Project description:Drosophila larval ventral nerve cord (VNC) shares many similarities with the spinal cord of vertebrates and has emerged as a major model for understanding the development and function of motor systems. We use high quality single cell RNA sequencing to create a comprehensive atlas of larval VNC cell types. Our atlas provides a high-resolution characterization of larval VNC capturing primary neurons, glia and the functional landscape that coordinates larval behavior. At the same time, this atlas offers unique insights into neurogenesis and into the strategies and signaling networks utilized for generation of the adult VNC.

Expression time series during the differentiation of ventral motor neurons from embryonic stem cells

Project description:The aim of this study is to profile gene expression dynamics during the in vitro differentiation of embryonic stem cells into ventral motor neurons. Expression levels were profiled using Affymetrix microarrays at six timepoints during in vitro differentiation: ES cells (Day 0), embryoid bodies (Day 2), retinoid induction of neurogenesis (Day 2 +8hours of exposure to retinoic acid), neural precursors (Day 3), progenitor motor neurons (Day 4), postmitotic motor neurons (Day 7). The differentiation of ventral motor neurons is induced by treating embryonic stem cell cultures with retinoic acid and hedgehog agonist. Here, gene expression patterns are profiled at various defined stages during the differentiation process using Affymetrix expression arrays.

Project description:Circulating sex steroids are critical for the development of neuroplasticity in the respiratory motor system. A primary locus of inducible respiratory neuroplasticity is the phrenic motor nucleus, a column of motor neurons in the ventral cervical spinal cord (C3-C5). In specific, exposure to acute intermittent hypoxia (AIH) induces strengthened synaptic connections to phrenic motor neurons and increased motor output; a form of neuroplasticity called phrenic long-term facilitation (pLTF). Gonadectomy (surgical removal of the testes in males and ovaries in females) reduces circulating sex steroids and eliminates AIH-induced phrenic pLTF in both females and males, implicating sex steroids as critical mediators for the development of neuroplasticity. To begin interrogating mechanisms of sex steroid influence on phrenic motor neurons and to determine the effect of gonadectomy on local gene expression, we performed RNA sequencing on ventral cervical spinal cord samples containing the phrenic motor nucleus. Our findings identified sex-specific gene expression in gonadally-intact rats and suggested that gonadectomy caused greater changes to the transcriptomes of male rats compared to females.

Motor cortex after C3 lesion

Project description:Gene expression analysis of motor cortex after spinal C3 lesion Dorsal column wire knife lesions: Adult female Fischer 344 rats weighing 150-200 gm were used. Animals underwent a laminectomy at spinal level C3. Dorsal funiculus lesions were made in the middle of C3 using a Kopf microwire device (Kopf Instruments, Tujunga, CA). After fixation in a spinal stereotaxic unit, a small dural incision was made. The wire knife was lowered into the spinal cord to a depth of 1.1 mm ventral to the dorsal cord surface and 1.1 mm to the left of the midline. The tip of the wireknife was extruded, forming a 2.25 mm-wide arc that was raised to the dorsal surface of the cord. To ensure complete axotomy of the dorsal funiculus, spinal tissue was compressed against the microwire knife surface using a microaspiration pipette until all visible white matter was transected. Cortical microdissection: The forelimb and hindlimb motor cortex were microdisected from rat cortices: Rostral to Bregma: an area from 2.0 to 4.5 mm mediolateral and from 0 to 2 mm anterior-posterior Caudal to Bregma: an area from 2.0 to 3.5 mediolateral and from 0 to 3 mm caudal to Bregma. Only the inferior half of the cortex containing layer V corticospinal motor neurons was sampled in each region.
