Expression data from porcine adrenal gland

ABSTRACT: To gain insight into the adrenal stress response, we analyzed differential mRNA expression of genes associated with stress induced by aggressive behaviour in the pig In an animal experiment various levels of aggressive behaviour and stress were induced by mixing groups of unfamiliar pigs with different aggressiveness. To analyze adrenal stress response we selected two experimental groups for comparison, each comprising eight animals (n=16) which differed significantly in aggressive behaviour and plasma cortisol levels. To identify differentially expressed genes, we compared the adrenal transcriptome of these two groups of pigs, using the Affymetrix GeneChip porcine Genome Array.

Role of Adrenal Steroid Metabolites in Osteoporosis in Patients with Autonomous Cortisol Secretion

Project description:Background: Autonomous cortisol secretion (ACS), results from cortisol-producing adenomas (CPA), causes endogenous steroid-induced osteoporosis (SIOP). However, how other adrenal steroid metabolites affect bone status is unclear. Methods: ACS was diagnosed at serum cortisol after 1-mg dexamethasone suppression test (DST-cortisol) ≥ 1.8 g/dL. Using liquid chromatography-tandem mass spectrometry, we measured 21 plasma steroid metabolites in 73 patients with ACS and 85 with non-functioning adrenal tumors (NFAT). We also examined expression of steroidogenic enzymes and relevant steroid metabolites in some of CPA tissues. Results: In discriminant and principal component analysis, steroid profiles distinguished between the ACS and NFAT groups in premenopausal women. Premenopausal women with ACS exhibited higher levels of a mineralocorticoid metabolite; 11-deoxycorticosterone (11-DOC), with lower androgen metabolites; dehydroepiandrosterone-sulfate and androsterone-glucuronide. In premenopausal women with ACS, DST-cortisol was negatively correlated with trabecular bone score (TBS). Additionally, 11-DOC was negatively correlated with lumbar spine-bone mineral density (r = -0.603), while androsterone-glucuronide was positively correlated with TBS (r = 0.681), which was supported by Bayesian kernel machine regression analysis. There were no such correlations in postmenopausal women and men. The CPA tissues showed increased levels of 11-DOC, with increased expression of CYP21A2, which is essential for 11-DOC synthesis. The adrenal non-tumor tissues were atrophied with reduced expression of CYB5A, which is required for androgen synthesis. Conclusion: This study provides the first evidence that unbalanced production of adrenal steroid metabolites, which are derived from both adrenal tumors and non-tumor tissues, play a role in the pathogenesis of endogenous SIOP in premenopausal women with ACS.

Time course of the response to ACTH in pig: biological and transcriptomic study

Project description:To explore individual variation in the biological response to cortisol, the main stress hormone, in pigs, we have developed a microarray analysis to study the kinetics of the response to an ACTH injection measured at 4 time points. ACTH is a hormone secreted by the pituitary gland and triggering the release of cortisol by the adrenal glands in response to a stress. The objective was to identify genes up- or down-regulated during the stress response triggered by cortisol in the whole blood. To explore individual variation in the biological response to cortisol, whole blood from 30 juvenile Large White (LW) pigs was collected at 4 time point ( t = 0, +1 h, +4 h, +24 h) for a total of 120 samples. All pigs were females and were raised in two batches. Arrays were designed such that all observations of a sole individual were kept on the same array, with two individuals (one from each batch) on one array. This design secured the kinetics of the response for each individual and prevented confounding effects between batch and array. After quality and control, 120 samples were kept for normalization and analyses (represented here).

Domestication Effects on Stress Induced Steroid Secretion and Adrenal Gene Expression in Chickens

Project description:Background Understanding the genetic basis of phenotypic diversity is a challenge in contemporary biology. Domestication provides a model for unravelling aspects of this, for example with respect to stress sensitivity. Ancestral Red Junglefowl (RJF) show more fearful behaviour and a more pronounced HPA-axis reactivity than its domesticated descendant White Leghorn (WL). By comparing adrenal global gene transcription profiles between WL and RJF as well as their plasma levels of adrenal hormones in response to an acute stress event, we aimed at investigating the molecular basis for the altered physiological stress responsiveness in domesticated chicken.

Cell Atlas at Single-Nuclei Resolution of the Adult Human Adrenal Gland and Adrenocortical Adenomas

Project description:The human adrenal glands are highly dynamic endocrine organs that are involved in the secretion of various hormones such as steroids and catecholamines. Here we present a single-nuclei and spatial transcriptomic analysis of healthy adult human adrenal glands to provide a complete adrenal gland atlas. With this, we show how such an atlas can be taken advantage when studying adrenocortical diseases, such as adrenocortical adenomas (ACA). Using nornal adrenal as reference, we showed a high intra-tumoural heterogeneity in the single-nuclei transcriptome of ACA, revealing the presence of specific cell populations associated with cortisol secretion and genetic background.

Adrenal gland gene expression in male pigs from dams fed diets supplemented with algae or fish oil and challenged with LPS in late gestation

Project description:Maternal stress occuring during gestation can influence the functioning of the stress and immune responses in offspring. Maternal supplementation with immunomodulatory compounds such as omega-3 polyunsaturated fatty acids may reduce inflammation associated with maternal stress, promoting offspring health and growth Sows were fed diets supplemented with microalgae or fish oil from gestation day 75 and were challenged with LPS on gestation day 112. Previously, LPS challenge in the male piglets of these sows resulted in differences in fever and cortisol responses among piglets from different maternal treatments. Therefore, the adrenal gland was chosen for microarray analysis to assess the effects of the maternal treatment on the gene expression within the hypothalamic-pituitary-adrenal axis.
