Whole genome sequencing of aureobasidin A resistant mutants in Candida albicans

ABSTRACT: We treated C. albicans reference strain SC5314 with 20ng/ml of aureobasidin A, and obtained two mutants which were resistant to aureobasidin A. Then we did whole genome sequencing of these two mutants.

Project description:Candida albicans lab strain SC5314 was grown in YPD broth supplemented with sub-MIC concentration (5 ng/ml) of aureobasidin A (AbA) for 24h. The culture was then washed and diluted with distilled water. Approximately 200 cells were spread on YPD agar plate. Randomly 40 colonies were tested with spot assay for tolerance to AbA. 21 colonies were tolerant and were sequenced.

Then the culture was washed and diluted with distilled water, then plated on YPD plate. From each culture, we randomly tested 120 colonies to see if they obtained tolerance to brefeldin A. In total, we got 30 tolerant adaptors. We sequenced the adaptors as well as the parent.
