Project description:This experiment was conducted in order to evaluate the potential contribution of oil droplets to the toxicity of dispersed oil to fish larvae. Atlantic cod larvae were exposed to five concentrations of either dispersed oil (D1-D5) (containing oil droplets [medium size 10-14 µm based on volume] and water soluble fraction [WSF]) or the filtered dispersion containing only WSF of oil (W1-W5) for four days and harvested for transcriptional analysis at 13 days post hatching. The most significant differently expressed genes were observed in cod larvae exposed to the highest concentration of the dispersed oil (containing 10.41 ± 0.46 µg ∑PAH/L), with CYP1A showing the strongest response. Functional analysis further showed that the top scored network as analyzed with Ingenuity Pathway Analysis was “Drug Metabolism, Endocrine System Development and Function, Lipid Metabolism”. Oil exposure also increased the expression of genes involved in bone resorption and decreased the expression of genes related to bone formation. In conclusion, oil exposure affects drug metabolism, endocrine regulation, cell differentiation and proliferation, apoptosis, fatty acid biosynthesis and tissue development in Atlantic cod larvae. The altered gene transcription was dominated by the WSF and the oil droplet fraction only had a moderate impact on the observed changes.

Project description:The use of dispersants can be an effective way to deal with acute oil spills to limit environmental damage, however very little is known about whether chemically dispersed oil have the same toxic effect on marine organisms as mechanically dispersed oil. We exposed Atlantic cod larvae to chemically and mechanically dispersed oil for four days during the first-feeding stage of development, and collected larvae at 14 days post hatch for transcriptional analysis. A genome-wide microarray was used to screen for effects and to assess whether molecular responses to chemically and mechanically dispersed oil were similar, given the same exposure to oil (droplet distribution and concentration) with and without the addition of a chemical dispersant (Dasic NS).

Project description:Polar cod (Boreogadus saida), a key arctic fish species spawning during the polar night, may be at risk for crude oil exposure during this potential sensitive life stage. This study investigates the effects of crude oil exposure on Polar cod during spawning season. Wild-caught polar cod were exposed to crude oil water accommodated fraction (WAF) (start concentrations of sum of 44 polycyclic aromatic hydrocarbon (sum 44 PAHs) at 12.5 μg/L) from pre-spawning to post-spawning. The exposure system consisted of an oiled-rock-column system where running though sweater delivers WAF in the fish tank at decreasing concentrations over time. Samples were taken at three time-points (pre-spawning, spawning window, and post-spawning) and used to evaluate changes in reproductive endpoints such as gonad histology, plasma steroid hormones and sperm motility. For RNA-extraction and sequencing, liver samples from both oil-exposed and control groups of female fish (n = 5 per group) were taken at 47 days after experiment start and submitted for sequencing. RNA-seq analysis showed that hundreds of genes were differentially expressed in the liver.

Project description:This study was performed to investigate the effect of camelina oil-based diets on the immune function of Atlantic cod, as measured by gene expression in spleen. Atlantic cod were fed with one of three practical diets (three tanks per diet): a control diet using herring oil as a lipid source (FO diet), and two experimental diets using vegetable oil from Camelina sativa to replace 40% or 80% of herring oil (40CO and 80CO diets). We studied both the effect of the diet alone on basal spleen gene expression levels, as well as the effect of the diet after fish were injected with the synthetic double-stranded RNA polyriboinosinic polyribocytidylic acid (pIC), which mimics a viral immune stimulus.

Project description:Polar cod, a key fish species in the arctic marine foodweb is vulnerable to effects of pollution from offshore petroleum related activities in the Arctic and sub-arctic region. The study was conducted to map transcriptome responses to in Polar cod (Boreogadus saida) liver slice culture exposed to benzo[a]pyrene (BaP) in the presence or absence of physiological levels of ethynylestradiol (EE2). BaP is a polycyclic aromatic hydrocarbon (PAH), also found in crude oil contaminants. PAHs such as BaP are among the most toxic compounds of crude oil. Precision-cut liver slice cultures from five female polar cod (n = 5/ group, paired design) were exposed to BaP alone (10 µM), or in combination with low concentrations of EE2 (5 nM), to mimic physiological estradiol levels in early vitellogenic female fish. Transcriptome analysis (RNA-seq) was performed after 72 h exposure in culture. The results provide a global view of transcriptome responses to BaP, EE2 and their mixture. In the mixture exposure, BaP resulted attenuation of EE2 stimulated gene expression (anti-estrogenic effects). The results from this ex vivo experiment suggest that pollutants that activate the Ahr pathway such as the PAH compound BaP can result in anti-estrogenic effects that may lead to endocrine disruption in polar cod.

Project description:This study was performed to validate the newly developed CGP Atlantic cod 20K oligonucleotide microarray. Atlantic cod (Gadus morhua) received an intraperitoneal injection of either formalin-killed, atypical Aeromonas salmonicida (Asal) or PBS and transcriptional profiles of spleen tissues from Asal-injected fish were compared to those from pre-injection control fish and PBS-injected control fish. Gene expression profiles resulting from this study were compared to those from suppression subtractive hybridization library studies, that were previously performed on the same samples, and to literature. Gene expression patterns of single genes were confirmed by QPCR analysis. This study has shown that the newly developed CGP Atlantic cod 20K oligo microarray platform is a valuable tool for cod genomic research.

Project description:In order to investigate the underlying mechanisms of PCB 153 mediated toxicity to Atlantic cod (Gadus morhua), we analyzed the liver proteome of fish exposed to various doses of PCB 153 (0, 0.5, 2 and 8mg/kg body weight) for two weeks and examined the effects on expression of liver proteins using quantitative proteomics. Label-free mass spectrometry enabled quantification of 1272 proteins, and 78 were differentially regulated between PCB 153 treated samples and controls. Two proteins downregulated due to PCB 153 treatment, Glutathione S-transferase theta 1 (GSTT1) and sulfotransferase family protein 1 (ST2B1), were verified using selected reaction monitoring (SRM). Supported by bioinformatics analyses, we concluded that PCB 153 perturbs lipid metabolism in the Atlantic cod liver and that increased levels of lipogenic enzymes indicate increased synthesis of fatty acids and triglycerides.
