RNA-seq of Drosophila exposed to trametinib under normal and high sugar dietary conditions

ABSTRACT: The transcriptional response to the small molecule MEK inhbitor, trametinib, was analysed from RNA extracts prepared from 10-day old adult female Drosophila that had been maintained under normal and high sugar dietary conditions post-eclosion.

Effect of PTEN knockout on dabrafenib-trametinib treatment in A375 melanoma cells - replicate 2

Project description:Approximately 50% of melanomas harbor an activating BRAFV600E mutation. Standard of care involves a combination of inhibitors targeting mutant BRAF and MEK1/2, the substrate for BRAF in the MAPK pathway. PTEN loss of function mutations occur in 40% of BRAFV600E melanomas, resulting in increased PI3K/AKT activity that enhances resistance to BRAF/MEK combination inhibitor therapy. To compare the response of PTEN null to PTEN wild type cells in an isogenic background, CRISPR was used to knock out PTEN in the A375 melanoma cell line that harbors a BRAFV600E mutation. Kinome profiling was performed using the parental line and two PTEN KO clones (5 and 11), treated with DMSO, or treated with 100nM dabrafenib and 10nM trametinib for 1 day or for 7 days. PTEN KO cells showed dramatically increased binding of HER3 and AKT3 compared to wild type. The activation of the SOX10-FOXD3-HER3-AKT axis in PTEN KO cells could be targeted with the ERBB/HER inhibitor neratinib.

Project description:Whole-exome sequencing was performed on DNA samples extracted from seven melanoma cell lines resistant to either vemurafenib (BRAF V600E inhibitor) or trametinib (MEK1/2 inhibitor). The aim of the experiment was to search for genetic alterations responsible for phenotypic diversity of melanoma cell lines reported at the level of cell morphology, activity of signaling pathways essential for melanoma development and progression, and resistance to targeted therapeutics.

Transcriptional profiling of fruit flies overexpressing the Atg1 gene

Project description:Overexpression of Atg1 using either weaker CGGAL4 or stronger HRGAL4 driver. Both drivers have same expression pattern and are specific for intestine, Malpighian tubules and fat body of the fruit fly Drosophila melanogaster. Weaker Atg1 overexpression results in longer lifespan whereas stronger Atg1 overexpression shortens lifespan compared to controls. Overexpressor lines were combined with tubGAL80ts, which is a temperature sensitive GAL4 repressor. This enabled induction of Atg1 overexpression in day-2 adult fly, thereby bypassing development and any potential developmental effects that Atg1 might have. Fly eggs of appropriate genotype were raised under standard density at 18C and then emerged flies were switched to 27C at day 2 of adulthood for Atg1 overexpression, and were kept at 27C onwards for longevity analyses. Dissected tissue was collected for the transcriptomic analysis at two weeks of age.
