Transcription profiling of yeast wild type BY4741, single deletion mutants delta-pbf1, delta-pbf2 and double mutant delta-pbf1 delta-pbf2 before and after heat shock

ABSTRACT: We used microarrays to detail the global program of gene expression underlying rRNA processing gene regulation during heat shock. PBF1 is YBL054W (TOD6) and PBF2 is YER088C (DOT6). Experiment Overall Design: S. cerevisiae cells were grown @ 25 ÂºC to O.D. 600 of 0.3~04, in triplicate. Half of the cells were collected at T0 and half of the cells were subjected to 25 ÂºC-->37 ÂºC heat shock and cells were collected after 20 min, as samples T20. Total RNAs were collected and used to perform expression microarray analysis on Affymetrix Yeast Array 2.0 platform. The changes of gene expression upon heat shock were analyzed for four strains, wild type BY4741, single deletion mutants delta-pbf1, delta-pbf2 and double mutant delta-pbf1 delta-pbf2.

Project description:Fusarium graminearum (teleomorph Gibberella zeae) is a prominent pathogen that infects major cereal crops, such as wheat, barley, and maize. To dissect cellular responses toward heat stress in the plant pathogenic fungus F. graminearum, we compared transcriptomes of the fungal cultures incubated in normal temperature condition (25 ÂºC) and in high temperature condition (37 ÂºC) for 15 min. 6 samples examined: 24 h-old mycelia from complete medium (CM) of Fusarium graminearum wild-type Z-3639 were incubated in normal temperature condition (25 ÂºC) and in high temperature condition (37 ÂºC) for 15 min.

Genome-wide expression analysis of dikaryotic colonies of Schizophyllum commune under various conditions: wild-type, delta-wc2, delta-wc2 hsp3(pr)-wc2

Project description:The blue light receptor WC-2 was shown to be involved in mushroom development in the basidiomycete Schizophyllum commune. When the gene encoding WC-2 was deleted, no mushrooms formed and colony morphology was radial. This phenotype was similar to the wild-type colony grown in the dark. This phenotype could be complemented by transforming the wc-2 deletion strain with a construct encompassing the wc-2 coding sequence under the control of the heat inducible promoter hsp3. A daily heat shock of 1 hour at 42 degrees Celsius resulted in mushroom development and an asymmetrical colony. In this study we performed a genome-wide expression analysis on dikaryons of wild-type (not heat shocked), delta-wc2 (heat shocked or not heat shocked) and the complemented strain delta-wc2 hsp3-wc2 (heat shocked or not heat shocked). A total of five samples were analyzed. One wild-type (not heat shocked), delta-wc2 (heat shocked or not heat shocked) and the complemented strain delta-wc2 hsp3-wc2 (heat shocked or not heat shocked).

Transcription profiling of Arabidopdis seedlings - heat, anoxia, and combined heat-anoxia treatments

Project description:Anoxia induces several heat shock proteins and a heat pre-treatment can acclimatize Arabidopsis seedlings to a subsequent anoxic treatment. In this work we analyzed the response of Arabidopsis seedlings to anoxia, heat and a combined heat+anoxia stress. A significant overlapping between the anoxic and heat shock responses has been observed by whole-genome microarray analysis. Experiment Overall Design: We treated Arabidopsis seedling, 4-days old, dark germinated with: Experiment Overall Design: -Control (23Â°C, dark, liquid Murashige-Skoog medium containing 30mM sucrose). Experiment Overall Design: -Heat-treated (38Â°C for 90 minutes, dark, liquid Murashige-Skoog medium containing 30mM sucrose). Experiment Overall Design: -Anoxia-treated (23Â°C, under anoxia for 6h, dark, liquid Murashige-Skoog medium containing 30mM sucrose). Experiment Overall Design: -combined heat+Anoxia-treatment (23Â°C, treated at 38Â°C for 90 min and thereafter under anoxia for 6h, dark, liquid Murashige-Skoog medium containing 30mM sucrose).
