Project description:Fate determination and maintenance of fetal testes in most mammals occur cell autonomously as a result of the action of key transcription factors in Sertoli cell. In the mouse testis AMH and Activin B are required for the maintenance of Sertoli cell fate. In the absence of both AMH and Activin B initial testis differentiation (E12.5) occurs normally but, by embryonic day 15.5, Sertoli cells at the testis poles transdifferentiate into granulosa cells.

Project description:The ability of male reproduction is seriously dependent on Sertoli cells. However, the mechanisms governing the functional integrity of Sertoli cells remained largely unexplored. Tyrosine phosphatase protein Shp2 is expressed in germ, Leydig and Sertoli cells of mice testes. But the physiological role of Shp2 in the spermatogenesis was not fully understood. Thus, we conditionally deleted Shp2 gene in Sertoli cells using two transgenic models, and demonstrated that Shp2 deficiency caused infertility, excessive differentiation of SSCs and abnormal BTB in mice. To further discover the underlying mechanism of Shp2 regulation, we collected the mRNA of testes from wild type or knockout mice at 16.5 fetal day, Postnatal 3 days, 1 weeks, 2 weeks, and then screened the gene expression. Testis tissues from Shp2f/f and SCSKO mice were for RNA extraction and hybridization on Affymetrix microarrays.

Project description:This dataset represents genes that are dysregulated in the postnatal day 12 (P12) mouse testis when ATRX is specifically inactivated in Sertoli cells (ScAtrxKO mice). The differentially expressed genes included in the dataset may play important roles in the testicular phenotypes observed in the ScAtrxKO mice, which were first reported in our previous work (Bagheri-Fam et al., 2011). In fetal ScAtrxKO mice, Sertoli cells undergo apoptosis due to cell cycle defects, resulting in smaller testes with reduced tubule volume. Adult ScAtrxKO mice show a wide range of spermatogenesis defects probably due to a failure of the dysfunctional ATRX protein to interact with the androgen receptor (AR). The chromatin remodeling protein ATRX is widely expressed in the human testis including Sertoli cells. In XY individuals, the loss of ATRX leads to ATR-X (alpha thalassemia, mental retardation, X-linked) syndrome associated with a wide range of genital abnormalities such as hypospadias, ambiguous genitalia, and small testes with reduced tubule volume. Our dataset contributes towards understanding the mechanism underlying ATRX regulation of testis development and spermatogenesis.

Project description:The androgen receptor is a steroid receptor belonging to the superfamily of hormone-activated transcriptional factors, displaying distinct expression profiles in Sertoli cells during testis development, tightly correlated to the stages of spermatogenesis.The aim of the project was to better understand the AR signaling pathways, and identify androgen regulated genes in a mature Sertoli cell line (ST38c). In order to identify androgen regulated candidate genes we compared the gene expression of mature Sertoli cells (ST38c) in the absence (condition B) and in the presence of a androgenic ligand, dihydrotestosterone (condition ST). In order to understand the AMH gene repression in mature Sertoli cells, which occurs at puberty, soon after the augmentation of the AR expression, we also compared the gene profile of an immature, not expressing the androgen receptor, prepubertal Sertoli cell line (SMAT1-condition A) with the mature Sertoli cell line (ST38c- condition C), expressing the androgen receptor. The Sertoli cell line SMAT1 has been obtaiend from a 6-day old transgenic male mouse (using the targeted oncogenesis SV40). This cell line has been kindly provided to us by Dr Jean Yves Picard (UMR 782), Universite Paris Sud. (Dutertre M, et al., Mol Cell Endocrinol. 1997 Dec 31;136(1):57-65. PMID 9510068).The Sertoli cell line ST38c was obtained in the INSERM U-693 from 8-wk-old male murine testis; from a transgenic mouse carrying the construct in which the SV40 large T Antigen (TAg) was placed under the control of the human vimentin promoter. The development and description of this new cell line have not yet been published.

Project description:To unravel the molecular mechanisms mediating the effects of androgens on spermatogenesis, testicular gene expression was compared in mice with a Sertoli cell-selective androgen receptor knockout (SCARKO) and littermate controls on postnatal d 10. At this age testicular cell composition is still comparable in SCARKOs and controls. Microarray analysis identified 692 genes with significant differences in expression. A more than 2-fold up- or downregulation by androgen action in Sertoli cells was observed for 28 and 6 genes respectively. The biological relevance of the "strongly" upregulated genes was supported by the finding that several of them were previously described to be androgen-regulated or essential for spermatogenesis. Serine protease inhibitors were overrepresented in the same subgroup suggesting a role for androgens in cell junction dynamics and tissue restructuring events during spermatogenesis. A time course experiment (d8-d20), followed by cluster analysis allowed the identification of typical expression patterns of differentially expressed testicular genes during initiation of spermatogenesis. Three genes with a pattern closely resembling that of Pem, a prototypal androgen-regulated gene in Sertoli cells, were selected for confirmation by RT-PCR and further analysis. The data confirm that the SCARKO model allows identification of novel androgen-regulated genes in the testis. This series represents the time course data from d 8, d 10, d 12, d 16 and d 20. The expression data from the additional SCARKO vs control comparison on d 10 is represented by series GSE2260 (Testicular gene expression in SCARKO mice at day 10).
