ABSTRACT: Candida albicans lab strain SC5314 was spread on YPD+uniconazole (0.06-4μg/mL) plates. Randomly 4 adaptors from each drug plate were sequenced.
Project description:Gene AFR1 was deleted from Cryptococcus neoformans lab strain H99. Approximately 1 million cells of the deletion strain were spread on YPD-agar plate supplemented with 0.125ug/mL uniconazole. Randomly 30 adaptors were chosen. 16 adaptors were resistant to uniconazole and were sequenced.
Project description:Approximately 1 million cells of Crypotoccus neoformans lab strain H99 were spread on YPD-agar plate supplemented with 1 ug/mL uniconazole. After 5 days incubation of the plate at 30℃, randomly 30 adaptors were chosen and sequenced.
Project description:Candida albicans lab strain SC5314 was spread on YPD plates supplemented with echinocandins (caspofungin, anidulafungin, micafungin, rezafungin). The adaptors were tested for tolrance to fluconazole. Randomly 43 tolerant adaptors, as well as the parent SC5314, were sequenced.
Project description:Candida albicans lab strain SC5314 was spread on YPD plates supplemented with ketoconazole (0.125-16μg/ml); Randomly 3 adaptors from each plate were sequenced.
Project description:H99 derived 2 ug/mL uniconazole adaptors: Approximately 1 million cells of Crypotoccus neoformans lab strain H99 were spread on YPD-agar plate supplemented with 2 ug/mL uniconazole. After 5 days incubation of the plate at 30℃, randomly 30 adaptors were chosen. 21 adaptors were resistant and were sequenced. H99 derived 0.25 ug/mL uniconazole adaptors: Cells of H99 were adjusted to 2500 cells/mL. The culture was supplemented with 0.25 ug/mL uniconazole. After 72 h incubation, the culture was washed, diluted and approximately 200 cells were spread on YPD-agar plate. Randomly 95 colonies were tested for resistance to uniconazole. One colony (TJ3832) was resistant and was sequenced.
Project description:Approximately 200 cells of Candida albicans lab strain SC5314 were spread on YPD plates supplemented with 2 fold increase from 50 ng/ml to 12800ng/ml caspofungin. All adaptors appeared were collected. Randomly 36 tolerant adaptors were sequenced.
Project description:In Candida albicans, the lab strain SC5314 was grown in YPD broth supplemented with 4ug/ml brefeldin A for 24h. The culture was washed, diluted and spread on YPD. Randomly 120 colonies were tested for tolerance to brefeldin A. Two adaptors (FY200 and FY201) obtained tolerance. These two adaptors were sequenced.
Project description:Candida albicans strains SC5314 and YJB-T1891 were used as wild type to delete VMA11. The deletion strains (TJ926 and TJ927) were spread on YPD plate supplemented with 1μg/ml fluconazole. Randomly 96 adaptors derived from TJ926 and 96 adaptors derived from TJ927 were tested with disk diffusion assay. Among the resistant adaptors, randomly 15 adaptors (A4-A18) and 48 adaptors (A19-A66) derived from TJ926 were sequenced.
Project description:From C. albicans reference strain SC5314, CDR1, CDR2, MDR1, CMP1, CNB1, CRZ1 were deleted. The deletion strains were spread on YPD plates supplemented with tebuconazole. Randomly 4 adaptors derived from each deletion strain were sequenced.
