RNA-Seq of vehicle- vs LPS-treated primary BMDMs from WT or interleukin-1alpha nuclear localisation sequence mutant mice

ABSTRACT: We generated a mouse with a mutated IL-1alpha nuclear localisation sequence, causing loss of pro-IL-1alpha nuclear localisation. We tested whether this loss of nuclear pro-IL-1alpha affected the transcriptional response of primary BMDMs to LPS priming. Primary BMDMs from WT or mNLS mice were treated with vehicle or LPS (1ug/ml, 4h).

We tested whether this loss of nuclear pro-IL-1alpha affected the response of these mice to Influenza A infection. WT or mNLS mice were anaesthetized by 2.5% inhalation isoflurane and intranasally infected with 10^3 PFU of the IAV strain X31 (H3N2), in a volume of 30 µL PBS, on day 0. Infected WT and mNLS mice, and heterozygous control uninfected mice, were culled on day 7 post-IAV X31 infection, and the lung RNA was extracted.

Host resistance to pulmonary infection with Mycobacterium tuberculosis is mediated by IL-1alpha.

Project description:Background: M.tuberculosis is one of the most prevalent and deadly human pathogens. The molecular mechanisms determining the outcomes of an infection with M.tuberculosis, that range from resistance to an active progressive disease, remain incompletely understood. Here we provide the evidence that IL-1alpha plays a critical and non-redundant role in enabling host resistance to pulmonary M.tuberculosis infection in mice that develop functional pathogen-specific adaptive immunity. Mechanistically, IL-1alpha-deficient mice fail to control M.tuberculosis replication in vivo at a level of individual infected cells and succumb to progressive disease at the late phase of infection. Furthermore, we show that IL-1alpha from hematopoietic compartment through IL-1RI operates upstream of TNFRI-signaling pathway and the lack of IL-1alpha leads to the continuous influx of monocytes that acquire a hyper-inflammatory phenotype and contribute to pathology, rather than to a pathogen control. Cell-type-specific restoration of IL-1alpha expression in CD11c+ subsets of lung leukocytes resulted in reduced levels of inflammatory marker expression on lung cells and improved survival IL-1alpha-deficient mice after M.tuberculosis infection. In humans, genetic analysis of single nucleotide polymorphisms in 14 genes implicated in IL-1-IL-1R signaling pathway revealed association of genetic variations in IL-1alpha and IL-1RAP genes with human susceptibility to pulmonary tuberculosis. Our results implicate IL-1alpha as a principal factor of host resistance to M.tuberculosis and IL-1alpha-driven cell-cell crosstalk as a key step in triggering M.tuberculosis control mechanisms that are critical for host survival.

Project description:Bone marrow was extracted from mice that are COP1-wt Rosa26-CreERT2 or COP1-floxed Rosa26-CreERT2 BMDMs were obtained by culturing bone marrow precursors in media containing 20% of supernatant from L929 cells. m6A modification maps of mRNA in bone marrow-derived macrophages (BMDMs) from mouse

Project description:We report the application of sequencing technology for high-throughput profiling of mRNA m6A methylation in mice BMDMs with or without PBS treatment. By obtaining over thirty billion bases of sequence from m6A antibody immunoprecipitated mRNA, we generated m6A methylation maps of mouse BMDMs in the presence or absence of LPS. We find that SOCS1 mRNA methylation is robustly enhanced by LPS treatment in WT BMDMs, leading to the increase of SOCS1 expression. However, LPS challenge fails to increase SOCS1 expression in the M14-/- BMDMs, indicating m6A modification plays a cirital role in the SOCS1 expression. This study provides a framework for the application of comprehensive mRNA m6A profiling towards characterization of BMDMs.

Project description:BMDMs or iMACs were generated via differentiation of bone marrow cells or immortalized precursors, respectively, in culture media containing m-CSF for 7 day. Upon differentiation, BMDMs or iMacs were left untreated or stimulated as described below. 1) BMDM WT were stimulated with LPS, PGE2, IL-4, IL-10, LPS+PGE2, LPS+IL-4, LPS+IL-10 for 4 hours. Fragmented chromatin was immuno-precipitated for H3K27Ac (each condition at least in duplicate). These experiments were performed to investigate the cross-antagonism in the concomitant presence of pro-inflammatory (LPS) and anti-inflammatory stimuli (IL-4, IL-10, or PGE2). 2) BMDM WT were stimulated with LPS, PGE2, LPS+PGE2 for either 2 or 4 hours. Fragmented chromatin was immuno-precipitated for IRF1(4h), STAT1 (4h), PU.1 (2h, also for IL-10 and LPS+IL-10 stimulation), NFkB (2h), MEF2A (2h), MEF2D (2h), MEF2C (2h), JUNB (2h) (each condition in single). These experiments were performed to assess TFs occupancy at previously defined PGE2-sensitive and PGE2-resistant enhancers. 3) MEF2A-deficient iMac clones (D7, A7, A8, C7) and MEF2A-proficient (referred to as wild-type) iMac clones (NE, B3 and D10) were generated via CRISPR/Cas9 and stimulated or not with LPS for either 2 or 4 hours: each condition in single. Fragmented chromatin was immuno-precipitated for H3K27Ac (4h) or for PU1 (2h). These experiments were performed to investigate the role of MEF2A on LPS-induced chromatin remodeling.
