Effects of gene scilencing of USP2 on expression profiles of HL60 cells

ABSTRACT: So far, we have found PMA induced USP2b isoform in myeloid leukemia cell lines such as HL60, THP-1, and U937. To explore molecular function of USP2 in the cells, we assess expression profiles of HL60-derivatives continuously expressing shRNA for USP2 and control shRNA. HL60 derivatives continuously expressing shRNA for USP2 and control shRNA were treated with PMA (30nM) for 0, 1, 2, and 3 days. Total RNA was extracted from the cells and subjected to microarray experiments using Human Genome U133 GeneChips.

Project description:Myeloid leukemia cell lines HL60, THP-1, and U937 undergo macrophage-like differentiation after treatment with phorbol ester. To explore genes whose exon usage was altered during macrophage differentiation, we compared exome of PMA-treated (differentated) and vehicle-treated (undifferentiated) myeloid cell lines. HL60, THP-1, and U937 cells were treated with either phorbol 12-myristate 13-acetate (PMA,30nM) or its vehicle (DMSO) for 3 days, and subjected to exome analysis using Affymetrix human exon 1.0ST arrays.

Effects of supernatants of USP2 knockdown HL-60 cells and mock transfected HL60 cells on differentiated 3T3-L1 cells

Project description:Ubiquitin specifc protease (USP) 2 modulates various cellular responces including carcinogenesis, spermatogenesis, and glucose metabolism. So far, we established USP2 knockdown HL-60 cells to explore roles of USP2 in macrophages. In this experiment, we investigated whether USP2 knockdown HL-60 cells more prominently modulate function of adipocytes compared with control cells. USP2 knock down(KD) HL-60 cells and mock vector-transfected mock control cells were differentiated into macrophage-like cells in the presence of 30nM of phorbol 12-myristate 13-acetate for 5 days. Subseqently, supernatants of the USP2 KD cells and control cells were added to differentiated 3T3-L1 cells for 3 and 6 hrs. Total RNA was isolated from the supernatant-treated 3T3-L1 cells, and subjected to microarray analysis. Duplicate samples were prepared for each cell and time point.

Project description:Analysis of gene expression changes associated with lentiviral vector mediated expression of pre-microRNA-150 expression in the AML cell lines PL21 and HL60 cells. To identify genes, direct putative miRNA-150 targets, biological and molecular functions and pathways that distinguish cells that express miR-150 from those that do not. Total RNA isolated from HL60 or PL21 cells transduced with pre-microRNA-150 or empty vector control and assayed by gene expresssion analysis 20 days post transduction. Two replicates were performed for each condition (miR-150 and control) in each of two cell lines (HL60 and PL21).

EPA inhibited cell proliferation in HL60 cells in a dose dependent manner. Gene expression profiling of HL60 cells treated with EPA was performed by Affymetrix GeneChip. System and key markers of relevant pathways were verified at protein level by western blotting.
