Bacterial Adaptation Through Loss of Function

ABSTRACT: Transcriptional profiling of mutants with beneficial in-frame null mutations. The goal of the study is to understand how expression of the mutants and their parental strain differs in conditions where the mutations are beneficial. For each of two conditions (media with either glutamine or alanine as the sole carbon source) four mutants were compared with the parental strain. Two biological replicates were done for each comparison. A total of 16 arrays were used.

ORGANISM(S): Escherichia coli str. K-12 substr. MG1655

The metabolic capabilities and regulatory networks of bacteria have been optimized by evolution in response to selective pressures present in each species' native ecological niche. In a new environment, however, the same bacteria may grow poorly due to regulatory constraints or biochemical deficiencies. Adaptation to such conditions can proceed through the acquisition of new cellular functionality due to gain of function mutations or via modulation of cellular networks. Using selection experimen  ...[more]
