Project description:While exercise effects on the immune system received increasing attention in recent years, it remains unclear to what extent gender and fluctuations in sex hormones during menstrual cycle influence immunological responses to exercise. Using a previously published microarray data set, we investigated mRNA changes induced through exhaustive exercise (half-marathon; pre-exercise and post-exercise [30 min, 3h, 24h], whole blood culture ± LPS (1h)) with a specific focus on sex differences (women in luteal phase vs men). Inflammation related pathways, TLRs, cytosolic DNA sensing and RIG-I like receptors were differentially activated between sexes in LPS-stimulated cultures. Genes differentially regulated between sexes included TNIP-1, TNIP-3, IL-6, HIVEP1, CXCL3, CCR3, IL-8 and CD69, revealing a bias towards less anti-inflammatory gene regulation in women compared to men. In addition, several genes relevant to brain function (OLIG2, TMEM106B, DDIT4, and KMO) showed differential activation between sexes. Some of these genes (e.g., KMO in women, DDIT4 in both sexes) potentially constitute neuroprotective mechanisms. These data reveal that the exercise-induced change in gene expression might be gender and menstrual cycle phase dependent. Project description:Autoimmune polyendocrine syndrome type I (APS-1) is a rare and devastating organ-specific autoimmune disease characterised by mutations in the Autoimmune Regulator (AIRE) gene. As AIRE is crucial for negative selection in the thymus, increased numbers of undesirable autoreactive T cells are released into the blood with the potential to cause tissue injury, including endocrine organ failure, chronic mucocutaneous candidiasis and hepatitis. B cells are also affected and produce high amounts of neutralising autoantibodies against both cytokines and organ-specific targets. The Aire-deficient mouse model has informed to some extent about immunological aspects of APS-1, but the rarity of APS-1 and inaccessibility of thymic tissue have severely limited immunological studies in patients. Sampling of APS-I patients and controls was performed in standardized manners in PAXgene blood RNA tubes (PreAnalytix, Qiagen, Hombrectikon, Switzerland) and stored at 80ºC until use. Purification of RNA was achieved by the PAXgene blood RNA kit following the instructions from the manufacturer. Samples were quality assessed by Agilent Bioanalyzer, using the Agilent 6000 Nano kit (Agilent, Santa Clara, CA, USA), providing RNA with RNA integrity numbers (RIN) above 6.0. The samples were randomly distributed into 4 batches for RNA extraction, each with 4 patients and 4 sex- and age matched controls and was extracted by the same person on the same day. Following the procedures from Illumina, RNA was subsequently transformed to cRNA, and these constructs were labeled, amplified and quality-checked again by the Agilent Bioanalyzer. The cRNAs were then hybridized to 4 Illumina HumanRef-8 BeadChip microarrays, followed by washing and scanning according to the protocol. Quality control of the arrays was done by BeadStudio.

A Dexamethasone Suppression Test was performed in 297 subjects. Project description:While exposure to adverse life events and subsequent dysregulation of the stress hormone response broadly confer risk for depression, the specific molecular mechanisms mediating this risk are poorly understood. Through pharmacologic activation of the stress hormone response in blood cells we demonstrate that common genetic variants in long-range enhancer elements moderate the immediate transcriptome response to stress, and that these genetic differences are associated with increased risk for depression in the context of early adversity. Using imaging genetics we then link these common risk variants with dysregulated amygdala reactivity, an important trigger of the stress hormone response. The transcripts regulated by these risk variants in peripheral blood were also responsive to stress and stress hormone receptor activation in murine brain. Network modeling approaches suggest that these differences in transcriptional activation may mediate stress-related risk for depression by altering a functional gene network related to proteasome degradation and synaptic plasticity. A Dexamethasone Suppression Test was performed in 160 male subjects. Baseline and stimulated (3 hours after 1.5 mg dexamethasone p.o.) whole blood samples were analyzed using Illumina Human HT-12 v3 arrays.

SNP-probe pairs with a high likelihood of cross-hybridization and SNP-in-probe effects were excluded to exclude false positive results. We identified 102 cis-acting and 5 trans-acting eQTL regions. The cis-eQTL regions were widely distributed both upstream and downstream of the gene, as well as within the gene. RNA microarray data obtained from 3 independent samples originally recruited for other studies investigating the gene expression levels in psychiatric disorders were used in the present study. For the purpose of the present analyses, genomic DNA was collected from 24 subjects (13 men and 11 women, mean age [SD] = 39.9 [7.6] years) in sample 1, 24 subjects in sample 2 (12 men and 12 women, 34.1 [11.5] years), and 28 subjects (14 men and 14 women, 41.4 [11.8] years) in sample 3. Some of the subjects had depressive symptoms, but all were physically healthy and without clinically significant systemic disease (e.g., malignant disease, diabetes mellitus, hypertension, renal failure, or endocrine disorders). Subjects were recruited from the outpatient clinic of the National Center of Neurology and Psychiatry Hospital, Tokyo, Japan, through advertisements in free local information magazines or through our website announcement. All the subjects were biologically unrelated Japanese individuals who resided in the same geographical area (western Tokyo). The study protocol was approved by the ethics committee at the National Center of Neurology and Psychiatry, Japan. Written informed consent was obtained from every subject after the study was explained to them. Venous blood was collected between 1100 and 1200 h in PAXgene tubes (Qiagen, Valencia) from each subject and was incubated at room temperature for 24 h for RNA stabilization. RNA was extracted from whole blood according to the manufacturer’s guidelines by using the PAXgene Blood RNA System Kit (PreAnalytix GmbH, Hombrechtikon, Switzerland). The RNA was quantified by optical density readings at A260nm by using the NanoDrop ND-1000 (Thermo Scientific, Rockford). Gene expression analysis was performed using Agilent Human Genome 4 × 44 K arrays (Agilent Technologies, Santa Clara).
