In this experiments, we used several ubiquitin activity-based probes (Ub-Prg, Ub-VME, and Ub-VS) to covalently capture ubiquitin-interacting proteins in Legionella pneumophila. These enriched interactome were analyzed by LC-MS/MS to identify Legionella effectors uniquely captured by the probes.

Ubiquitin-specific protease 30 (USP30) is a deubiquitylating enzyme (DUB) localized in the mitochondrial membrane, which is related to PINK1/Parkin-mediated mitophagy, pexophagy, BAX/BAK-dependent apoptosis, and IKKβ-USP30-ACLY-regulated lipogenesis/tumorigenesis. Mission therapeutics pointed their ...

The nucleophilic thiol-alkyne addition reaction can form a covalent thiovinyl bond with cysteine reactivity specific,as the classical standards for cysteine-reactive warheads in activity-based probes (ABPs).Utilizing this classical reactivity property, we designed two potentially reactive unnatural...

The cullin-RING E3 ligase (CRL) network comprises over 300 unique complexes that switch from inactive to activated conformations upon site-specific cullin modification by the ubiquitin-like protein NEDD8. Assessing cellular repertoires of activated CRL complexes is critical for understanding eukaryo...

Upon infection of host cells Legionella pneumophila releases a multitude of effector enzymes into the hosts cytoplasm that hijack a plethora of cellular activities, including the hosts ubiquitination pathways. Effectors belonging to the SidE-family are involved in non-canonical serine phosphoribosyl...

Post-translational modification of proteins by Ubiquitin (Ub) and Ubiquitin-like proteins (Ubls) can be reversed by deconjugating enzymes, which have been implicated in different pathways and associated with various human diseases. To understand the activity and dynamics of deconjugating enzymes, mu...

Almost all cellular processes are influenced by ubiquitination. A large family of enzymes known as E3 ligases provide the specificity for ubiquitination, with the largest class among them, the RING E3s, comprising over 600 members in humans. RING E3s facilitate transfer of ubiquitin to substrates by...

The 26S proteasome is the endpoint of the ubiquitin-proteasome system, an essential pathway for maintaining cellular homeostasis through targeted degradation of misfolded, damaged, and obsolete proteins. Substrates labeled with ubiquitin are directed to the 26S proteasome by binding to one or more u...
