Project description:To further investigate the therapeutic potential of NBXH in treating tuberculosis, we employ whole genome microarray expression profiling as a discovery platform. The study aimed to explore the impact of NBXH treatment on the gene expression profile of tuberculosis-infected animal models. The animal models were treated with high, medium, and low doses of NBXH. Following three months of treatment, PBMCs from each treatment group were used for whole genome microarray expression profiling analysis, alongside a control group. With a screening criterion of P <0.05 and an absolute log2FC value >1, 3454 up-regulated and 3597 down-regulated DEGs were respectively identified in the gene expression profiles of the mouse TB model treated with NBXH.

Comparative label-free lipidomic analysis of Mycobacterium tuberculosis during dormancy and reactivation

Project description:Mycobacterium tuberculosis employs several strategies to combat and adapt to adverse conditions encountered inside the host. The non-replicative dormant state of the bacterium is linked to drug resistance and slower response to anti-tubercular therapy. It is known that alterations in lipid content allow dormant bacteria to acclimatize to cellular stress. Employing comparative lipidomic analysis we profiled the changes in lipid metabolism in M. tuberculosis using a modified Wayne's model of hypoxia-induced dormancy. Further we subjected the dormant bacteria to resuscitation, and analyzed their lipidomes until the lipid profile was similar to that of normoxially grown bacteria. An enhanced degradation of cell wall-associated and cytoplasmic lipids during dormancy, and their gradual restoration during reactivation, were clearly evident. This study throws light on distinct lipid metabolic patterns that M. tuberculosis undergoes to maintain its cellular energetics during dormancy and reactivation.

In this work we have analyzed by DNA microarray technique the global transcription profile of Mtb infecting primary human macrophages in order to identify putative bacterial pathogenic factors that can be relevant for the intracellular survival of Mtb. Keywords: time course We compared the global gene expression of the H37Rv strain of Mtb after 4 hours and 24 hours of infection of human macrophage-like THP-1 cells with the gene expression profile of the strain growing exponentially in broth cultures.

Proteomic Profiling of Mycobacterium tuberculosis in Response to ILE Treatment

Project description:This study aims to elucidate the global proteomic changes in Mycobacterium tuberculosis upon treatment with the novel compound ilamycin E(ILE), which targets the Clp protease system. Label-free quantitative proteomics was employed to assess differential protein expression after ILE exposure. The results reveal that ILE induces significant alterations in the expression of proteins involved in proteolysis, stress response, redox homeostasis, and energy metabolism, providing valuable insights into the adaptive response and potential mechanism of action of ILE in M. tuberculosis. These data support further development of ILE as a candidate anti-mycobacterial agent targeting intracellular proteostasis.
