Project description:Neuroinflammation plays a role in the progression of several neurodegenerative disorders. We used a lipolysaccharide (LPS) model of neuroinflammation to characterize the gene expression changes underlying the inflammatory and behavioral effects of neuroinflammation. A single intracerebroventricular injection of LPS (5 ug) was administered into the lateral ventricle of mice and, 24 hours later, we examined gene expression in the cerebral cortex and hippocampus using microarray technology. Gene Ontology (GO) terms for inflammation and the ribosome were significantly enriched by LPS, whereas GO terms associated with learning and memory had decreased expression. We detected 224 changed transcripts in the cerebral cortex and 170 in the hippocampus. Expression of Egr1 (also known as Zif268) and Arc, two genes associated with learning and memory, was significantly lower in the cortex, but not hippocampus, of LPS-treated animals. Overall, altered expression of these genes may underlie some of the inflammatory and behavioral effects of neuroinflammation. Mice were given intracerebroventricular injections of saline vehicle (n = 4) or lipopolysaccharide (n = 4). Twenty-four hours later, we dissected the hippocampus and cerebral cortex and processed the tissue for microarray analysis. Gene expression changes observed in the microaray data were validated with quantitative real-time PCR.

Project description:Neuroinflammation is a central driver of cognitive decline in many neurodegenerative diseases and is marked by persistent activation of pro-inflammatory pathways. N,N-Dimethylacetamide (DMA), an FDA-approved excipient with emerging anti-inflammatory properties, was evaluated for its therapeutic potential in a murine model of chronic neuroinflammation. Both male and female C57BL/6 mice were administered lipopolysaccharide (LPS; 0.5 mg/kg) to induce neuroinflammation over seven days, followed by treatment with DMA. Behavioral assays—including the Radial Arm Maze and Novel Object Recognition tests—revealed significant cognitive deficits in LPS-treated animals, which were attenuated in DMA-treated mice, indicating preserved spatial memory and object recognition. To elucidate underlying molecular changes, RNA sequencing was performed on brain tissue, and differential gene expression analysis was followed by KEGG pathway enrichment. LPS exposure upregulated pathways related to immune activation, including TNF signaling, NF-κB signaling, MAPK signaling, and cytokine–cytokine receptor interactions. DMA treatment reversed many of these transcriptional changes, downregulating pro-inflammatory signaling and upregulating genes involved in neuroprotection and synaptic signaling. These findings suggest that DMA not only modulates key inflammatory pathways at the transcriptomic level but also improves cognitive function in a neuroinflammatory context. Collectively, this study highlights DMA’s potential as a therapeutic agent for preventing inflammation-associated cognitive decline.

Project description:Reactive astrogliosis is characterized by a profound change in astrocyte phenotype in response to all CNS injuries and diseases. To better understand the reactive astrocyte state, we used Affymetrix GeneChip arrays to profile gene expression in populations of reactive astrocytes isolated at various time points after induction using two different mouse injury models, ischemic stroke and neuroinflammation. Young adult male mice underwent middle cerebral artery occlusion (MCAO) to produce ischemic stroke or control sham surgery. Young adult mice were injected intraperitoneally with 5 mg/kg lipopolysaccharide (LPS) to produce neuroinflammation or saline for control. Astrocytes were acutely purified from control and injured brains at 1 day after injury for LPS/saline injection and 1, 3 days and 7 days after MCAO/sham surgeries.

Project description:Reactive changes of glial cells during neuroinflammation impact brain disorders and disease progression. Elucidating the mechanisms that control reactive gliosis may help understand brain pathophysiology and improve outcomes. Here, we report that adult ablation of the ASD-associated CHD8 in astrocytes attenuates reactive gliosis via remodeling chromatin accessibility, changing gene expression. Conditional Chd8 deletion in astrocytes, but not microglia, suppresses reactive gliosis by impeding astrocyte proliferation and morphological elaboration. Astrocyte Chd8 ablation alleviates lipopolysaccharide-induced neuroinflammation and septic-associated hypothermia in mice. Astrocytic CHD8 plays an important role in neuroinflammation by altering the chromatin landscape, regulating metabolic and lipid-associated pathways, and astrocyte-microglia crosstalk. Moreover, we show that reactive gliosis can be directly mitigated in vivo using an AAV-mediated Chd8 gene editing strategy. These findings uncover a role of ASD-associated CHD8 in the adult brain, which may warrant future exploration of targeting epigenetic pathways in reactive gliosis and neuroinflammation in injury and neurological diseases.
