We infected adult human intestine epithelium stem cell derived colonic chip with wild-type S.Tm 14028 with GFP label. To link intracellular replication levels to host response, we performed single-cell total RNA sequencing (VASA-seq) on index-sorted cells from GFPLow, GFPInt and GFPHigh levels.

scRNA-seq of FACS sorted epithelium after infections with S.Tm WT and mutants
Infection of LUVA mast cells with Salmonella Typhimurium wt and S.Tm ∆invG
Infection of bone marrow-derived mast cells with Salmonella Typhimurium WT and S.Tm ∆invG
To resolve host transcriptional responses induced by luminal or intracellular S.Tm 14028, we performed scRNA-seq on GFP+ (invaded, “In”) and GFP− (non-invaded, “Out”) cells infected with WTGFP, ΔSPI-1/2GFP, ΔssaVGFP (\"ΔSPI-2\"), or ΔsteEGFP S.Tm strains at 24 hpi

Human LUVA mast cells were infected for 4 h with Salmonella Typhimurium SL1344 wildtype (wt) and S.Tm ∆invG to explore LUVA mRNA expression levels upon infection and the role of type-3-secretion-system mediated invasion.

Murine bone marrow-derived mast cells (BMMC) were infected for 4 h with Salmonella Typhimurium SL1344 wildtype (WT) and S.Tm ∆invG to explore BMMC mRNA expression levels upon infection and the role of type-3-secretion-system mediated invasion.

Single-cell total RNA sequencing (VASA-seq) on index-sorted human colonic chip infected by wild-type S.Tm
Intracellular pathogens, such as Salmonella enterica serovar Typhimurium (S.Tm), are able to sense and respond to a changing host cell environment. Macrophages exposed to microbial products undergo metabolic changes that are increasingly understood to drive a productive inflammatory response. Howeve...
