility to virus-induced apoptosis. In immunocompetent IDH1-R132H murine glioma models, intratumoral administration of rQNestin34.5v.2 elicits robust immune activation, characterized by increased immune cell infiltration into the tumor and systemic IFN-γ release. However, elevated expression of poliovirus receptor (PVR or CD155) and the immune checkpoint T-cell immunoreceptor with immunoglobulin and ITIM domain (TIGIT) on tumor-infiltrating lymphocytes suggested a potential resistance mechanism to virotherapy. Combining rQNestin34.5v.2 with TIGIT blockade enhanced therapeutic efficacy and improved survival outcomes compared to monotherapy. These data show that IDH1-R132H modulates viral entry and immune evasion, identifying it as a predictive biomarker for oncolytic virotherapy response.
