RNA Seq Data of adult Drosophila melanogaster heads with Mushroombody-specific knockdown of Atg5 in comparison to wildtype controls

ABSTRACT: This Dataset contains global mRNA expression data of 10 day old female adult Drosophila melanogaster heads in which autophagy, a fundamental cellular degradation pathway in contexts of health and disease, is impaired in a major brain region, the Mushroombody, by RNAi mediated knockdown of Atg5 (ok107-Gal4>UAS-Atg5-RNAi). The control group consists of an age- and sex-matched wildtypic Drosophila strain (ok107-Gal4/+). 4 biological replicates per genotype were processed, each sample containing around 50 Drosophila heads. Samples labelled with AB1-AB4 refer to the control group, samples labelled with XY1-XY4 to the Atg5 knockdown group.

Project description:Gene expression profiling of nuclei isolated from genetically defined neuronal subpopulations of the adult Drosophila brain. Gene expression profiles were generated from nuclei isolated from all neurons (R57C10-GAL4), Kenyon cells (OK107-GAL4), and octopaminergic (Tdc2-GAL4) neurons in the adult Drosophila head using a method similar to INTACT (Deal and Henikoff, 2010; Steinner et al., 2012). Sequencing was performed with an Illumina HiSeq 2000.

Transcription profiling of Drosophila EcR-dependent gene expression in MB neurons at the onset of axon pruning

Project description:This study identifies genes that show EcR-dependent gene expression in MB neurons at the onset of axon pruning. Experiment Overall Design: A dominant negative form of the ecdysone receptor (EcRDN) was expressed in MB neurons by OK107-GAL4. Animals were staged at 0 and 5 hours relative to puparium formation. RNA was isolated from MB neurons labeled with mCD8::GFP driven by OK107-GAL4 by laser capture microdissection, labeled and hybridized to Affymetrix Drosophila Genome Arrays. These microarrays were compared to RNA isolated from y,w animals at similar time points.

Cell type-specific histone modification maps were generated from nuclei isolated from all neurons (R57C10-GAL4), Kenyon cells (OK107-GAL4), and octopaminergic (Tdc2-GAL4) neurons using a method similar to INTACT (Deal and Henikoff, 2010; Steinner et al., 2012). Three histone modifications were profiled: H3K4me3, H3K27ac, and H3K27me3. Sequencing was performed with an Illumina HiSeq 2000.

Project description:List of protein scores for putative TANGO10 interactors from mass spectrometry analysis of FLAG-tagged TANGO10 using either elav-GAL4 or tim-GAL4 at both ZT10 and ZT 22, as determined using Proteome Discoverer software (ThermoFisher). Fly heads from GAL4 heterozygotes were used as controls. The hits from FLAG-tagged twenty-four (TYF) proteomics were also excluded from the list to increase TANGO10-specificity. Proteins listed are those present in at least one elav-GAL4 sample and at least one tim-GAL4 sample but no negative controls.

Microarray analysis of a Drosophila dopamine transporter mutant, fumin (fmn)

Project description:We previously found a short sleeper mutant, fmn, and identified its mutation in the dopamine transporter gene. In an attempt to discover additional sleep related genes in Drosophila, we carried out a microarray analysis comparing mRNA expression in heads of fmn and control flies and found differentially expressed genes. Next, using the pan-neuronal Gal4 driver elav-Gal4 and UAS-RNA interference (RNAi) to knockdown individual genes, we performed a functional screen. We found that knockdown of the NMDA type glutamate receptor channel gene (Nmdar1) reduced sleep.
