Phosphoproteomics Uncovers a Neuroimmune Perspective on Trigeminal Neuralgia: Sexually Dimorphic Regulatory Networks Linking Calcium Channels to the Complement Cascade

Project description:Background: Trigeminal neuralgia (TN) is a debilitating neuropathic pain disorder with a higher prevalence in females, yet its pathogenic mechanisms and underlying sexual dimorphism remain incompletely understood. Phosphorylation modifications are critical regulators of pain signaling, but comprehensive phosphoproteomic profiling of TN has not been reported. Methods: We established a chronic constriction injury model of the infraorbital nerve (ION-CCI) in male and female rats via an intraoral approach. Mechanical allodynia was behaviorally confirmed. Integrated proteomic and phosphoproteomic analyses of trigeminal ganglia were performed at postoperative day 7 using data-independent acquisition mass spectrometry. Bioinformatic analyses included pathway enrichment, kinase-substrate prediction, and drug target annotation. Results: Key findings include: (1) Proteomics identified 5,820 proteins and phosphoproteomics revealed 8,830 phosphopeptides mapping to 8,139 phosphoproteins; (2) Complement component C1qa was upregulated in both sexes, while kininogen (KNG1/2) showed female-specific elevation; (3) MAPK pathway activation was prominent, with significantly increased HSPB1 phosphorylation at S86, predicted to be regulated by MAPKAPK2/3; (4) N-type calcium channel (CACN1B) phosphorylation was enhanced in both sexes, suggesting a CACN1B-MAPK-HSPB1 signaling axis; (5) Drug annotation identified Ziconotide (CACN1B blocker) as a therapeutic candidate. Conclusions: This study reveals a neuroimmune phosphorylation network in TN pathogenesis, establishing complement activation and the CACN1B-MAPK-HSPB1 axis as central mechanisms. Sexually dimorphic expression of complement and kinin components provides molecular insights into TN's female predominance. These findings offer therapeutic targets and support sex-specific immunomodulatory strategies for TN.

Genome-wide binding signature of Signal transducer and activator of transcription (STAT) family in colorectal cancer HCT 116 cell

Project description:The excessive and continuous activated Janus kinase/signal transducer and activator of transcription (JAK/STAT) pathway leads to the proliferation and migration of malignant cells resulting in the occurrence and development of almost all of cancers. The defined gene sets specifically activated by different STAT proteins are relied on their genomic accessibility by the interplay of certain STAT proteins with other potential cofactors. However, we have no clue about the status of human activated STAT dimers in the nucleus, as well as the intercrossing modules of synergic, supplementary or competitive relationship among each other in colorectal cancer. In current study, chromatin immunoprecipitation sequencing (ChIP-seq) was conducted to explore the genome-scale binding signatures of STAT1, STAT2, STAT3, STAT5A/B and STAT6 in human HCT-116 CRC cells. Moreover, STAT3 binding on genomic DNA was also investigated in HCT116 cells with NR5A2 knockdown.

Transcriptome changes in trigeminal ganglia in the mouse chronic constriction injury of the infraorbital nerve model - role of TRPM3.

Project description:In this project, we performed bulk RNAseq of mouse trigeminal ganglia at different time points following a constriction of the infraorbital nerve (IoN-CCI model). Samples include both ipsilateral and contralateral samples, as well as samples from sham-operated mice. To evaluate a possible role of TRPM3, trigeminal ganglia were isolated from both wild type and TRPM3-knockout mice. Our data indicate profound and time-dependent transcriptomic changes induced by the IoN-CCI model, but did not reveal IoN-CCI-specific differences between wild type and TRPM3-knockout mice.

A Light-Driven Bioactive Nerve Conduit for Trigeminal Neuralgia: Integrated Decompression and Photosynthetic Oxygenation Restores Mitochondrial Function

Project description:Vascular compression and concomitant hypoxia exacerbate trigeminal neuralgia (TN), yet current therapies rarely target both pathologies. We developed a photosynthetic nerve guide conduit (SFM-Gel@ NGC) by embedding auto fluorescent microalgae in silk fibroin hydrogel, enabling simultaneous mechanical decompression and oxygen release. The construct exhibited suitable mechanical properties, tunable oxygen generation, and excellent biocompatibility. Under hypoxia, SFM-Gel promoted neuronal cell proliferation and function by reducing ROS and restoring mitochondrial activity, while downregulating HIF-1α and ATP/P2X3 pathways. In a rat TN model, auto fluorescent SFM-Gel@ NGC implantation alleviated mechanical allodynia, repaired nerve morphology, and suppressed BDNF/NGF and ATP/P2X3 inflammatory signaling. Transcriptomic analysis confirmed modulation of hypoxia/ROS/NF-κB pathways during immune/inflammatory responses to compression. This integrated decompression-oxygenation strategy offers a transformative therapy for TN.

Overcoming ABCB1 mediated multidrug resistance in castration resistant prostate cancer

Project description:The few available therapies for mCRPC patients include taxanes docetaxel (DTX) and cabazitaxel (CBZ). However, development of resistance limits their clinical use. PCa that relapses after hormonal therapies, referred to as castration resistant prostate cancer (CRPC), often presents with metastases (mCRPC) that are the major cause of mortality. The few available therapies for mCRPC patients include taxanes docetaxel (DTX) and cabazitaxel (CBZ). However, development of resistance limits their clinical use. To study taxanes resistance, we produced CBZ resistant C4-2B cells (RC4-2B) and documented resistance to both CBZ and DTX in cell culture and in 3D prostaspheres settings. RNAseq identified increased expression of ABCB1 in RC4-2B, that was confirmed by immunoblotting and immunofluorescent analysis. ABCB1-specific inhibitor elacridar reversed CBZ and DTX resistance in RC4-2B cells, confirming ABCB1-mediated resistance mechanism.
