Transcription profiling of human embryonic stem cells (hES) during the differentiation.

ABSTRACT: The transcriptome analysis was performed in triplicate using two human embryonic stem cells lines (hES_VUB01 and hES_SA01) by comparing the expression profiles of the undifferentiated hES cells and two types of progenitors derived from the hES cell lines: Neural progenitors (NPC) and Mesodermal progenitors (MSC). Keywords: Cell type comparison hES (6 samples originated from the two cell line, in triplicate for each : hES_VUB01_1, hES_VUB01_2, hES_VUB01_3, hES_SA01_1, hES_SA01_2, hES_SA01_3) ; NPC (6 samples originated from the two cell line, in triplicate for each : NPC_VUB01_1, NPC_VUB01_2, NPC_VUB01_3, NPC_SA01_1, NPC_SA01_2, NPC_SA01_3) ; MSC (6 samples originated from the two cell line, in triplicate for each : MSC_VUB01_1, MSC_VUB01_2, MSC_VUB01_3, MSC_SA01_1, MSC_SA01_2, MSC_SA01_3)

Human embryonic stem cells can be differentiated along different lineages, providing the possibility of a precise analysis of genes profiles associated with specific commitments. Subtractive gene expression profiling between differentiated and undifferentiated cells provides lists of potential actors in this commitment. This combines, however, genes that are specifically associated with development and others that are over expressed because of nonlineage-specific differentiation systems. As a wa  ...[more]
